The chemoselective polymer blotting method allows for rapid and efficient synthesis of glycopeptides based on a "catch and release" strategy between solid-phase and water-soluble polymer supports. We have developed a heterobifunctional linker sensitive to glutamic acid specific protease (BLase). The general procedure consists of five steps, namely (i) the solid-phase synthesis of glycopeptide containing BLase sensitive linker, (ii) subsequent deprotections and the release of the glycopeptide from the resin, (iii) chemoselective blotting of the glycopeptide intermediates in the presence of water-soluble polymers with oxylamino functional groups, (iv) sugar elongations using glycosyltransferases, and (v) the release of target glycopeptides from the polymer platform by selective BLase promoted hydrolysis. The combined use of the solid-phase chemical syntheses of peptides and the enzymatic syntheses of carbohydrates on water-soluble polymers would greatly contribute to the production of complicated glycopeptide libraries, thereby enhancing applicative research. We report here a high-throughput synthetic system for the various types of MUC1 glycopeptides exhibiting a variety of sugar moieties. It is our belief that this concept will become part of the entrenched repertoire for the synthesis of biologically important glycopeptides on the basis of glycosyltransferase reactions in automated and combinatorial syntheses.  相似文献

High-frequency dielectric relaxation behavior up to 20 GHz was investigated for plain (alpha, beta, gamma) and (62 and 100%) methylated cyclodextrins, CDs, in dimethyl sulfoxide, DMSO, solution. Each hydrogen atom of OH groups of the CDs solvated a DMSO molecule for a residence time of 130-180 ps due to the hydrogen bond formation to an oxygen atom of DMSO, and a few DMSO molecules were included in cavities of the CDs for a while similar to the residence time. The overall rotational relaxation modes of solvated CDs were also observed depending on the effective sizes of the solvated CDs.  相似文献

TiO2 nanosized powders controlling by ultrasound sol-gel reaction   总被引:1,自引:0,他引:1  
We studied that anatase-TiO2 powders prepared from sol-gel process of titanium tetra-isopropoxide (TTIP) were developed under ultrasonic irradiation with different frequency of 28, 45 and 100 kHz. The irradiated ultrasound (US) was controlled by using semi-cylindrical reflection plate that was placed onto the vicinity of reaction vessel. The focused US influenced the reduction of particles size and increased the surface area of resultant nanosized TiO2 powders. We also examined photodegradation of rhodamine 640 dye (Rh-640) solution by the resultant TiO2 under UV light exposure. It was observed that low frequency for TiO2 photocatalyst preparation and low calcination temperature were more affected onto the photodegradation of the dye.  相似文献

Titania–silica composite have been prepared using polyethylene glycol (PEG) with different molecular weights (M w), PEG20000, PEG10000, and PEG2000, as template in supercritical carbon dioxide (SC CO2). The composite precursors were dissolved in SC CO2 and impregnated into PEG templates using SC CO2 as swelling agent and carrier. After removing the template by calcination at suitable temperature, the titania–silica composite were obtained. The composite were characterized by X-ray diffraction, Fourier transform infrared spectroscopy, and nitrogen sorption–desorption experiment. Photocatalytic activity of the samples has been investigated by photodegradation of methyl orange. Results indicate that there are many Si–O–Ti linkages in the TiO2/SiO2 composite; the PEG template has a significant influence on the structure of TiO2/SiO2. In addition, the TiO2/SiO2 prepared with PEG10000 exhibited high photocatalytic efficiency. So this work supplies a clue to control and obtain the TiO2/SiO2 composite with different photocatalytic reactivity with the aid of suitable PEG template in supercritical CO2.  相似文献

N2 adsorption isotherms of molecular sieve carbon were measured at 77 K and 303 K. The Ar adsorption isotherms of molecular sieve carbon samples were also measured at 303 K. The grand canonical Monte Carlo (GCMC) simulation technique was applied to calculate the N2 and Ar adsorption isotherms at 303 K using the ultramicropore volume determined by H2O adsorption. The comparative method of experimental and simulated isotherms of supercritical N2 and Ar at 303 K gave the width of the micropore mouth of the molecular sieve carbon, which can be applied to the ultramicropore width determination for other noncrystalline porous solids.  相似文献

Sugarcane bagasse was used as a cellulose resource, and the transparent cellulose hydrogel films were obtained from the purified cellulose by phase inversion process without chemical cross‐linking, when the dissolved cellulose in lithium chloride/N,N‐dimethyl acetamide was transformed into the solid film. On these processes, bagasse was pre‐treated by 10 wt% sodium hydroxide in the absence and presence of bleaching of 10 vol% sodium hypochlorite (NaOCl) solution in order to obtain cellulose fibers. Here, the bleaching temperature was varied from 40 to 50°C. The effect of pre‐treatment conditions on the resultant cellulose solution and hydrogel films was investigated. It was seen that strong bleaching removed most of lignin component from the bagasse. However, viscosity and size exclusion chromatogram of the cellulose indicated that this operation decreased average molecular weight of the cellulose fibers from 2.1 × 106 to 4.8 × 105. These property changes of fibers also caused increase of water content and weakening of mechanical strength of the resultant hydrogels. In addition, scanning probe microscopy in wet state revealed that the porous fiber network structure in the hydrogel was greatly influenced by bleaching with NaOCl. The average pore size of fiber network was decreased from 8.1 to 5.9 nm as the NaOCl treatment was at 50°C, because of expanded fibers in the swollen hydrogel.
