A generalized Gibbs-ensemble methodology with a fluctuating particle is used to determine the coexistence vapor-liquid densities for a square-well fluid. It is shown that the presence of the fluctuating particle in sub-states of the Markov chain of states supresses considerably density fluctuations which makes it possible to carry out simulations efficiently even for temperatures very close to the critical temperature. 相似文献

A concise synthesis of lupane triterpenes with an elongated carbon chain at the C-28 position, as well as saponins containing d-mannose, l-arabinose, and l-rhamnose moieties at the C-3 position is described. The overall synthesis of the new triterpenes involved seven linear steps starting from natural betulin: selective protection of a hydroxyl group, oxidation, elongation of the carbon chain by Grignard reaction, and deoxygenation. O-Glycosides were obtained by glycosylation of triterpenes with classical Schmidt's donors. Additionally, all new compounds were evaluated in vitro for their cytotoxic activities. Several triterpenes and the corresponding saponins exhibited an interesting cytotoxic activity profile against human cancer cell lines. The therapeutical index of active triterpenes is very high, since almost none of them were cytotoxic for normal BJ fibroblasts. These results open the way to the synthesis of various lupane-type saponin derivatives as potentially bioactive compounds. 相似文献

Statins have been widely used for the treatment of hypercholesterolemia due to their ability to inhibit HMG-CoA reductase, the rate-limiting enzyme of de novo cholesterol synthesis, via the so-called mevalonate pathway. However, their inhibitory action also causes depletion of downstream intermediates of the pathway, resulting in the pleiotropic effects of statins, including the beneficial impact in the treatment of cancer. In our study, we compared the effect of all eight existing statins on the expression of genes, the products of which are implicated in cancer inhibition and suggested the molecular mechanisms of their action in epigenetic and posttranslational regulation, and in cell-cycle arrest, death, migration, or invasion of the cancer cells. 相似文献

Electrochemistry combined with mass spectrometry represents an emerging analytical technique used to study the oxidation pathway of various drugs and in vivo occurring compounds, continuously showing a capability to generate many known metabolites or new oxidation products. An on‐line HPLC/EC/HR ESI‐MS method had been used to investigate the oxidation of selected cytokinin compounds. This setup allowed rapid identification and general structure elucidation of the obtained products. An electrochemical oxidation of isopentenyladenine resulted in five products, including hydroxylated and dehydrogenated products, which correlates very well with its in vivo metabolism. Electrochemical conversion of trans‐zeatin revealed six products, with two dehydrogenation products corresponding to its in vivo occurring metabolites. cis‐Zeatin oxidation in the electrochemical cell gave rise to eight products, resembling similarity to trans‐zeatin oxidation. All three compounds underwent a complete turnover mainly through two oxidation reactions occurring in the electrochemical cell? dehydrogenation and a less typical aliphatic hydroxylation. Pyrazolo[4,3‐d]pyrimidines are of interest as potential kinase inhibitors. This article describes the formation of a novel highly conjugated, condensed, centrosymmetric heteroaromatic compound, 4,9‐dichloro‐2,7‐diisopropyl‐1,3,5,5b,6,8,10,10b‐octaazacyclopenta[h,i]aceanthrylene ( 3 ), during the chlorination of 5,7‐dihydroxypyrazolo[4,3‐d]pyrimidine ( 1 ) with phenylphosphonic dichloride. The nucleophilic attack of benzylamine on 3 afforded N‐benzyl‐5‐chloro‐3‐isopropyl‐1H‐pyrazolo[4,3‐d]pyrimidin‐7‐amine ( 6 ), which was further substituted to yield a pyrazolo[4,3‐d]pyrimidine analogue of roscovitine, a well‐known cyclin‐dependent kinase inhibitor. 相似文献

We describe a screening methodology that can be used to quickly determine the effectiveness of newly synthesized photocatalysts. We were particularly interested in measuring the destruction of organic molecules painted onto a photocatalytic surface by spraying, with destruction proceeding in ambient air (as a model for airborne toxin destruction). Our method can utilize photocatalysts that are synthesized as powders (such as doped and undoped titanium oxide) and which are then calcined onto a glass substrate disk at 600°C. Herein, we used UV illumination of Aeroxide P-25 TiO(2), but the method is general and can accommodate any region of the light spectrum. 相似文献

On‐line LC‐EC/ESI‐MS has been established as a fast and simple method to mimic some types of oxidation reaction of various drugs and to study the formation and structure of the resulting products. This technique has been applied to a 2,6,9‐trisubstituted purine, R‐roscovitine, which is known to be an inhibitor of some cyclin‐dependent kinases (CDKs) and a potential anticancer drug. Oxidation of R‐roscovitine in an electrochemical cell (EC), operated under various conditions, resulted in appearance of 6 major products. These were further analyzed by high‐resolution mass spectrometry, their structures were elucidated by accurate mass measurement and compared to previously identified R‐roscovitine in vitro/in vivo metabolites. Although none of the observed products was structurally identical to those identified previously in vitro/in vivo, all of them, except for the methoxylated products, resembled similarity due to appearing through the same reaction type. R‐roscovitine in the EC cell underwent N‐dealkylation of the isopropyl moiety, hydroxylation of the aromatic side‐chain, dihydroxylation, methoxylation and dimer formation. The hydroxylation product was identified as Olomoucine II, a R‐roscovitine derivative, which displays 10‐times higher CDK‐inhibiting activity than R‐roscovitine and the occurrence of which, as R‐roscovitine product, has not yet been observed in vitro/in vivo. 相似文献
