Summary The mathematical resolution of overlapped chromatographic peaks obtained in HPLC with a diode array UV detector has received considerable attention recently. The purpose of the proposed methods is the quantitation and identification of unknown solutes in complex mixtures by an efficient use of all available data. Basically two approaches have been proposed: the first one resolves the concentration profiles and calculates the pure spectra by applying a minimal number of assumptions, which is denoted self-modeling-curve resolution. The second one is based on a match of pure spectra available in a spectral library with the measured mixture spectra. In this paper both approaches are evaluated with respect to their performance to provide the pure spectra and an accurate quantitation of the concentrations.  相似文献

This paper reviews the use of lipid vesicles as model membranes in capillary electrophoresis (CE). The history and utility of CE in the characterization of microparticles is summarized, focusing on the application of colloidal electromigration theories to lipid vesicles. For instance, CE experiments have been used to characterize the size, surface properties, enclosed volumes, and electrophoretic mobilities of lipid vesicles and of lipoprotein particles. Several techniques involving small molecules or macromolecules separated in the presence of lipid vesicles are discussed. Interactions between the analytes and the lipid vesicles - acting as a pseudostationary phase or coated stationary phase in electrokinetic chromatography (EKC) - can be used to obtain additional information on the characteristics of the vesicles and analytes, and to study the biophysical properties of membrane-molecule interactions in lipid vesicles and lipoproteins. Different methods of determining binding constants by EKC are reviewed, along with the relevant binding constant calculations and a discussion of the application and limitations of these techniques as they apply to lipid vesicle systems.  相似文献

The reproducibility of two migration parameters (retention time and mobility) of a seven-component test mixture was examined under various operating conditions using laboratory-built capillary electrophoresis systems. It was found that the frequency of rinsing the capillary and the solutions used for rinsing had the greatest effect on migration reproducibility. In addition, it was found that the migration behavior of solutes that interact with micelles is not repeatable unless the proper rinse protocol is applied. Inconsistent migration behavior is linked to inconsistent total current of the system. Preliminary investigations indicate that the fluctuation in total current were associated with non-equilibrium conditions between the buffer and the capillary wall.  相似文献

Background  

Inflammation around cell bodies of primary sensory neurons and retinal ganglion cells enhances expression of neuronal growth-associated genes and stimulates axonal regeneration. We have asked if inflammation would have similar effects on corticospinal neurons, which normally show little response to spinal cord injury. Lipopolysaccharide (LPS) was applied onto the pial surface of the motor cortex of adult rats with or without concomitant injury of the corticospinal tract at C4. Inflammation around corticospinal tract cell bodies in the motor cortex was assessed by immunohistochemistry for OX42 (a microglia and macrophage marker). Expression of growth-associated genes c-jun, ATF3, SCG10 and GAP-43 was investigated by immunohistochemistry or in situ hybridisation.  相似文献

The nucleide 73Kr has been identified by on-line mass separation as a precursor of β-delayed proton emission. The proton branch is (6.8 ±1.2) × 10−3 proton/decay. The protons populate the ground state and also the first excited 2+ state at 866 keV in 72Se with a relative intensity of (35±9) %. The value of QECBp, where Bp is the proton separation energy for the nucleus 73Br, is found to be 4.85 ±0.30 MeV based on the fraction of proton events preceded by positron decay.  相似文献

An extension of the iterative regression optimization strategy to multi-parameter optimizations is described and applied to the separation of ionic compounds (amino acids and peptides) by means of micellar liquid chromatography. The parameters examined are the concentration of surfactant, the concentration of 2-propanol and pH. Fairly regular (linear, weakly curved) retention behaviour of the compounds as a function of the parameters results in an efficient optimization using a relatively small number of initial experiments.  相似文献
