Application of affinity capillary electrophoresis for charge heterogeneity profiling of biopharmaceuticals

Charge heterogeneity profiling is important for the quality control (QC) of biopharmaceuticals. Because of the increasing complexity of these therapeutic entities [1], the development of alternative analytical techniques is needed. In this work, flow‐through partial‐filling affinity capillary electrophoresis (FTPFACE) has been established as a method for the analysis of a mixture of two similar monoclonal antibodies (mAbs). The addition of a specific ligand results in the complexation of one mAb in the co‐formulation, thus changing its migration time in the electric field. This allows the characterization of the charged variants of the non‐shifted mAb without interferences. Adsorption of proteins to the inner capillary wall has been circumvented by rinsing with guanidine hydrochloride before each injection. The presented FTPFACE approach requires only very small amounts of ligands and provides complete comparability with a standard CZE of a single mAb.     

Disclosure of the imidazolium cation coordination and stabilization mode in ionic liquid stabilized gold(0) nanoparticles

A surface-enhanced Raman spectroscopy (SERS) study of imidazolium ionic liquid stabilized gold(0) nanoparticles (GNPs) furnished previously unknown knowledge about the coordination and stabilization mode of the imidazolium cation. GNPs were prepared by hydrazine reduction of a chloroauric acid solution in 1-triethylene glycol monomethyl ether-3-methylimidazolium methanesulfonate 2 as ether-functionalized room-temperature ionic liquid (RTIL). UV-vis spectroscopy showed the presence of GNP aggregates as absorptions extended to the NIR region. A parallel coordination mode for the imidazolium cation of RTIL 2 on the GNP surface was observed by SERS, which occurred without the simultaneous coordination of the 1-triethylene glycol monomethyl ether-functionality. Instead of this, the ether-functionality was directed away from the GNP surface and acted as steric barrier between the GNPs/GNP aggregates, thus preventing further aggregation. These new insights suggest that the imidazolium cation is responsible for electrosteric stabilization.     

Conservative methods for the Toda lattice equations

We are concerned with the numerical integration of the Todalattice equations by using different conservative methods. Numericalexperiments suggest that the global error for isospectral schemesdecreases exponentially with time but it is almost constantfor either symplectic or more general integrators. We providea theoretical explanation for these experimental findings.
