Abstract— De-etiolation of maize seedlings reduces their sensitivity for red light potentiation of rapid chlorophyll accumulation in white light. An earlier proposal (Raven and Spruit, 1973) attributes this to migration of the far-red absorbing form of phytochrome (Pfr) to receptors essential for chlorophyll synthesis, thereby increasing the local Pfr/total phytochrome (Ptot)ratio. We have studied etioplasts as possible loci for such P(r receptors. The level of spectrophotometric phytochrome in purified etioplasts isolated from red preirradiated maize seedlings was higher than that of dark grown plants. The difference was marginally significant, however. We argue that migration of a fraction of cytoplasmic Pfr to the etioplasts, too small to be spectrophotometically demonstrable, could still meet the requirements of the model. Dark destruction of bulk spectrophotometric Pfr following saturating red irradiation of seedlings is not paralleled by a decrease of etioplast phytochrome. the latter remaining essentially constant over long periods. On the other hand, the potentiating effect of red light in intact seedlings is still partially reversible by far red light even after 24 h of darkness when destruction of bulk Pfr is complete. Since this demonstrates persistent presence of Pfr active in potentiation, we propose that at least part of this Pfr is associated with the etioplasts.  相似文献

Abstract— In epicotyl tissue of Pisum , irradiation of Pr at – 196°C forms a stable product P698, whereas Pfr forms a stable product P650. On warming P698, dark transformation to Pr predominates. On warming P650 to – 70°C an intermediate P690 is formed which bleaches on further warming to –10°C. When tissue is cooled to –196°C during actinic irradiation, difference spectra for subsequent warming to –10°C indicate that Pr, Pfr and an intermediate P710 are formed from weakly absorbing intermediates. Complete photoconversion of Pr to Pfr is not possible at temperatures below –5°C. As the temperature is reduced, the amount of Pfr produced from Pr decreases, while P710 increases. P710 can be photoconverted at –20°C and above, ultimately forming Pr, but in contrast to Pfr it is not photoconvertible at –196°C.  相似文献

Summary A peroxyoxalate chemiluminescence detection system for liquid chromatography is described. The excitation of fluorophores is generated by the reaction of bis-(2,4,6-trichlorophenyl)oxalate or bis(2,4-dinitrophenyl)oxalate and hydrogen peroxide, which are added to the column effluent. The influence of the solvents and the concentrations of the reagents have been investigated. The influence of the flow cell volume on sensitivity and on band broadening have also been studied and a chemical band narrowing effect has been observed. Different types of apparatus have been compared for detection of the emitted light. The system has been used for the detection of the dansyl derivative of a drug with a secondary amine functional group in serum samples. The detection limits are in the 1–10pg range.  相似文献

1. The determination of low degrees of hardness in water according to the usual methods does not yield satisfactory results. Therefore an investigation was carried out with a soap solution of sodium, oleate according to clarke, and also nephelometric measurements were made. 2. Measurements were cairied out at degrees of hardness from 0.5 to 39.0 p.p.m. (i.e. 0°.005–3°.09 FH; or 0°.003–2°.18 DH), made out of pure calcium chloride solutions, at different pH values; also with calcium chloride solations, containing NaCI and alcohol. Magnesium, chloride solutions were investigated in the same way. 3. The calcium oleate, that precipitates when the hardness is determined, has a composition of 5 CaOl2-2NaOl. The magnesium oleate has a corresponding composition. Dependent on the circumstances also 5 CaOl2-3NaOl and calcium oleates. containing yet more sodium oleate may be formed. This phenomenon occurs especially at very low degrees of hardness. 4. No turbidity of calcium oleate arises when precipitation takes place at degrees of liardness lower tlian 1.2 p.p.m. CaCO3(0°.07 DH), Magnesium olcate does not precipitate at degrees of hardness lower than 3.2 p.p.m. (0°.18 DH). 5. As a result of the phenomenon referred to in. point 3 the determination according to clarke for degrees of hardness below 5 p.p.m. CaCO3(0°.3 DH) are only reliable if very important errors (up to 30%) are tolerated. In order to determine these degrees of hardness it is therefore advisable to add so much concentrated CaCl2 solution as to reach a handy hardness of the water sample.  相似文献

Abstract— Accumulation of weakly absorbing phytochrome intermediates has been demonstrated in Pisum epicotyl tissue under conditions of pigment cycling using a quasi-continuous measuring spectrophotometer. An action spectrum shows 690–700 nm to be the most efficient wavelength range in this process. Difference spectra for the decay of intermediates maintained by 690 nm light show that, if the experiment is done at 0°C, only Pfr is formed. At – 11°C, intermediates decaying to Pr can also be observed. At – 20°C, Pr is produced as well as a pigment with peak absorption at 710nm. Kinetic analysis of intermediate decay at – 11°C reveals that at least two intermediates are maintained by 690 nm light. The level of intermediate maintained by incandescent light at 0°C was 25% higher in air than in nitrogen.  相似文献

Summary The combination of an ion-pair extraction detection system with peroxyoxalate chemiluminescence detection has been investigated. Ion-pairs of protonated tertiary amines with a chemiluminescent counter ion are on-line post-column extracted to 1,2-dichloroethane containing bis(2,4,6-trichlorophenyl)oxalate (TCPO). Hydrogen peroxide is added to the organic phase by means of a solid-state perhydrit reactor. The influence of the base catalyst (imidazole) on the chemiluminescence reaction in apolar solvents was studied. Some sulfonated chemiluminophores were compared with respect to their chemiluminescence, ion-pairing and extraction properties, and 5-dimethylaminonaphthalene-1-sulfonate was found to be the most suitable reagent. The detection limit of the potential drug secoverine is in the sub-nanogram range.  相似文献

Abstract— Irradiation of small phytochrome from oat in its Pr form with 15 ns laser pulses of different wavelengths(605–655 nm) gave rise to a difference absorption with maxima at 400 and 685 nm for the first detectable transient. Bleaching of a 660 nm band was observed, non-recuperable up to 1 ms. The transient absorption has a lifetime of 70±15 μs at 273 K. The transient is tentatively identified as lumi-R and the conformation of its chromophore is postulated to be more extended than that of Pr. A deviation from the exponential decay of the lumi-R absorption at 284 and 300 K and the lack of observable enhancement of the far-red absorption within 1 ms are interpreted in terms of the appearance of still other intermediates on this time scale between lumi-R and Pfr phytochrome.  相似文献

Abstract— Kinetic experiments have provided evidence for a series of light and dark reactions of phytochrome intermediates at low temperature in Pisum epicotyl tissue. A photoequilibrium exists between Pr and P698, and between Pfr and P650. A dark reversion of P698Pr and P650pfr at –70°C has been demonstrated. When cooled to 70°C under incandescent light, most of the phytochrome in the tissue is driven into photochemically unreactive intermediates. About 2% of the phytochrome remains as weakly absorbing intermediates that form Pr and Pfr in darkness. A scheme is presented for phytochrome phototransformation in vivo.  相似文献
