This work serves as a follow-up to Part I of experiments designed to determine the underlying principles in the formation of pseudomolecular, or adduct, ions during electrospray ionization. Aromatic acids were studied by flow injection analysis in the negative ionization mode of electrospray ionization mass spectrometry. Part I dealt with common acidic anti-inflammatory pharmaceuticals. such as ibuprofen and related analogues. Part II deals with functionally less complex molecules, namely benzoic acid (BA) and substituted benzoic acids. Halide-substituted molecules are investigated to deduce the effect of electron-withdrawing substituents (bromo-, chloro-, and fluoro-) and ring position (ortho-, meta- and para-) on the response of a traditional deprotonated molecular ion ([M-H]-) and a sodium-bridged dimer ion ([2M-2 H+Na]-). Amino-substituted benzoic acids are also analyzed in order to study the effect of an additional ionizable group on the molecule, and para-tert.-butyl-BA is analyzed to study the effect of increased hydrophobicity, as they relate to the formation of pseudomolecular ions. This study shows that solution character [octanol-water partition coefficient (or log P) and pKa] of the model compounds controls the relative efficiency of formation of [M-H]- and [2M-2H+Na]- ions. However the relative gas phase character (gas phase basicity and proton affinity) also has a significant effect on the formation of the sodium-bridged dimer ion. For the halide-substituted species, placement of the electron-withdrawing atom at the meta-position gives the greatest enhancement in sensitivity. Observations also show that as the structural complexity of the model compound increases, predictions relating analyte acidity to sodium-bridged dimer ion formation give way to a stronger dependence between log P values and ionization efficiency. Supporting this hypothesis is the nearly ten-fold enhancement in signal for tert.-butyl BA relative to BA. due to the greater hydrophobicity, and consequently, increased surface activity in an electrosprayed droplet of the analyte molecule.  相似文献

Chiral resolution of arginine (Arg) and Arg derivatives is demonstrated using electrospray ionization-tandem mass spectrometry (ESI-MS). Calcium ion (Ca(II))-mediated trimeric clusters are generated, which incorporate the analyte of interest and an enantiomerically pure reference molecule of similar metal ion affinity. Two methods, one based on the measurement of a competitive-dissociation-based branching ratio (R(chir)) by the kinetic method (KM) and one based on the measurement of a chiral recognition ratio (CR) by a similar method, are compared. Incorporating N-blocked Arg derivatives (Z-Arg and Boc-Arg) as chiral references provides chiral resolution greater than that previously reported for Arg enantiomers. In a reciprocal manner, pure Arg enantiomers can be used as references for discriminating enantiomers of these N-blocked Arg derivatives. Condensed-phase and gas-phase Ca(II) ion affinity relative to Arg is also addressed qualitatively for other acidic, basic, and neutral amino acids. In some cases, when only one offspring ion is observed (insufficient for KM analysis), the CR method can be applied as an alternative to obtain a measurable stereoselectivity value for the system. The results of these experiments demonstrate the applicability of, and the difference between, the KM and the CR method for improved quantitative analysis of enantiomeric excess for Arg.  相似文献

DETECTION OF DNA-PSORALEN PHOTOADDUCTS in situ   总被引:1,自引:0,他引:1  
Abstract— An immunological method, with the use of specific immune serum, has been developed for detection of 8-methoxypsoralen (8-MOP) photoadducts to DNA, formed in situ in cell nuclei, after combined treatment with 8MOP and UV-A irradiation (Zarçbska et al. , 1978). Lymphocytes fixed on slides or in suspension, and cryostat sections of different mammalian tissues, served as antigenic substrate, after treatment with 8-MOP and UV-A in vitro. Specific fluorescence in these substrates was detected in the nuclei after treatment with 30 ˜ 140 kJ/m2 UV-A in the presence of 0.1-0.3 μg/cm2 8-MOP. PHA-stimulated-lymphocytes appeared to be the most sensitive substrate.
However, hairless mice treated with high doses of UV-A in vivo , 70 ˜ 360 kJ/m2 did not reveal a specific fluorescence of epidermal nuclei, unless a high local concentration of 8-MOP was attained.
The apparent discrepancy in the level of photoadduct detection between the in vitro and in vivo treated specimens was explained by the low number of DNA-8-MOP-photoadducts formed in vivo under these experimental conditions. The relevance of these findings to the role of DNA-8-MOP-photoadducts formed during PUVA photochemotherapy is discussed.  相似文献

Ab initio projected-unrestricted Hartree-Fock calculations have been carried out on a number of excited and ionic states of the water molecule. Results have been compared with large-scale CI calculations, with IVO calculations, and with those of Mrozek and Golebiewski obtained by the 2 × 2 rotation method applied to orbitals. It is concluded that the PUHF method may provide the most useful alternative to large-scale CI for calculating properties of open-shell systems. But it will not be generally useful for calculating spectral transition energies.  相似文献

MNDO molecular orbital calculations have been employed to investigate limited reaction pathways and potential energy surfaces for a series of SN2 reactions. Model calculations for X? + CH3X (X = H, F, OH, OCH3, and CN) indicate that the MNDO method gives qualitative agreement with ab initio studies except for the hydride–CH4 exchange. Studies involving alkylation of pyridine (Menschutkin reaction) were also carried out. For the reaction of pyridine with CH3Cl, which involves charge separation, our MNDO studies (which do not include solvation effects) do not produce a characteristic SN2 pathway. For the reaction of pyridine with trimethyloxonium cation [(CH3)3O+] as the alkylating agent, a well defined SN2 reaction pathway was obtained; this reaction involves charge transfer. A potential energy surface for the pyridine–trimethyloxonium cation reaction shows the presence of a saddle point transition state that resembles starting materials, in agreement with the Hammond postulate for this exothermic reaction.  相似文献

Steroid hormones and their metabolites are currently undergoing clinical trials as potential therapeutics for traumatic brain injury (TBI). To support this work, it is necessary to develop improved procedures for differentiating isobaric species in this compound class. Equilin sulfate (E-S), estrone sulfate (E1-S), 17α-dihydroequilin sulfate (ADHE-S), and 17β-dihydroequilin sulfate (BDHE-S) are primary constituents in hormone replacement therapies, such as Premarin, which are among pharmaceuticals being investigated for TBI treatment. The latter three compounds are isomers and can be difficult to differentiate in trace analytical determinations. In this work, a systematic study of the fragmentation of ADHE-S, BDHE-S, E1-S, and E-S under different stages of higher order tandem mass spectrometry (MSn) and variation of collision energy, allowed optimization of conditions for distinguishing the isomeric structures. For epimeric variants (e.g., ADHE-S versus BDHE-S; α- versus β-stereoisomerization in the C-17 position), differentiation was achieved at MS4 and fragmentation was demonstrated through MS5. Computational analysis was performed to further explore differences in the fragmentation pathways due to changes in stereochemistry.   相似文献

Online restricted access media with liquid chromatography and tandem mass spectrometry for the direct analysis of small molecules in biological fluids represents an interesting alternative to time‐demanding traditional sample preparation techniques. In this study, important considerations concerning the development of a restricted access media with liquid chromatography and tandem mass spectrometry method for the analysis of dansylated estrogens in biological matrix are presented. Parameters influencing peak tailing and trapping efficiency were evaluated. The key factors included the ion strength of the mobile phase, a loading flow rate of the sample onto the trap column, and selection of a proper stationary phase of the trap column for a given set of analytes. These parameters have proven to be essential for minimizing any unwanted chromatographic peak tailing. The bulk derivatization of the analytes in the biological fluids and its relationship to the observed matrix effects was evaluated as well.  相似文献

A Lamb-shift polarimeter, which has been built for a fast determination of the polarization of protons and deuterons of an atomic-beam source and which is frequently used in the ANKE experiment at COSY-Jülich, is shown to be an excellent device for atomic-spectroscopy measurements of metastable hydrogen isotopes. It is demonstrated that magnetic and electric dipole transitions in hydrogen can be measured as a function of the external magnetic field, giving access to the full Breit-Rabi diagram for the 22S1/2 and the 22P1/2 states. This will allow the study of hyperfine structure, g factors and the classical Lamb shift. Although the data are not yet competitive with state-of-the-art measurements, the potential of the method is enormous, including a possible application to anti-hydrogen spectroscopy.  相似文献
