We have developed a method to make real-time, continuous, noninvasive measurements of muscle oxygenation (Mox) from the surface of the skin. A key development was measurement in both the visible and near infrared (NIR) regions. Measurement of both oxygenated and deoxygenated myoglobin and hemoglobin resulted in a more accurate measurement of Mox than could be achieved with measurement of only the deoxygenated components, as in traditional near-infrared spectroscopy (NIRS). Using the second derivative with respect to wavelength reduced the effects of scattering on the spectra and also made oxygenated and deoxygenated forms more distinguishable from each other. Selecting spectral bands where oxygenated and deoxygenated forms absorb filtered out noise and spectral features unrelated to Mox. NIR and visible bands were scaled relative to each other in order to correct for errors introduced by normalization. Multivariate Curve Resolution (MCR) was used to estimate Mox from spectra within each data set collected from healthy subjects. A Locally Weighted Regression (LWR) model was built from calibration set spectra and associated Mox values from 20 subjects using 2562 spectra. LWR and Partial Least Squares (PLS) allow accurate measurement of Mox despite variations in skin pigment or fat layer thickness in different subjects. The method estimated Mox in five healthy subjects with an RMSE of 5.4%.  相似文献

Systematically modified octyl galactosides and octyl N-acetyllactosamines were assessed as inhibitors of, and substrates for, T. cruzi trans-sialidase (TcTS) in the context of exploring its acceptor substrate binding site. These studies show that TcTS, which catalyses the α-(2→3)-sialylation of non-reducing terminal β-galactose residues, is largely intolerant of substitution of the galactose 2 and 4 positions whereas substitution of the galactose 6 position is well tolerated. Further studies show that even the addition of a bulky sugar residue (glucose, galactose) does not impact negatively on TcTS binding and turnover, which highlights the potential of 'internal' 6-substituted galactose residues to serve as TcTS acceptor substrates. Results from screening a 93-membered thiogalactoside library highlight a number of structural features (notably imidazoles and indoles) that are worthy of further investigation in the context of TcTS inhibitor development.  相似文献

NADPH‐cytochrome P450 reductase (CPR) serves as electron donor for cytochrome P450 catalyzed monooxygenase reactions utilizing flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD) as electron transfer cofactors. Here, stable films of human and rabbit CPRs with didodecyldimethylammonium bromide (DDAB), dimyristoylphosphatidyl choline (DMPC), and poly(diallyldimethylammonium) (PDDA) were made on pyrolytic graphite (PG) electrodes for comparative structural and electrochemical studies. CD and UV‐VIS absorbance spectra suggested that near native CPR conformation is retained in PDDA films, and some conformational changes occur in DMPC or DDAB films. Cyclic voltammetry of these films gave quasireversible pairs of peaks at average formal potential ?0.246±0.008 V vs. NHE. In human CPR‐DDAB (H‐CPR‐DDAB), a second pair of peaks at +0.317 V vs. NHE was found that depended strongly on identity of buffer and salt. Excepting H‐CPR in DDAB, films showed similar voltammetry, formal potentials, and ks values. While CPR‐PDDA films had near native CPR structures, electrochemical parameters did not differ significantly from CPR‐DMPC films. The relative independence of film voltammetry from the influence of film materials for CPRs is in contrast with heme iron proteins that, while retaining near native structures, have formal potentials that depend significantly on identity of the film material.  相似文献

This paper describes several applications of polyion-biomolecule films on electrodes related to future development of in vitro chemical toxicity sensors. In the first example, composite films of DNA and ionomers cast onto pyrolytic graphite (PG) electrodes are shown to be useful for detecting DNA damage during incubation with the carcinogen styrene oxide at pH 5.5. Single electrodes can be used to estimate relative damage rates by derivative square wave voltammetry. Films containing the ionomer Nafion gave better reproducibility than another ionomer, Eastman AQ38. In the second example, films containing redox proteins myoglobin (Mb) and cytochrome (cyt) P450cam were constructed in alternate layers with polyions including DNA on rough PG electrodes. Films with reversible protein FeIII/FeII electrochemistry with up to 7 electroactive layers were made. Amounts of electroactive protein on rough PG that were 7 to 17-fold larger than in similar films on smooth gold were achieved because many more layers were electroactive. Films of Mb/DNA also showed oxidation peaks after short incubations with styrene oxide that may be attributable to DNA damage. Results are relevant to the future design of enzyme-DNA films which convert pollutants and drugs to reactive metabolites, followed by electrochemical detection of the resulting DNA damage.  相似文献

Assuming the Riemann Hypothesis, Montgomery showed by meansof his pair correlation method that at least two-thirds of thezeros of Riemann's zeta-function are simple. Later he and Taylorimproved this to 67.25 percent and, more recently, Cheer andGoldston increased the percentage to 67.2753. Here we proveby a new method that if the Riemann and Generalized LindelöofHypotheses hold, then at least 70.3704 percent of the zerosare simple and at least 84.5679 percent are distinct. This paper concerns a Markov operator T on a space L1, and aMarkov process P which defines a Markov operator on a spaceM of finite signed measures. For T, the paper presents necessaryand sufficient conditions for:
a the existence of invariant probabilitydensities (IPDs)
b the existence of strictly positive IPDs,and
c the existence and uniqueness of IPDs.
Similar resultson invariant probability measures for P are presented.
