Detection of staphylococcal toxins presents a great interest for medical diagnostics. Screening of clinical samples for the presence of several types of staphylococcal toxins using traditional methods—biological tests on animals or cell cultures as well as ELISA—is laborious. Multiplex detection methods would simplify testing. We have designed an xMAP-based assay to detect three staphylococcal toxins—enterotoxins A and B (SEA and SEB) and toxic shock syndrome toxin (TSST)—in cultural supernatants obtained from different strains of Staphylococcus aureus. The limits of detection of SEA, SEB, and TSST multiplex detection in S. aureus growth medium were 10, 1,000, and 5 pg/mL, respectively. Fifty-nine samples of S. aureus cultural supernatants were tested with the xMAP assay. The developed assay has proved highly effective detection of the natural toxins in the samples obtained due to bacterial cells cultivation. In prospect, the developed test system can be used in clinical diagnostics and in monitoring of foodstuffs and environmental objects.  相似文献

HPLC separation of the products of high-temperature reaction of a sublimed mixture of C60–C70 (10: 1) with CF3I in a sealed ampoule allowed isolation and determination of molecular structures (X-ray crystallography and 19F NMR) of two new isomers of C60(CF3)12 and one isomer of C60(CF3)14. The goal of the work was to study changes in the activity of the angiotensin-converting enzyme (ACE) and production of reactive oxygen species (ROS) in the aorta of rats after the intraperitoneal injection of stereoisomers of catechin and gallate. The activity of ACE in the aorta sections was determined by measuring the hydrolysis of hippuryl-l-histidyl-l-leucine. The production of ROS in the aorta sections was estimated from the oxidation of dichlorodihydrofluorescein. The time and dose dependences of the effect of catechin stereoisomers and gallate on ACE activity and ROS production in the aorta were studied. It was shown that (+)-catechin and gallate increased the ACE activity and ROS production, and (−)-catechin and (−)-epicatechin did not influence these parameters. The doses of (+)-catechin and gallate that increased the ACE activity to a half-maximal value (AD50) were 0.04 and 0.03 µg/kg, respectively. Fucoidin, a blocker of leukocyte adhesion to the endothelium, reduced the ACE activity to the control level in the aortas of (+)-catechin-treated rats.  相似文献

MINCRYST is an original interrelated combination of the Crystal Structure Database for Minerals, Subbase of Calculated Polycrystal Standards (Calculated Powder X-ray Diffraction Standard Subbase, CPDS Subbase), and the Applied Program Package. The Information fund of the Crystal Structure Database comprises about 4800 files. Two MINCRYST versions are developed—the local (L) version with the original Database Management System designed for use on a personal computer and the WWW version with an original interface integrated into the WWW server of the Institute of Experimental Mineralogy of the Russian Academy of Sciences (Chernogolovka) accessible to all the Internet users.  相似文献
