The effects of cholesterol, a lipid mostly found in the sarcolemmal membranes, on the interaction of amiodarone with synthetic
models of dimyristoylphosphatidylcholine (DMPC) and with native models of mitochondria and brain microsomes was studied. Alterations
on the structural order of lipids were assessed by fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene (DPH) probing
the bilayer core, and of the propionic acid derivative 3-(p-(6-phenyl)-1,3,5-hexatrienyl)phenylpropionic acid (DPH-PA) probing the outer regions of the bilayer. As detected by the probes
and according to classic observations, cholesterol progressively increased the molecular order in the fluid phase of DMPC.
Additionally, it modulated the type and extension of amiodarone effects. For low cholesterol concentrations (≤10–15 mol%),
amiodarone (50 μM) ordered DMPC bilayers and the effects were almost identical to those observed in pure DMPC. For higher cholesterol concentrations,
amiodarone ordering effects decreased slightly and faded for cholesterol concentrations as high as 25 and 30 mol%, when detected
by DPH-PA and DPH, respectively. Above these high cholesterol concentrations, a crossover from ordering to disordering effects
of amiodarone was apparent, either in the upper region of the bilayer or the hydrophobic core. The effects of amiodarone in
native membranes of mitochondria and brain microsomes, in which "native" cholesterol accounts for about 0 and 25 mol%, respectively,
correlated reasonably with the results in models of synthetic lipids. There is a close relationship between cholesterol concentration
and amiodarone effects, in either synthetic models or native model membranes. Therefore, it may be predicted that the lipid
physicochemical properties regulated by cholesterol concentration will also modulate the effects of amiodarone in sarcolemma. 相似文献

A method for analyzing the influence of noise on newborns is proposed. The method consists of defining three different types of time interval (quiet, noisy and nursing) and, for each period, environmental noise levels, heart rate, mean arterial pressure and oxygen saturation is continuously measured. The statistical analysis of the influence of the equivalent noise level, rather than instantaneous noise level, on the behavior of the physiological variables is carried out. Great influence of noise is found by using this method, which is also easily translatable to other intensive care units as actual noise conditions are used in the investigation. Moreover, episodes of Bradycardia, Hypoxia and Hypertension are easily related to simultaneous direct nursing activity or a short but high enough noise event, suggesting that both sustained noisy environment and isolated peak noises lead to the alteration of the physiological variables. 相似文献

The use of CO2 as a massive and polarizable drift gas is shown to greatly improve peak‐to‐peak resolution (Rp‐p), as compared with N2, for the separation of disaccharides in a Synapt G2 traveling wave ion mobility cell. Near or baseline Rp‐p was achieved for three pairs of sodiated molecules of disaccharide isomers, that is, cellobiose and sucrose (Rp‐p = 0.76), maltose and sucrose (Rp‐p = 1.04), and maltose and lactose (Rp‐p = 0.74). Ion mobility mass spectrometry using CO2 as the drift gas offers therefore an attractive alternative for fast and efficient separation of isomeric disaccharides. Boron-doped diamond (BDD) films are excellent electrode materials, whose electrochemical activity for some analytes can be tuned by controlling their surface termination, most commonly either to predominantly hydrogen or oxygen. This tuning can be accomplished by e.g. suitable cathodic or anodic electrochemical pretreatments. Recently, it has been shown that amorphous carbon nitride (a-CNx) films may present electrochemical characteristics similar to those of BDD, including the influence of surface termination on their electrochemical activity toward some analytes. In this work, we report for the first time a complete electroanalytical method using an a-CNx electrode. Thus, an a-CNx film deposited on a stainless steel foil by DC magnetron sputtering is proposed as an alternative electrode for the simultaneous determination of dopamine (DA) and ascorbic acid (AA) in synthetic biological samples by square-wave voltammetry. The obtained results are compared with those attained using a BDD electrode. For both electrodes, a same anodic pretreatment in 0.1 mol L−1 KOH was necessary to attain an adequate and equivalent separation of the DA and AA oxidation potential peaks of about 330 mV. The detection limits obtained for the simultaneous determination of these analytes using the a-CNx electrode were 0.0656 μmol L−1 for DA and 1.05 μmol L−1 for AA, whereas with the BDD electrode these values were 0.283 μmol L−1 and 0.968 μmol L−1, respectively. Furthermore, the results obtained in the analysis of the analytes in synthetic biological samples were satisfactory, attesting the potential application of the a-CNx electrode in electroanalysis. 相似文献

The main objective of the reference materials for biofuel specifications (BIOREMA) project is the development of two test materials (one bio-ethanol material and one biodiesel material) with well-established reference values. Of a series of three papers, this part describes the material preparation, homogeneity study, stability study, and characterisation of the bio-ethanol material. The test material thus obtained was used in an interlaboratory comparison (ILC) to assess current practices and comparability amongst laboratories providing bio-ethanol testing services. Only 13 participants provided data, resulting in a small dataset for evaluation. Further, it appeared that for a number of laboratories, there was not sufficient material for the determination of all requested parameters. In most cases, as far as the data permit, it can be concluded that the consensus values (based on participant’s results) are in good agreement with the reference or the BIOREMA values (obtained by NMIs participating in the project). For three parameters, namely ethanol content, water content, and density, there is good agreement between the reference and consensus values. For these parameters, the reproducibility standard deviation is close to, or even smaller than, the expanded uncertainty associated with the reference value. A number of parameters show very poor reproducibility, for example, pHe, electrolytic conductivity, and acidity. The same applies to sodium and copper content, which are very low and therefore challenging parameters to measure accurately. The results of the ILC underpin the need for certified reference materials and demonstrate the requirement for more robust quality control to improve the precision and trueness of the results from testing laboratories. 相似文献

A cathodically pretreated boron‐doped diamond electrode was used for the simultaneous anodic determination of ascorbic acid (AA) and caffeine (CAF) by differential pulse voltammetry. Linear calibration curves (r=0.999) were obtained from 1.9×10?5 to 2.1×10?4 mol L?1 for AA and from 9.7×10?6 to 1.1×10?4 mol L?1 for CAF, with detection limits of 19 μmol L?1 and 7.0 μmol L?1, respectively. This method was successfully applied for the determination of AA and CAF in pharmaceutical formulations, with results equal to those obtained using a HPLC reference method. 相似文献

The procedure for spatial sampling in order to find out the relation between L(day) and L(night) for urban noise is presented. From that data, other parameters like L(dn) or L(den) can be easily obtained from L(day) values. To this end, a long-term measuring campaign was carried out in eight cities of different types and sizes in northeastern Spain. The statistical treatment of the measures was based on a characterization of the streets in view of their type, including factors such as traffic and land use, which enabled a final classification of six types of streets. The results show that streets of the same type located in different cities do not have the same value for L(day)-L(night), due to socioeconomic factors, but analyzed city by city have a close to normal (or t-Student) distribution. Under this behavior, it is demonstrated that a sample of between 14 and 25 points, depending on the city characteristics, is needed in order to calculate L(day)-L(night), with a confidence level of 95% and a margin of error of +/- 1 dB(A). 相似文献
