Addition of guanidine to a 6-methylhexahydroindenone in MeOH at 85 degrees C afforded 7-epineoptilocaulin. A similar reaction with a 6-propylhexahydroindenone afforded netamine E. MnO2 oxidation of 7-epineoptilocaulin and netamine E afforded mirabilin B and netamine G, respectively. The netamines have the side chains trans, not cis as was initially proposed. A unified biosynthetic scheme for the batzelladines and ptilocaulin family is proposed. Conjugate addition of guanidine to a bis enone followed by an intramolecular Michael reaction of the enolate to the other enone, aldol reaction, dehydration, and enamine formation will lead to a tricyclic intermediate at the dehydroptilocaulin oxidation state. 1,4-Hydride addition will lead to ptilocaulin or 7-epineoptilocaulin depending on which face the hydride adds to. 1,2-Hydride addition will lead to isoptilocaulin. The key tricyclic intermediate was prepared from a tetrahydroindenone and guanidine and reduced with NaBH4 to give a mixture rich in ptilocaulin and isoptilocaulin.  相似文献

NMR resonance assignments in the vicinity of paramagnetic metals in proteins are often difficult or impossible to make using conventional 1H detected 2-D and 3-D methods due to paramagnetic line broadening. The applicability of 13Calpha{13C'} and 13C'{15N} multiple quantum coherence methods for residue-specific assignments of resonances near paramagnetic centers is described, using the Ni2+-containing enzyme acireductone dioxygenase as an example.  相似文献

The orientation of the substrate camphor in the active site of reduced CO-bound cytochrome P450cam (CYP101) as a function of reduced putidaredoxin (Pdxr) addition has been examined by NMR using perdeuterated CYP101 and perdeuterated Pdx as well as isotopically labeled d-camphor. This permits the 1H resonances of CYP101-bound camphor to be observed without interference from the signals of CYP101 or Pdx and confirms assignments of the methyl signals of camphor in the bound form. The Cys4Fe2S2 ferredoxin Pdx is the physiological redox partner and effector of CYP101. The addition of Pdx to the reduced CYP101-camphor-CO complex results in a conformational selection that is slow on the chemical shift time scale with spectral effects observed primarily at the 8-CH3 group of the camphor. The camphor signals are ring current shifted by the heme, and for the 9- and 10-CH3 resonances, these shifts are reasonably well predicted by ring current calculations from the crystal structure of CO-bound CYP101. However, in the absence of Pdx, the 8-CH3 resonance of CYP101-bound camphor is observed at considerably higher field than predicted. Dynamic simulations using ring current shift restraints generated a structure with low chemical shift violations in which the hydrogen bond between the camphor carbonyl oxygen and the OH of Tyr96 is lost, and an expansion of the active site takes place that permits reorientation of the camphor within the active site.  相似文献

Marine molluscs have long been recognised as potential records of palaeoclimate change using the patterns and differences in the stable isotopic composition of the carbonate shells. The aim of this study is to improve the robustness of this approach for aragonitic molluscs by completing the first experimental calibration of the fractionation between water and biogenic aragonite. Fractionation factors were calibrated by growing specimens of the freshwater mollusc Lymnaea peregra under controlled conditions of water temperature and isotopic composition. Fifteen populations of L. peregra were maintained at constant temperature and isotopic conditions for five months (at five different temperatures and using three different water compositions). Water samples and temperature measurements were taken regularly throughout the experiment. The temperature dependence of the fractionation factor, between 8 and 24 degrees C, is given by: 1000 ln alpha=16.74x(1000T(-1))-26.39 (T in Kelvin) and the relationship between temperature (T), delta(18)O(carb) and delta(18)O(wat) is given by: T=21.36-4.83xdelta(+ degrees )O(carb)-delta(+ degrees )O(wat) (T is in degrees C, delta(18)O(carb) is with respect to Vienna Pee Dee Belemnite (PDB), the International Atomic Energy Agency (IAEA) replacement standard for PDB, and delta(18)O(wat) is with respect to Vienna standard mean ocean water (VSMOW)) The outcome of the controlled experiment is compared with previous studies on synthetic, and biogenic, calcite and aragonite from field and laboratory investigations. These comparisons suggest that although a vital offset exists between the fractionation of isotopes in synthetic and biogenic aragonite for molluscs in general, there is no vital effect that is specific either to freshwater, or to individual, genera. Therefore, the calibrated relationship may be used for any freshwater or marine mollusc to derive palaeotemperatures providing the isotopic composition of the environmental water can be reliably constrained. Summary A homologeous series of stereoisomericbis-amides derived from racemic N-(3,5-dinitrobenzoyl)leuchine and various α,ω-diamines has been chromatographed on chiral stationary phases (CSP) derived from N-(2-naphthyl)alanine. Plots of α, the separation factor for enantiomers, versus N, the number of methylene groups in the α,ω-diamines, are convex in shape and show a maximum in α when N=B. This maximum is attributed to optimal “bridging” between adjacent binding sites on the CSP. “Bridging” is simultaneous interaction of the two ends of thebis-derivative with sites on the CSP. Separation factors for thebis-analytes are approximately the square of those of correspondingmono-amides.
