Potentiometric stripping analysis (PSA) was investigated to assay simultaneously cadmium, lead and thallium present as contaminants in highly saline solutions used in hemodialysis. The saline matrices were sodium, potassium, magnesium and calcium chlorides, sodium acetate, sodium bicarbonate and glucose, which constitute concentrates for hemodialysis. A 1000 μg mL−1 Hg(II) solution was used to prepare the mercury film electrode (MFE) and to carry out the stripping step. After a 30 s accumulation interval the analytes were simultaneously detected in the saline matrices without using masking agents. Determination limits of 80 ng L−1 for cadmium and thallium, and 50 ng L−1 for lead were calculated and a R.S.D. ranging from 0.5 to 2.2% (n = 3) was obtained measuring the analytes directly in commercial hemodialysis saline solutions. Recoveries from spiked samples ranging from 94.6 to 102.0% were obtained. The investigated metals were found in concentrations ranging from 2.7 to 5.7 μg L−1 for cadmium, 27.7 to 75.8 μ L−1 for lead and 9.6 to 18.7 μg L−1 for thallium in commercial hemodialysis solutions. The PSA method showed to be adequate to the quality control of saline concentrates for hemodialysis.  相似文献

In order, understanding the antimicrobial action of photodynamic therapy and how this technique can contribute to its application in the control of pathogens. The objective of the study was to employ a proteomic approach to investigate the protein profile of Staphylococcus aureus after antimicrobial photodynamic therapy mediated by rose bengal (RB-aPDT). S. aureus was treated with RB (10 nmoL L−1) and illuminated with green LED (0.17 J cm−2) for cell viability evaluation. Afterward, proteomic analysis was employed for protein identification and bioinformatic tools to classify the differentially expressed proteins. The reduction in S. aureus after photoinactivation was ~2.5 log CFU mL−1. A total of 12 proteins (four up-regulated and eight down-regulated) correspond exclusively to alteration by RB-aPDT. Functionally, these proteins are distributed in protein binding, structural constituent of ribosome, proton transmembrane transporter activity and ATPase activity. The effects of photodamage include alterations of levels of several proteins resulting in an activated stress response, altered membrane potential and effects on energy metabolism. These 12 proteins required the presence of both light and RB suggesting a unique response to photodynamic effects. The information about this technique contributes valuable insights into bacterial mechanisms and the mode of action of photodynamic therapy.  相似文献

In the present work, traveling wave ion mobility spectrometry-mass spectrometry (TWIMS-MS) was applied to speciation analysis of metalloproteins. The influence of pH on complexation conditions between some metals and bovine carbonic anhydrase was evaluated from pH 6 to 9, as well as the time involved in their complexation (0–24 h). Employing TWIMS-MS, two conformational states of bovine carbonic anhydrase were observed with charge states of +12 and +11; these configurations being evaluated in terms of the folded state of the apo form and this protein (at charge state +11) being linked to barium, lead, copper, and zinc in their divalent forms. Metalloprotein speciation analysis was carried out for copper (Cu+ and Cu2+), lead (Pb2+ and Pb4+), and selenium (Se4+ and Se6+) species complexed with bovine carbonic anhydrase. Mobilities of all complexed species were compared, also considering the apo form of this protein.  相似文献

Solid-phase extraction (SPE) procedures for cleanup and preconcentration followed by HPLC-UV method were investigated for the simultaneous determination of seven low-dosed pesticides in saline concentrates for hemodialysis. The target compounds were ametryn, desmetryn, prometryn, terbutryn, molinate, triallate and butylate. Polyethylene (three different types), teflon, polyurethane and polystyrene, in powder form, were investigated as adsorbents for solid-phase extraction of the analytes from the saline samples. Quantification was performed at 222 nm and the analytes were separated on a LiChrosorb RP-18 (5 μm, 125 mm × 4 mm i.d.) column using gradient elution with water/acetonitrile as mobile phase. The duration each chromatographic run was 18 min including column reconditioning. The efficiency of the different SPE substrates for retaining the analytes from the highly concentrated saline (HCS) samples was discussed. The best performance was achieved with polystyrene as SPE material considering preconcentration factor, precolumn clogging, reusing capability and similarity between the mobile phases for SPE and HPLC procedures. Analyte concentrations as low as 1 μg L−1 could be determined in spiked HCS samples after preconcentration on polystyrene SPE precolumns. Recoveries between 98.7 and 102.2% were obtained from commercial spiked samples. Detection limits ranging from 4.8 (for prometryn) to 46 μg L−1 (for butylate) were calculated (without preconcentration). The within-day relative standard deviations (n = 9) ranged from 2.3 to 4.8%.  相似文献
