A high yield method to extract alkaloids from Aconitum coreanum using pulsed electric field (PEF) was developed, and the optimized extraction method compared to the other four extraction methods, consisting of cold maceration extraction, percolation extraction, heat reflux extraction (HRE), and ultrasonic-assisted extraction (UE). The experimental factors of the extraction methods such as electric field intensity, pulse frequency and solid-to-solvent ratio were evaluated. The content of Guanfu base A (GFA) was quantified by liquid chromatography coupled with electrospray ionization tandem mass spectrometry. The results indicated that the highest yield of GFA was 3.94 mg g?1 by PEF with conditions of 20 kV cm?1 electric field intensity, 8 pulse, 1:12 solid-to-solvent ratio, and 90 % ethanol–water solution. Meanwhile, the extraction time of PEF was <1 min, which is much less than the HRE of 10 h and even the newly used technique UE of 40 min. Moreover, the results of PEF extraction method showed obvious advantages, with the highest efficiency (120 L h?1), the shortest extraction time (0.5–1 min), and the lowest energy costs, which could be applied in the industrial production of alkaloids from A. coreanum. Therefore, the application of the PEF extraction method is a promising and constructive method for extraction of GFA. 相似文献

An efficient and refined method for the separation of six aconitine‐type alkaloids from the alkaline prepared “Kusnezoff monkshood root” was established. It is the first study that two new lipo‐alkaloids were successfully isolated from refined sample by pH‐zone‐refining counter‐current chromatography rather than synthetic method. It was of interest that a great deal of lipo‐alkaloids was produced in crude extract from the alkalization of “Kusnezoff monkshood root.” A refined sample method was proposed to enrich two types of alkaloids by liquid–liquid extraction, i.e. lipo‐alkaloids and monoester‐diterpenoid alkaloids. The pH‐zone‐refining counter‐current chromatography was performed with an optimized two‐phase solvent system composed of n‐hexane‐ethyl acetate–methanol–water (3:5:4:5, v/v), where upper organic phase was added to 3 mmol/L triethylamine as a retainer and lower aqueous mobile phase was added to 3 mmol/L hydrochloric acid as an eluter. As a result, six aconitum alkaloids, including two lipo‐alkaloids (8‐lino‐14‐benzoylaconine, 8‐pal‐14‐benzoylaconine), three monoester‐diterpenoid alkaloids (14‐benzoylmesaconine, 14‐benzoylaconine, beyzoyldeoxyaconine), and one aconine alkaloid (neoline) were acquired from the plant at the same time. The anti‐inflammatory activities of the two new lipo‐alkaloids were compared to the six alkaloids in vitro, in cyclo‐oxygen‐ase‐2 inhibition assays. In standalone ion mobility spectrometry (IMS) instruments, the effect of drift gas turbulence reduces the sensitivity and resolution of the instrument. A traditional ion detector constructed with a Faraday plate and used to detect ions in an IMS is positioned at the end of the drift region. Drift gas flowing through this detector may introduce turbulence near the detector, possibly affecting the sensitivity and resolution of the device. To address this problem, a novel Faraday detector with a double layer structure was constructed. A number of dense and staggered holes were created on each layer of the detector. This design enabled the drift gas to pass through the holes of the detector, and the staggered nature of holes in the detector ensured that the ions could be detected. Theoretical simulations were conducted using the finite element method to obtain velocity distributions for both a standard Faraday detector and the modified Faraday detector. The results indicated that the novel ion detector created a homogenous gas under at high inlet flow rate while turbulence was still evident for the traditional Faraday detector. When the inlet flow rate was 1000 mL/min, the range of the unstable region of the drift gas in the axis of the drift tube with the novel ion detector was reduced by 97% relative to that for the traditional detector. The data suggests that due to such gains, sensitivity and resolution may be improved for standalone IMS instruments. 相似文献
