Irradiation of o-acetylphenylacetonitriles 1a,b in methanol (lambda >280 nm) leads to essentially quantitative photohydrolysis with formation of the corresponding ketal amides 3a,b. The mechanism proposed for this novel reaction is supported by the qualitatively different photochemical behavior of 1c and by a labeling experiment with 1a-L that confirms transfer of the carbonyl oxygen atom of 1a to the amide oxygen atom of 3a.  相似文献

The benzene tolerant Acinetobacter baylyi isolated from marine sludge in Angsila, Thailand could constitutively secrete lipolytic enzymes. The enzyme was successfully purified 21.89-fold to homogeneity by ammonium sulfate precipitation and gel-permeable column chromatography with a relative molecular mass as 30 kDa. The enzyme expressed maximum activity at 60°C and pH 8.0 with p-nitrophenyl palmitate as a substrate and found to be stable in pH and temperature ranging from 6.0-9.0 to 60-80°C, respectively. A study on solvent stability revealed that the enzyme was highly resisted to many organic solvents especially benzene and isoamyl alcohol, but 40% inhibited by decane, hexane, acetonitrile, and short-chain alcohols. Lipase activity was completely inhibited in the presence of Fe2+, Mn2+, EDTA, SDS, and Triton X-100 while it was suffered detrimentally by Tween 80. The activity was enhanced by phenylmethylsulfonyl fluoride (PMSF), Na+, and Mg2+ and no significant effect was found in the presence of Ca2+ and Li+. Half of an activity was retained by Ba2+, Ag+, Hg+, Ni2+, Zn2+, and DTT. The enzyme could hydrolyze a wide range of p-nitrophenyl esters, but preferentially medium length acyl chains (C8-C12). Among natural oils and fats, the enzyme 11-folds favorably catalyzed the hydrolysis of rice bran oil, corn oil, sesame oil, and coconut oil in comparison to palm oil. Moreover, the transesterification activity of palm oil to fatty acid methyl esters (FAMEs) revealed 31.64 ± 1.58% after 48 h. The characteristics of novel A. baylyi lipase, as high temperature stability, organic solvent tolerance, and transesterification capacity from palm oil to FAMEs, indicate that it could be a vigorous biocatalyzer in the prospective fields as bioenergy industry or even in organic synthesis and pharmaceutical industry.  相似文献

A novel β-glucosidase from higher termite Microcerotermes annandalei (MaBG) was obtained via a screening method targeting β-glucosidases with increased activities in the presence of glucose. The purified natural MaBG showed a subunit molecular weight of 55 kDa and existed in a native form as a dimer without any glycosylation. Gene-specific primers designed from its partial amino acid sequences were used to amplify the corresponding 1,419-bp coding sequence of MaBG which encodes a 472-amino acid glycoside hydrolase family 1 (GH1) β-glucosidase. When expressed in Komagataella pastoris, the recombinant MaBG appeared as a ~?55-kDa protein without glycosylation modifications. Kinetic parameters as well as the lack of secretion signal suggested that MaBG is an intracellular enzyme and not involved in cellulolysis. The hydrolytic activities of MaBG were enhanced in the presence of up to 3.5-4.5 M glucose, partly due to its strong transglucosylation activity, which suggests its applicability in biosynthetic processes. The potential synthetic activities of the recombinant MaBG were demonstrated in the synthesis of para-nitrophenyl-β-D-gentiobioside via transglucosylation and octyl glucoside via reverse hydrolysis. The information obtained from this study has broadened our insight into the functional characteristics of this variant of termite GH1 β-glucosidase and its applications in bioconversion and biotechnology.  相似文献

The combination of catalytic amounts of p-toluenesulfonic acid and 1 equiv of N-halosuccinimide afforded highly selective ring-halogenation of N-acetyl-l-tyrosine, furnishing either N-acetyl-3-halo-l-tyrosine analogues or mixed 3,5-dihalo derivatives in a one-pot reaction with excellent yields at room temperature.  相似文献

Two bioactive cyclic dipeptides, cyclo(L-Pro-L-Tyr) and cyclo(D-Pro-L-Tyr), were isolated from the culture broth of Streptomyces sp. strain 22-4 and tested against three economically important plant pathogens, Xanthomonas axonopodis pv. citri, Ralstonia solanacearum and Clavibacter michiganensis. Both cyclic dipeptides were active against X. axonopodis pv. citri and R. Solanacearum with MIC of 31.25 μg/mL. No activity could be observed against C. michiganensis.  相似文献

Microfluidic paper-based analytical devices (µPADs) have attracted much attention over the past decade because they offer clinicians the ability to deliver point-of-care testing and onsite analysis. Many of the advantages of µPADs, however, are limited to work in a laboratory setting due to the difficulties of processing data when using electronic devices in the field. This review focuses on the use of µPADs that have the potential to work without batteries or with only small and portable devices such as smartphones, timers, or miniaturized detectors. The µPADs that can be operated without batteries are, in general, those that allow the visual judgment of analyte concentrations via readouts that are measured in time, distance, count, or text. Conversely, a smartphone works as a camera to permit the capture and processing of an image that digitizes the color intensity produced by the reaction of an analyte with a colorimetric reagent. Miniaturized detectors for electrochemical, fluorometric, chemiluminescence, and electrochemiluminescence methods are also discussed, although some of them require the use of a laptop computer for operation and data processing.  相似文献

A simple and efficient liquid-phase microextraction (LPME) technique using a supported liquid hollow fiber membrane, in conjunction with gas chromatography-electron capture detector has been developed for extraction and determination of trihalomethanes (THMs) in water samples. THMs were extracted from water samples through an organic extracting solvent impregnated in the pores and filled inside the porous hollow fiber membrane. Our simple conditions were conducted at 35 degrees C with no stirring and no salt addition in order to minimize sample preparation steps. Parameters such as types of hollow fiber membranes, extracting solvents and extraction time were studied and optimized. The method exhibited enrichment factors ranged from 28- to 62-fold within 30 min extraction time. The linearity of the method ranged from 0.2 to 100 microg l(-1). The limits of detection were in the low microg l(-1) level, ranging between 0.01 and 0.2 microg l(-1). The recoveries of spiked THMs at 5 microg l(-1) in water were between 98 and 105% with relative standard deviations (RSDs) less than 4%. Furthermore, the method was applied for determination of THMs in drinking water and tap water samples was reported.  相似文献

para-Regioselective bromination of phenol and analogues, promoted by p-toluenesulfonic acid, is achieved in high to excellent yields at room temperature with N-bromosuccinimide. Chlorination with N-chlorosuccinimide and catalysed by p-toluenesulfonic acid also gives para-chlorinated phenol analogues in good yields at room temperature.  相似文献

A new approach for the detection of trimethylamine (TMA) using recombinant Xenopus laevis melanophores was developed. The cells were genetically modified to express the mouse trace amine-associated receptor 5 (mTAAR5), a G protein-coupled receptor from the olfactory epithelium, which conferred high sensitivity to TMA. A focused chemical screen allowed the discovery of additional, previously unknown stimuli of mTAAR5. The cell-based sensor demonstrated no sensitivity to trimethylamine N-oxide (TMAO), making it suitable for a convenient evaluation of TMA levels in fish tissue extracts. The developed gas measurement platform was able to detect TMA from 1 to 100 ppm within thirty-five minutes.  相似文献
