Abstract— The Fourier-transform infrared spectra of chloroform-d solutions of conjugated imines CH3CH=CHCH=NCH(CH3)2 and CH3CH2CH=CHCH=CHCH=NCH(CH3)2 and the related protonated species with HCl, HBr, HI, trichloro, dichloro, monobromo and monochloroacetic acids or propionic acid are presented. The effects of conjugation and protonation are examined. The results show that conjugation slightly increases the basicity of the Schiff bases. HCl, HBr and HI protonate the Schiff bases completely. The carboxylic acids protonate partially depending on their p K a, values. When the Schiff base contains two (or more) C=C bonds conjugated with C=N, the main C=C stretching band undergoes a strong intensification showing that sizeable dipole moment variations occur along the conjugated chain.  相似文献

A simple and efficient stereoselective synthesis of fluorine containing beta-aminophosphonates by reduction of beta-enaminophosphonates is described. Reduction with sodium cyanborohydride in the presence of zinc chloride and the catalytic hydrogenation of beta-enaminophosphonates gives beta-aminophosphonates. beta-Enaminophosphonates are also used as intermediates for the regioselective synthesis of fluoroalkyl-substituted pyridines.  相似文献

The interpretation of factors constitutes a major problem in factor analysis. Until now the underlying semantic aspects of the question have been studied only occasionally. However, there are some indications that the mechanical interpretation of the results may constitute in the immediate future a real need in the realm of very large problems. On the other hand, the semantic study of multivariate statistical methods is the only way to warrant the validity of the mechanization procedures and the means to identify their limits.  相似文献

A simple method for preparation of fluoroalkyl beta-enaminophosphonates 1 from alkylphosphonates 2 and perfluoroalkyl nitriles 3 is reported. Olefination reaction of functionalized phosphates 1 with aldehydes gives alpha,beta-unsaturated imines 5. Acid hydrolysis of these fluoroalkyl derivatives 5 affords alpha,beta-unsaturated ketones 6, while their selective reduction with hydrides leads to the formation of allylamines 7, enamines 8, and saturated ketones 9 or amines 10. Selective oxidative cleavage of the carbon-carbon double bond of allylamines 7 gives fluorinated alpha-amino aldehydes 12, alpha-amino ketones 13, or alpha-amino acid derivatives 14.  相似文献

In this work, we analyze the structure–activity relationships (SAR) of epigenetic inhibitors (lysine mimetics) against lysine methyltransferase (G9a or EHMT2) using a combined activity landscape, molecular docking and molecular dynamics approach. The study was based on a set of 251 G9a inhibitors with reported experimental activity. The activity landscape analysis rapidly led to the identification of activity cliffs, scaffolds hops and other active an inactive molecules with distinct SAR. Structure-based analysis of activity cliffs, scaffold hops and other selected active and inactive G9a inhibitors by means of docking followed by molecular dynamics simulations led to the identification of interactions with key residues involved in activity against G9a, for instance with ASP 1083, LEU 1086, ASP 1088, TYR 1154 and PHE 1158. The outcome of this work is expected to further advance the development of G9a inhibitors.

Due to the huge amount of data generated in drug discovery programs, their success strongly depends on both the workflows and platforms to manage and, more importantly, to integrate different chemical and biological data sources. At Experimental Therapeutics Program in the Spanish National Cancer Research Center (CNIO), we have addressed our efforts in the design and optimal implementation of those key processes that enable dynamic workflows and interfaces between the different information blocks. Our approach focuses on the development of a common chemical and biological repository (CCBR) that gathers all data that pass quality control criteria. An integral web application (WACBIP) was designed to query against CCBR while providing decision making tools. Currently, our CCBR contains more than 43,000 unique structures as well as experimental data from more than 350 different biological assays. As input sources of the CCBR, we federated a series of Laboratory Information Management Systems (LIMS) which cover sections as follows: chemical synthesis, analytical department, compound logistics, biochemical and cellular data (including high-throughput and high-content screenings; HTS and HCS), computational chemistry (in-silico chemogenomics and physico-chemical profiling) and in-vivo pharmacology. With regard to the last section, an integral In-Vivo Management e-Biobook (IVMB) that handles the entire workflow of in-vivo labs was designed and implemented. Herein we describe the processes and tools that we have developed and implemented, balancing purchase and development, for centralizing discovery information as well as providing decision-making and project management tools - a clear unmet need in public organizations and networks.  相似文献

In this paper, the influence on corrugation of the most significant track parameters has been examined. After this parametric study, the optimization of the track parameters to minimize the undulatory wear growth has been achieved. Finally, the influence of the dispersion of the track and contact parameters on corrugation growth has been studied. A method has been developed to obtain an optimal solution of the track parameters which minimizes corrugation growth, thus ensuring that this solution remains optimum despite dispersion of track parameters and wheel–rail contact uncertainties. This work is based on the computer application RACING (RAil Corrugation INitiation and Growth) which has been developed by the authors to predict rail corrugation features.  相似文献
