Arsenic-tolerant freshwater alga Chlorella vulgaris which had been collected from an arsenicpolluted environment were tested for uptake and excretion of inorganic arsenic. Approximately half the quantity of arsenic taken up by C. vulgaris was estimated to be adhered to the extraneous coat (10 wt %) of the cell. The remainder was bioaccumulated by the cell. Both adhered and accumulated arsenic concentrations increased with an increase in arsenic(V) concentration of the aqueous phase. Arsenic(V) accumulation was affected by the growth phse: arsenic was most actively accumulated when the cell was exposed to arsenic during the early exponential phase and then accumulation decreased with an increase in culture time exposed to arsenic. The alga grew well in the modified Detmer (MD) medium containing 1 mg As(III) dm?3 and the growth curve was approximated by a ‘logistic equation’. Arsenic(III) was accumulated up to the second day of the culture time and arsenic(III) accumulation decreased with an increase in the culture time after that. Arsenic accumulation was also largely affected by various nutrients, especially by managanese, iron and phosphorus compounds. A modified MD medium with the three nutrients was proposed for the purpose of effective removal of arsenic from the aqueous phase. Using radioactive arsenate (Na2H74AsO4), the arsenic accumulated was found to be readily excreted under conditions which were unfavourable for the multiplication of C. vulgaris.  相似文献

Lipophilic acyclic dibenzopolyether diamides, 12 kinds, have been designed to prepare solvent polymeric membrane ion-selective electrodes (ISEs) for Pb(2+). The ionophores include 1,5-bis[2-(N,N-dialkylcarbamoylmethoxy)phenoxy]-3-oxapentanes1-4, 1,5-bis[2-(N,N-dialkylcarbamoylpentadecyloxy)phenoxy]-3-oxapentanes 5-8, and 1,2-bis[2-(2'-N,N-dialkylcarbamoylpentadecyloxy)phenoxy]ethanes 9-12. Linear response concentration range of the ISE based on 9 is 3 x 10(-2) - 1 x 10(-6) M of Pb(2+) (average slope = 28.5 mV decade(-1)). Potentiometric selectivities of the ISEs based on 1-12 for Pb(2+) over other heavy metal cations, alkali metal cations, and alkaline earth metal cations have been assessed. These ISEs exhibit remarkably high selectivities for Pb(2+) relative to heavy metal cations, such as Cu(2+), Fe(2+), and Ni(2+), the selectivity coefficients (K(Pot)(Pb,Cu)) being 5 x 10(-5) - 6 x 10(-5) for 1-4 and ca. 6 x 10(-4) for 9. For the Pb(2+) selectivities over alkali metal cations, such as Na(+) and K(+), 9 which has an ethylene glycol spacer and a N,N-diethyl group is superior to other dibenzopolyether diamide ionophores 1-8 and 10-12.  相似文献

The organic acids in the urine and serum of diabetic patients with ketoacidosis and disturbance of consciousness were studied using acidification, extraction, evaporation, methoxime formation and trimethylsilylation, gas chromatographic separation and mass spectrometric identification procedures. The organic acid profile of 1 ml of serum ultrafiltrate was obtained with good separation using a gas chromatograph equipped with a glass capillary column and a splitless injector. 5-Hydroxyhexanoic acid and 3-hydroxyvaleric acid were identified for the first time in the urine of diabetic patients with ketoacidosis. Urinary excretion and serum concentrations of 2,3-dideoxypentonic acid were increased in diabetic patients.  相似文献

Phase transition behavior of polyesters derived from 2,7-phenanthrene dicarbonic acid diethylester and alkanediols with even methylene carbon number was investigated by the simultaneous DSC-XRD method. The smectic A phase was observed on cooling from the molten state. The transition entropy from the isotropic state to the smectic A phase was about 9.0 J mol-1 K-1, which depended on the methylene carbon number. The linear expansion coefficients, based on the (001) spacing of the crystalline phase at room temperature, were 1.3·10-4 K-1 (crystalline phase), 5.7·10-4 K-1 (crystallization region), 1.7·10-3 K-1 (smectic A phase) during cooling, and 1.5·10-4 K-1 (crystalline phase), and 1.0·10-3K-1(melting region) on heating. The microwave-assisted acid-digestion for the determination of metals in coal by ICP-AES was investigated, especially focusing on the necessity of adding HF. By testing five certified reference materials, BCR-180, BCR-040, NIST-1632b, NIST-1632c, and SARM-20, it was found that the two-stage digestion without HF (HNO3 + H2O2 was used) was very effective for the pretreatment of ICP-AES measurement. Both major metals (Al, Ca, Fe, and Mg) and minor or trace metals (Co, Cr, Cu, Mn, Ni, Pb, and Zn) in coal gave good recoveries for their certified or reference values. The possibility of ‘HF-memory effect’ was cancelled by the use of a set of vessels which had been never contacted with HF. Twenty-four Japanese standard coals (SS coals) were analyzed by the present method, and the concentrations of major metals measured by the present method provided very high accordance with those from the authentic JIS (Japanese Industrial Standard) method.  相似文献

Summary HPLC determination of pheophytinatonicke(II) (Pheo-Ni) prepared by the replacement of magnesium(II) in chlorophyll with nicke(II) is described. The good separation of PheoNi was obtained by using chemically bonded C18 as the stationary phase and acetone-methanol (50∶50, vol/vol) as the mobile phase. Conventional spectrophotometric method was also used for the determination of PheoNi. For the synthetic samples prepared by mixing (pheophytinato a) nicke(II) [(Pheo-a) Ni] and (pheophytinato b) nicke(II) [(Pheo-b) Ni], analytical values obtained by the spectrophotometric method were very high compared to those obtained by HPLC. In the proposed HPLC method, (Pheo-a) Ni and (Pheo-b). Ni could be determined in the concentration range of 0.028–30μg/ml and 0.038–30μg/ml with relative standard deviations (n=10) of 3.1% and 0.8%, respectively.  相似文献

A series of sulfido-bridged tungsten-ruthenium dinuclear complexes Cp*W(mu-S)(3)RuX(PPh(3))(2) (4a; X = Cl, 4b; X = H), Cp*W(O)(mu-S)(2)RuX(PPh(3))(2) (5a; X = Cl, 5b; X = H), and Cp*W(NPh)(mu-S)(2)RuX(PPh(3))(2) (6a; X = Cl, 6b; X = H) have been synthesized by the reactions of (PPh(4))[Cp*W(S)(3)] (1), (PPh(4))[Cp*W(O)(S)(2)] (2), and (PPh(4))[Cp*W(NPh)(S)(2)] (3), with RuClX(PPh(3))(3) (X = Cl, H). The heterolytic cleavage of H(2) was found to proceed at room temperature upon treating 5a and 6a with NaBAr(F)(4) (Ar(F) = 3, 5-C(6)H(3)(CF(3))(2)) under atmospheric pressure of H(2), which gave rise to [Cp*W(OH)(mu-S)(2)RuH(PPh(3))(2)](BAr(F)(4)) (7a) and [Cp*W(NHPh)(mu-S)(2)RuH(PPh(3))(2)](BAr(F)(4)) (8), respectively. When Cp*W(O)(mu-S)(2)Ru(PPh(3))(2)H (5b) was treated with a Br?nstead acid, [H(OEt(2))(2)](BAr(F)(4)) or HOTf, protonation occurred exclusively at the terminal oxide to give [Cp*W(OH)(mu-S)(2)RuH(PPh(3))(2)](X) (7a; X = BAr(F)(4), 7b; X = OTf), while the hydride remained intact. The analogous reaction of Cp+W(mu-S)(3)Ru(PPh(3))(2)H (4b) led to immediate evolution of H(2). Selective deprotonation of the hydroxyl group of 7a or 7b was induced by NEt(3) and 4b, generating Cp*W(O)(mu-S)(2)Ru(PPh(3))(2)H (5b). Evolution of H(2) was also observed for the reactions of 7a or 7b with CH(3)CN to give [Cp*W(O)(mu-S)(2)Ru(CH(3)CN)(PPh(3))(2)](X) (11a; X = BAr(F)(4), 11b; X = OTf). We examined the H/D exchange reactions of 4b, 5b, and 7a with D(2) and CH(3)OD, and found that facile H/D scrambling over the W-OH and Ru-H sites occurred for 7a. Based on these experimental results, the mechanism of the heterolytic H(2) activation and the reverse H(2) evolution reactions are discussed.  相似文献

Dibromopyridines or dibromopyridone with -(CH2)m-SO3Na group(s) has been prepared via the reactions of the corresponding dibromopyridines with -OH and -NH2 groups with sultone. These compounds were converted into polymers with the -(CH2)m-SO3H groups via organometallic polycondensation. Background

The ubiquitin ligase COP1, COnstitutively Photomorphogenic 1, functions in many biological responses in mammalian cells, but its downstream pathway remains unclear.

Results

Here, we identified FIP200, a key regulator of mammalian autophagy, as a novel COP1-interacting protein by yeast two-hybrid screening. The interaction was confirmed by a GST-pulldown assay. Split-GFP analysis revealed that interaction between COP1 and FIP200 predominantly occurred in the cytoplasm and was enhanced in cells treated with UV irradiation. Different forms of FIP200 protein were expressed in cultured mammalian cells, and ectopic expression of COP1 reduced one of such forms.

Conclusions

These data suggest that COP1 modulates FIP200-associated activities, which may contribute to a variety of cellular functions that COP1 is involved in.  相似文献

Tolerance bioaccumulation and biotransformation of arsenic compounds by a freshwater prawn (Macrobrachium rosenbergii) were investigated. M. rosenbergii was exposed to 10, 20, 30 and 35 μg As cm−3 of disodium arsenate [abbreviated as As(V)], 25, 50, 100 and 120 μg As cm−3 of methylarsonic acid (MMAA), or 100,200, 300 and 350 μg As cm−3 of dimethylarsinic acid (DMAA). Tolerances (50% lethal concentration: LC50) of the prawn against As(V), MMAA, and DMAA were 30, 100, and 300 μg As cm−3, respectively. The prawn accumulated arsenic compounds directly from aqueous phase and biotransformed them in part. Both methylation and demethylation of the arsenicals were observed in vivo. Highly methylated and less toxic arsenicals were less accumulated in M. rosenbergii.  相似文献
