The non-proteinogenic amino acid 3-hydroxy-4-methylproline (HMP) is an active constituent of some potent antimicrobials including echinocandins, nostopeptins, pneumocandins, sporiofungin and mulundocandins. A synthesis has been achieved in 10 steps with 29% overall yield; the Evans’ aldol reaction using Crimmins’ modified method was pivotal to the success of the strategy.  相似文献

Efficient syntheses of the phytoalexins brassilexin, sinalexin, and analogues are demonstrated through the application of the Vilsmeier formylation to indoline-2-thiones followed by a new aqueous ammonia workup procedure. Similarly, a very concise two-pot synthesis of the phytoalexins wasalexins using sequential formylation-amination of indolin-2-ones is described. Remarkably, this novel aqueous ammonia workup allows the sequential one-pot formylation-amination, expanding substantially the scope of the Vilsmeier formylation of both indoline-2-thiones and indolin-2-ones. The examination of the formylation-amination reaction and optimization of conditions, as well as the syntheses and antifungal activities of several brassilexin analogues, are reported.  相似文献

The resulting dienes provide cyclooctenes in moderate to good yields after ring-closing metathesis (RCM). The central macrocyclic core of the macrocidins was constructed using RCM as the key reaction. A preliminary investigation dealing with the key reactions, that is, the Dieckmann cyclization and the RCM, revealed that RCM of the β-ketoamide is better than RCM of the corresponding acyltetramic acid.  相似文献

Enzyme stability studies in case of Sclerotium rolfsii UV-8 mutant have been investigated under the conditions used for saccharification of cellulose (50 degrees C, pH 4.5, 48 h). Avicelase (measure of exoenzymes) and xylanase were found to be less stable than CMCase (endoglucanase) and beta-glucosidase. Merthiolate (and other Hg compounds) added as a biocide, inactivated avicelase and xylanase about 60-70%. Of the antibiotics tested, tetracycline, chloramphenicol, and streptomycin sulfate were found suitable as an additive in cellulose hydrolysis system. The optimum hydrolysis of alkali-treated (AT)-rice straw, AT-bagasse, Solka Floc SW40, and Avicel P.H.101 was observed under shaking conditions at pH 4.5, 50 degrees C in CO2 atmosphere. It is suggested, all the studied parameters could be used for the evaluation of mutant strains.  相似文献
