Tubiferal A, a triterpenoid isolated from myxomycete Tubifera dimorphotheca, exhibits a reversal effect of vincristine (VCR) resistance against VCR-resistant KB cell lines. The compound possesses a complex structure involving the 6-7-6-5 polycyclic carbon framework with various functional groups. The stereoselective synthesis of the right-hand segment of tubiferal A was achieved on the basis of the cyclopentene annulation method and the semi-pinacol rearrangement reaction of an epoxy alcohol for constructing the trans-fused 6-5 bicyclic skeleton possessing two quaternary carbon atoms at the angular positions.  相似文献

Lembehsterols A (1) and B (2), two novel sulfated sterols, were isolated from the marine sponge Petrosia strongylata. Both sterols showed inhibitory activity against thymidine phosphorylase, which is an enzyme related to angiogenesis in solid tumors. The structures of these sulfated sterols were established on the basis of chemical and physicochemical evidence.  相似文献

Cationic polymerization of α‐methyl vinyl ethers was examined using an IBEA‐Et1.5AlCl1.5/SnCl4 initiating system in toluene in the presence of ethyl acetate at 0 ~ ?78 °C. 2‐Ethylhexyl 2‐propenyl ether (EHPE) had a higher reactivity, compared to corresponding vinyl ethers. But the resulting polymers had low molecular weights at 0 or ?50 °C. In contrast, the polymerization of EHPE at ?78 °C almost quantitatively proceeded, and the number‐average molecular weight (Mn) of the obtained polymers increased in direct proportion to the EHPE conversion with quite narrow molecular weight distributions (weight‐average molecular weight/number‐average molecular weight ≤ 1.05). In monomer‐addition experiments, the Mn of the polymers shifted higher with low polydispersity as the polymerization proceeded, indicative of living polymerization. In the polymerization of methyl 2‐propenyl ether (MPE), the living‐like propagation also occurred under the reaction conditions similar to those for EHPE, but the elimination of the pendant methoxy groups was observed. The introduction of a more stable terminal group, quenched with sodium diethyl malonate, suppressed this decomposition, and the living polymerization proceeded. The glass transition temperature of the obtained poly(MPE) was 34 °C, which is much higher than that of the corresponding poly(vinyl ether). Living cationic copolymerization of amide‐functional vinyl ethers with isobutyl vinyl ether (IBVE) was achieved using SnCl4 in the presence of ethyl acetate at 0 °C: the number–average molecular weight of the obtained polymers increased in direct proportion to the monomer conversion with relatively low polydispersity, and the amide‐functional monomer units were introduced almost quantitatively. To optimize the reaction conditions, cationic polymerization of IBVE in the presence of amide compounds, as a model reaction, was also examined using various Lewis acids in dichloromethane. The combination of SnCl4 and ethyl acetate induced living cationic polymerization of IBVE at 0 °C when an amide compound, whose nitrogen is adjacent to a phenyl group, was used. The versatile performance of SnCl4 especially for achieving living cationic polymerization of various polar functional monomers was demonstrated in this study as well as in our previous studies. Thus, the specific properties of the SnCl4 initiating system are discussed by comparing with the EtxAlCl3?x systems from viewpoints of hard and soft acids and bases principle and computational chemistry. © 2008 Wiley Periodicals, Inc. Four novel steroidal alkaloids named cortistatins E (1), F (2), G (3), and H (4) have been isolated from the marine sponge Corticium simplex. The chemical structures of these four cortistatins, which are unique abeo-9(10-19)-stigmastane-type steroidal alkaloids having oxabicyclo[3.2.1]octene and N-methyl piperidine or 3-methylpyridine units in the side chain, were elucidated by the detailed 2D-NMR analysis. These four compounds showed only weak anti-proliferative activity against human umbilical vein endothelial cells (HUVECs) at 0.35-1.9 μM concentrations in contrast to cortistatin A (5), which was isolated as a highly selective inhibitor of proliferation of HUVECs from the same marine sponge.  相似文献

Novel mono-O-alkyl-diglycosylglycerols named myrmekiosides A (1) and B (2) were isolated from a marine sponge of Myrmekioderma sp. and their absolute stereostructures were elucidated on the basis of chemical and physicochemical evidence. Myrmekiosides A (1) and B (2) reversed the phenotype of melanoma H-ras transformed NIH3T3 cells at 5 μg/ml.  相似文献

The absolute stereostructure of komodoquinone A (1), a neuritogenic anthracycline, which was isolated from a cultured marine Streptomyces sp. KS3, has been determined on the basis of chemical derivatization. Komodoquinone A (1) induces neuronal cell differentiation in the neuroblastoma cell line, Neuro 2A and arrests the cell cycle at the G1 phase. The effect of a solid-state medium on the production of 1 and its aglycone, komodoquinone B (2), was examined.  相似文献

Five new pregnene-type steroidal glycosides, named pregnedioside -a (1), 4′-O-acetyl-pregnedioside-a (2), 3′-O-acetyl-pregnedioside-a (3), pregnedioside-b (5), and 4′-O-acetyl-pregnedioside-b (6), were isolated from an Okinawan soft coral of Alcyonium sp. and their structures were elucidated.
