A generic strategy based on the use of CdSe/ZnS Quantum Dots (QDs) as elemental labels for protein quantification, using immunoassays with elemental mass spectrometry (ICP-MS), detection is presented. In this strategy, streptavidin modified QDs (QDs-SA) are bioconjugated to a biotinylated secondary antibody (b-Ab2). After a multi-technique characterization of the synthesized generic platform (QDs-SA-b-Ab2) it was applied to the sequential quantification of five proteins (transferrin, complement C3, apolipoprotein A1, transthyretin and apolipoprotein A4) at different concentration levels in human serum samples. It is shown how this generic strategy does only require the appropriate unlabeled primary antibody for each protein to be detected. Therefore, it introduces a way out to the need for the cumbersome and specific bioconjugation of the QDs to the corresponding specific recognition antibody for every target analyte (protein). Results obtained were validated with those obtained using UV–vis spectrophotometry and commercial ELISA Kits.  相似文献

Critical review of the atomic spectrometric analysis of arsenic in foods   总被引:1,自引:0,他引:1  
The literature on the determination of arsenic in foods by atomic spectroscopy involving hydride generation, platform furnace Zeeman and inductively coupled plasma has been critically reviewed. Hydride generation has been largely displaced as a powerful tool by platform furnace Zeeman, using the STPF (stabilized temperature platform furnace) concept for the control of interferences. Inductively coupled plasma with conventional pneumatic nebulization has been little used. An alternative is to employ hydride generation-ICP with extraction of the arsenic in an organic phase. This technique permits proper control of interferences together with an increase in sensitivity. Depending on the contents present in the foodstuff and the technique available, the sample can be solubilized by either wet or dry digestion.  相似文献

We present Raman spectroscopy experiments in dimethylacetylene (DMA) using a sapphire anvil cell up to 4 GPa at room temperature. DMA presents phase transitions at 0.2 GPa (liquid to phase I) and 0.9 GPa, which have been characterized by changes in the Raman spectrum of the sample. At pressures above 2.6 GPa several bands split into two components, suggesting an additional phase transition. The Raman spectrum of the sample above 2.6 GPa is identical to that found for the monoclinic phase II (C2/m) at low temperatures, except for an additional splitting of the band assigned to the fourfold degenerated asymmetric methyl stretch. The global analysis of the Raman spectra suggests that the observed splitting is due to the loss of degeneracy of the methyl groups of the DMA molecule in phase II. According to the above interpretation, crystal phase II of DMA extends from 0.9 GPa to pressures close to 4 GPa. Between 0.9 and 2.6 GPa, the methyl groups of the DMA molecules rotate almost freely, but the rotation is hindered on further compression.  相似文献

A warm standby n-system with operational and repair times following phase-type distributions is considered. The online unit goes through degradating levels, determined by inspections. Two types of repairs are performed, preventive and corrective, depending on the degradation level. The standby units undergo corrective repair. This systems is governed by a level-dependent-quasi-birth-and-death proces (LDQBD process), whose generator is constructed. The availability, rate of occurrence of failures, and other quantities of interest are calculated. A numerical example including an optimization problem and illustrating the calculations is presented. This system extend other previously studied in the literature.  相似文献

The current technology of air‐filtration materials for protection against highly toxic chemicals, that is, chemical‐warfare agents, is mainly based on the broad and effective adsorptive properties of hydrophobic activated carbons. However, adsorption does not prevent these materials from behaving as secondary emitters once they are contaminated. Thus, the development of efficient self‐cleaning filters is of high interest. Herein, we report how we can take advantage of the improved phosphotriesterase catalytic activity of lithium alkoxide doped zirconium(IV) metal–organic framework (MOF) materials to develop advanced self‐detoxifying adsorbents of chemical‐warfare agents containing hydrolysable P? F, P? O, and C? Cl bonds. Moreover, we also show that it is possible to integrate these materials onto textiles, thereby combining air‐permeation properties of the textiles with the self‐detoxifying properties of the MOF material.  相似文献

The objective of this study is to investigate the effects of high pressure processing on the molecular structure of some unsaturated fatty acids. Samples of elaidic acid, linoleic acid, ZE and EE conjugated linoleic acid are treated at 293 or 333?K at pressures up to 700?MPa. It is observed that the adiabatic heat generated from compression is able to bring the sample temperature above 373?K after 700?MPa. These relatively extreme conditions are of great interest for food sterilization, but they may induce undesirable change in fatty acid quality characteristics. To check for structural changes, Raman spectra of the samples are analysed after treatments. The comparison with Raman spectra of samples kept at atmospheric pressure shows that pressure induces some conformational changes at the hydrocarbon skeleton in solid samples, while the liquid ones remain unchanged. No cis/trans isomerization occurs, but gauche conformers are likely to be present.  相似文献

The arsenic species present in samples of the crayfish Procambarus clarkii caught in the area affected by the toxic mine‐tailing spill at Aznalcóllar (Seville, Southern Spain) were analyzed. The total arsenic contents ranged between 1.2 and 8.5 µg g?1 dry mass (DM). With regard to the different species of arsenic, the highest concentrations were for inorganic arsenic (0.34–5.4 µg g?1 DM), whereas arsenobetaine, unlike the situation found in marine fish products, was not the major arsenic species (0.16 ± 0.09 µg g?1 DM). Smaller concentrations were found of arsenosugars 1a (0.18 ± 0.11 µg g?1 DM), 1b (0.077 ± 0.049 µg g?1 DM), 1c (0.080 ± 0.089 µg g?1 DM), and 1d (0.14 ± 0.13 µg g?1 DM). The presence of two unknown arsenic species was revealed (U1: 0.058 ± 0.058 µg g?1 DM; U2: 0.12 ± 0.12 µg g?1 DM). No significant differences were seen with respect to the total arsenic contents between the sexes. However, significant differences in the total arsenic contents were revealed between the area affected by the spill and the area not affected, the contents being greater in the affected area.
