Immunochemical determination of dioxins in sediment and serum samples   总被引:3,自引:0,他引:3  
Polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) are considered highly toxic contaminants and the environmental and biological monitoring of these compounds is of great concern. Immunoassays may be used as screening methods to satisfy the growing demand for rapid and low cost analysis. In this work, we describe the application of an immunoassay that uses 2,3,7-trichloro-8-methyldibenzo-p-dioxin (TMDD) as a surrogate standard for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) to sediment and human serum samples. Sample extraction and preparation methods were developed with the aim to establish the simplest, cost-effective and efficient removal of the matrix interferences in the enzyme-linked immunosorbent assay (ELISA). The overall method for sediments is based on a hexane extraction; clean up by a multilayered silica gel column and an activated carbon column; an organic solvent exchange with DMSO–Triton X-100 and ELISA measurement. The gas chromatography–high resolution mass spectrometry (GC–HRMS) validation studies (n = 13) revealed that the method is suitable for the toxic equivalents (TEQ) screening of dioxin in sediments with a method detection limit of about 100 pg g−1 dry sediment with a precision of 13–33% R.S.D. The analysis of a large number of samples originating from different sources would be required to establish more precisely the screening level, as well as the number of false positives and negatives of dioxin TEQ by the immunoassay for sediments. The immunoassay method for sediment analysis offers improvement in speed, sample throughput, and cost in comparison to GC–HRMS. Dioxins were determined in serum samples after a simple liquid–liquid extraction and solvent exchange into DMSO–Triton X-100 without further dilution. The current method (approximate method LOQ of 200 pg ml−1 serum) is not sufficiently sensitive for the determination of dioxins in serum to measure acceptable exposure limit.  相似文献

Residual dipolar couplings (RDC) provide important global restraints for accurate structure determination by NMR. We show that nonuniform sampling in combination with maximum entropy reconstruction (MaxEnt) is a promising strategy for accelerating and potentially enhancing the acquisition of RDC spectra. Using MaxEnt-processed spectra of nonuniformly sampled data sets that are reduced up to one fifth relative to uniform sampling, accurate 13C'-13Calpha RDCs can be obtained that agree with an RMS of 0.67 Hz with those derived from uniformly sampled, Fourier transformed spectra. While confirming that frequency errors in MaxEnt spectra are very slight, an unexpected class of systematic errors was found to occur in the 6th significant figure of 13C' chemical shifts of doublets obtained by MaxEnt reconstruction. We show that this error stems from slight line shape perturbations and predict it should be encountered in other nonlinear spectral estimation algorithms. In the case of MaxEnt reconstruction, the error can easily be rendered systematic by straightforward optimization of MaxEnt reconstruction parameters and self-cancels in obtaining RDCs from nonuniformly sampled, MaxEnt reconstructed spectra.  相似文献

The type of hapten linkage to a carrier protein can play an important role in determining the nature of the resulting antibody response. Generic haptens using three types of linkers were synthesized (a monocarboxylic acid, an unsaturated hydrocarbon and a carboxamido spacer). These haptens were conjugated to bovine serum albumin (BSA) and used as immunogens to produce broad-specificity monoclonal antibodies (MAbs) to organophosphorus pesticides (OPs). Three-dimensional (3D) structures of hapten-lysine conjugates were optimized using molecular modeling (MM) to mimic conformations of hapten-BSA conjugates. The results from MM studies revealed a change of the 3D conformation and electrostatic potential of hapten 1 when the monocarboxylic acid linker was coupled to lysine. This result was consistent with the observed high-cross-reactivity of the corresponding MAb-H1 for the OPs. The competitive indirect enzyme-linked immunosorbent assay based on MAb-H1 is ideally suited to be used as a screening method for OP contaminants.  相似文献

Abstract

The temperature and enthalpy of melting for poly(ethy1ene oxide) have, for the first time, been studied as a fuction of pressure up to 1 GPa by means of differential scanning calorimetry. The initial increase of the temperature of melting with increasing pressure is 64 K/GPa, whereas the enthalpy decreases by 40% in the 1 GPa pressure range. Using Clausius-Clapeyrons equation the volume change on melting is estimated to be 1.5 cm3/mol. The glass transition temperature Tg for polystyrene has also been studied by the same technique for pressures up to 0.1 GPa. The measurements show that Tg increases with increasing pressure by 250 K/GPa.  相似文献

Functionalization of 2,1,3-benzothiadiazole (BTD) with thiols at C-5 position remains low explored. Moreover, the arylthiol-substitutions at this position are also unexplored and can not be found by a SN2 or SN1 reaction. In this sense, herein we present a new palladium-catalyzed methodology for a wide variety of unpublished 5-arylsulfanyl-benzo-2,1,3-thiadiazole derivatives synthesis with moderate to high yields using a low catalytic loading of Pd(L-Pro)2 as low-coast, and efficient catalyst in low reaction time. Besides, we concluded that the pKa of thiol species has an important role in this catalysis, mainly in the CMD like catalytic cyclo process, which strongly interferes in the reaction yields. Furthermore, arylsulfanyl-benzo-2,1,3-thiadiazoles derivatives have been assessed (in vitro) as potential acetylcholinesterase inhibitors.  相似文献

The stress-strain behavior in tension and the effect of temperature on the creep of poly(ether ether ketone) (PEEK) have been studied. At room temperature, 130° below the glass-transition temperature, the material does not become brittle, and the specimens show necking in tension over a wide range of elongation rates. The stress and strain at yield and the strain at break are almost linear functions of the logarithmic elongation rate. The values of stress and strain at yield increase slightly with increasing elongation rate, while the strain at break decreases markedly. The short-term creep tests were conducted at temperatures extending from 20 to 200°C. The glass-transition temperature was found to be about 155°C. The creep of PEEK is greatest at temperatures above 130°C. In the glass region the time dependence of the deformation is much weaker. It has been found that the time-temperature relation for PEEK corresponds well with its thermorheological simplicity in the temperature range investigated. The data on the temperature shift factor below and above the glass-transition temperature may be fitted separately to the Arrhenius and Williams-Landel-Ferry (WLF) equations, respectively. The long-term creep tests show that PEEK has excellent creep resistance at room temperature. After 14-month tests at a stress level of 30 MPa the total strain exceeds the instantaneous elastic strain only by a factor of 1.15.Institute of Polymer Mechanics. Multifunctional core-shell nanocomposites with a magnetic core and a silica shell doped with lanthanide chelate have been prepared by a simple method. First, citric acid-modified magnetite nanoparticles were synthesized by a chemical coprecipitation method. Then the magnetite nanoparticles were coated with silica shells doped with terbium (Tb3+) complex by a modified Stöber method based on hydrolyzing and condensation of tetraethyl orthosilicate (TEOS) and a silane precursor. These multifunctional nanocomposites are potentially useful in a variety of biological areas such as bio-imaging, bio-labeling and bioassays because they can be simultaneously manipulated with an external magnetic field and exhibit unique phosphorescence properties.  相似文献

A nanocrystalline TiO2 (anatase) nanosheet exposing mainly the (001) crystal faces was tested as photoanode material in dye-sensitized solar cells. The nanosheets were prepared by hydrothermal growth in HF medium. Good-quality thin films were deposited on F-doped SnO2 support from the TiO2 suspension in ethanolic or aqueous media. The anatase (001) face adsorbs a smaller amount of the used dye sensitizer (C101) per unit area than the (101) face which was tested as a reference. The corresponding solar cell with sensitized (001)-nanosheet photoanode exhibits a larger open-circuit voltage than the reference cell with (101)-terminated anatase nanocrystals. The voltage enhancement is attributed to the negative shift of flatband potential for the (001) face. This conclusion rationalizes earlier works on similar systems, and it indicates that careful control of experimental conditions is needed to extract the effect of band energetic on the current/voltage characteristics of dye-sensitized solar cell.  相似文献

All studies on oxygen-evolution reaction by Mn oxides in the presence of cerium(IV) ammonium nitrate (CAN) have been so far carried out by synthesizing Mn oxides in the first step. And then, followed by the investigation of the Mn oxides in the presence of oxidants for oxygen-evolution reaction (OER). This paper presents a case study of a new and promising strategy for in situ catalyst synthesis by the adding MnII to either CAN or KMnO4/CAN solution, resulting in the formation of Mn-based catalysts for OER. The catalysts were characterized by scanning electron microscopy, energy-dispersive spectroscopy, transmission electron microscopy, X-ray diffraction, Fourier transform infrared spectroscopy, X-ray absorption spectroscopy, and X-ray photoelectron spectroscopy. Both compounds contained nano-sized particles that catalyzed OER in the presence of CAN. The turnover frequencies for both catalysts were 0.02 (mmol /molMn⋅s).  相似文献
