Background
The common event in transmissible spongiform encephalopathies (TSEs) or prion diseases is the conversion of host-encoded protease sensitive cellular prion protein (PrPC) into strain dependent isoforms of scrapie associated protease resistant isoform (PrPSc) of prion protein (PrP). These processes are determined by similarities as well as strain dependent variations in the PrP structure. Selective self-interaction between PrP molecules is the most probable basis for initiation of these processes, potentially influenced by chaperone molecules, however the mechanisms behind these processes are far from understood. We previously determined that polymorphisms do not affect initial PrPC to PrPSc binding but rather modulate a subsequent step in the conversion process. Determining possible sites of self-interaction could elucidate which amino acid(s) or amino acid sequences contribute to binding and further conversion into other isoforms. To this end, ovine – and bovine PrP peptide-arrays consisting of 15-mer overlapping peptides were probed with recombinant sheep PrPC fused to maltose binding protein (MBP-PrP). 相似文献2.

Ferromagnetism in fullerene-based systems doped with metallocenes is reviewed. These compounds form a ferromagnetic state
by spin-coupling between π electrons on fullerene units, while the metallocene molecules do not contribute to the spin ordering.
One of these compounds has the highest critical temperature (19 K) for this class of compound. The magnetic properties of
these materials are very strongly dependent on the crystallization conditions.

The selective hydrolysis of proteins by non-enzymatic catalysis is difficult to achieve, yet it is crucial for applications in biotechnology and proteomics. Herein, we report that discrete hafnium metal-oxo cluster [Hf18O10(OH)26(SO4)13⋅(H2O)33] ( Hf18 ), which is centred by the same hexamer motif found in many MOFs, acts as a heterogeneous catalyst for the efficient hydrolysis of horse heart myoglobin (HHM) in low buffer concentrations. Among 154 amino acids present in the sequence of HHM, strictly selective cleavage at only 6 solvent accessible aspartate residues was observed. Mechanistic experiments suggest that the hydrolytic activity is likely derived from the actuation of HfIV Lewis acidic sites and the Brønsted acidic surface of Hf18 . X-ray scattering and ESI-MS revealed that Hf18 is completely insoluble in these conditions, confirming the HHM hydrolysis is caused by a heterogeneous reaction of the solid Hf18 cluster, and not from smaller, soluble Hf species that could leach into solution. 相似文献

We report on a study of the interactions between holes and molecular vibrations on dry DNA using photoinduced infrared absorption spectroscopy. Laser photoexcited holes are found to have a room-temperature lifetime in excess of tau > 1 ms, clearly indicating the presence of localization. However, from a quantitative model analysis of the frequency shifts of vibrational modes caused by the holes, we find the hole-vibrational coupling constant to be relatively small, lambda approximately 0.2. This interaction leads to a change in the conformational energy of DeltaE0 approximately 0.015 eV, which is too small to cause self-trapping at room temperature. We conclude that, at least in the dry (A) form, DNA is best understood in terms of a double chain of coupled quantum dots arising from the pseudorandom chain sequence of base pairs, in which Anderson localization prevents the formation of a metallic state. 相似文献

We present the results of a dc transport and optical investigation of WO3 and NaxWO3 with x
=0.01. Upon Na-doping we find a (Drude) metallic component in the optical conductivity, while the transport data display a crossover
from an activated to a variable range hopping regime around 210 K. We suggest the possible formation of polarons (and bipolarons)
and speculate that superconductivity could be induced, provided the dc percolation threshold is achieved.

We investigate the relaxation dynamics of nonequilibrium carriers in organic conductors κ-(BEDT-TTF)(2)Cu[N(CN)(2)]X (X=Br and Cl) using ultrafast time-resolved optical spectroscopy. The dynamics for both salts show similar temperature dependences, which is well characterized by the carrier relaxation across the pseudogap (PG) of the magnitude Δ(PG) ≈ 16 meV for Br salt and 7.0 meV for Cl salt. On the other hand, only the Br salt shows an abrupt increase of the decay time at low temperature, indicating an additional decay component associated with the superconducting (SC) gap below T(c). The fluence dependent dynamics at low temperature evidences the superposition of the SC component onto the PG component. These results indicate a metallic-insulating phase separation in the Br salt triggered by photoexcited nonequilibrium carriers. 相似文献

Metal/superconductor/semiconductor (Ag/Hg-1212/CdSe) hetero-nanostructures have been fabricated using pulse-electrodeposition
technique and are characterized by X-ray diffraction (XRD), full-width at half-maximum (FWHM) and scanning electron microscopy
(SEM) studies. The junction capacitance of Ag/Hg-1212, Hg-1212/CdSe and Ag/Hg-1212/CdSe heterojunctions is measured in dark
and under laser irradiation at room temperature. The nature of the junction formed and built-in-junction potentials were determined.
The increase in carrier concentration across the junction due to photo-irradiation has been observed. 相似文献

The electric conductivity is calculated for regular inhomogeneous two-component isotropic medium in which droplets of one phase with conductivity σ2 are embedded in another, with conductivity σ1. An expression is formulated that can be used in many different situations and is of particular relevance in the case where the relative proportion of the components is temperature-dependent and varies over a wide range. heteroepitaxial multilayer structures such as superconductor-semiconductor heterostructures. In the present investigation,
metal/superconductor/semiconductor (Ag/Tl-2223/CdSe) hetero-nanostructures have successfully been fabricated using dc electrodeposition
technique and were characterized by X-ray diffraction (XRD), full-width at half-maximum (FWHM) and scanning electron microscopy
(SEM) studies. The measurement of junction capacitance as a function of biasing voltage was used for the estimation of junction
built-in-potential (V
D) and to study the charge distribution in a heterojunction. The Mott-Schottky plots were measured for each junction in dark
