A novel method for the simultaneous quantification of both glutathione (GSH) and its oxidized form glutathione disulfide (GSSG) by hydrophilic interaction chromatography–MS/MS has been developed and is critically discussed. Internal standardization based on isotopically labeled standards for both analytes is an absolute prerequisite for accurate quantification of this redox pair. Hence, a highly efficient and selective miniaturized procedure for the synthesis of isotopically labeled GSSG from commercially available glutathione-(glycine-13C2,15N) was established using H2O2 as oxidant and NaI as catalyst. Moreover, a tool is presented to monitor and hence uncover artifactual GSSG formation due to oxidation of GSH during sample preparation, which is the main source of systematic error in GSSG analysis. For this purpose, we propose to monitor the oxidation product formed by reaction of naturally occurring GSH with the isotopically labeled GSH used as internal standard. For the determination of GSH/GSSG ratios in yeast, different extraction methods based on (1) hot extraction with aqueous, acidic, or organic solvents, (2) mechanical cell lysis, and (3) extraction at subambient temperature were investigated in terms of recovery, extraction efficiency, and artifactual formation of GSSG. Total combined uncertainties of as low as 25–30 % (coverage factor?=?2) for the determination of GSH/GSSG ratios without derivatization were made possible by the addition of the internal standards early in the analytical procedure (before extraction) and immediate analysis of the analytes. 相似文献

The cornea sits at the anterior aspect of the eye and, like the skin, is highly exposed to ultraviolet radiation (UVR). The cornea blocks a significant proportion of UVB from reaching the posterior structures of the eye. However, UVA can penetrate the full thickness of the cornea, even reaching the anterior portion of the lens. Epidemiological data indicate that UVR is a contributing factor for a multitude of diseases of the cornea including pterygium, photokeratitis, climatic droplet keratopathy and ocular surface squamous neoplasia (OSSN), although the pathogenic mechanisms of each require further elucidation. UVR is a well‐known genotoxic agent, and its effects have been well characterized in organs such as the skin. However, we are only beginning to identify its effects on the cornea, such as the UVR signature C → T and CC → TT transversions identified by sequencing and increased proliferative and shedding rates in response to UVR exposure. Alarmingly, a single low‐dose exposure of UVR to the cornea is sufficient to elicit genetic, molecular and cellular changes, supporting the consideration of using protective measures, such as wearing sunglasses when outdoors. The distribution of the isoscalar, T = 0, and isovector T = 1, strength of quadrupole modes in the deformed nucleus 24Mg is investigated. A finite-range residual interaction is used in the particle-hole basis of a rotating two-center shell-model potential of two 12C nuclei. The model calculation describes semi-quantitatively the main features of the available experimental data for the T = 0, E2 excitations and predicts the location and structure of the isovector, T = 1, E2 strength for which no data has been published to date. 相似文献

In this paper we construct a finitely based variety, whose equational theory is undecidable, yet whose word problems are recursively solvable, which solves a problem stated by G. McNulty (1992). The construction produces a discriminator variety with the aforementioned properties starting from a class of structures in some multisorted language (which may include relations), axiomatized by a finite set of universal sentences in the given multisorted signature. This result also presents a common generalization of the earlier results obtained by B. Wells (1982) and A. Mekler, E. Nelson, and S. Shelah (1993).

The reaction 40Ca(16O, 15N)41Scg.s. has been investigated in the context of a model where the final single-particle state in 41Sc was modified or polarized due to the proximity of the other nucleus. Calculations, performed with the finite-range distorted waves Born approximation code KUNDRY, using two-centre shell model wavefunctions, demonstrate that this polarization produces effects on the cross section which indicate a resolution to some discrepancies between previous DWBA predictions and experiment. 相似文献

We investigate properties of the endomorphism monoid of the countable random graph R . We show that End(R) is not regular and is not generated by its idempotents. The Rees order on the idempotents of End(R) has 2N0 many minimal elements. We also prove that the order type of Q is embeddable in the Rees order of End(R) . 相似文献
