Negative-ion fast atom bombardment mass spectrometry and metastable ion studies were applied to the characterization of three isomeric fluorine-containing sugars. Peculiar fragmentation processes were evidenced for each isomer, enabling their differentiation. Comparison with the behaviour of a non-fluorinated analogue allowed investigation of the role of fluorine in the decomposition processes of these compounds.  相似文献

Biodetection is one of the most important challenges for the twenty-first century: many fields are concerned, mainly environmental and medical. The quartz crystal microbalance (QCM) may offer great possibilities for this purpose: a direct response signal, which characterizes the binding event between a sensitive layer, immobilized onto the surface transducer, and the analyte to be detected, can be obtained. However, for the detection of small biomolecules such as antigens, it is quite difficult to obtain an observable signal that corresponds directly to the binding event. In general, this is owing to the lack of mass sensitivity of the commonly used QCM, with 5- to 10-MHz quartz crystals. For improving this mass sensitivity, a 27-MHz quartz resonator was developed and incorporated in a flow-through microcell. Two biospecies, IgG rabbit and peroxidase enzyme, were studied with this ultra-sensitive QCM in terms of specificity, detection limit, and calibration curve.  相似文献

The gas-liquid chromatographic fractionation of the commercial biodegradable surfactants RO(CH2CH2O) H (where R is an n-alkyl group and is the number-average degree of polymerization) can be approached only for 4, as the truncation of the molecular-weight distribution occurs at higher values. Although conversion into trimethylsily (TMS) derivatives improves the volatility of the oligomers, there is only a slight advantage in using this conversion procedure: TMS derivatives have the same elution temperature as untreated compounds above ca. 330°. The use of an internal standard as a reference compound for comparison of peak areas is a useful procedure, which has been tentatively applied to the reconstruction of the distribution of the surfactant with R = n-dodecyl and = 7.1. The choice of the internal standard is discussed.  相似文献

Partially modified retro- (PMR) and retro-inverso (PMRI) psi[NHCH(CF(3))]Gly peptides, a conceptually new class of peptidomimetics, have been synthesized in wide structural diversity and variable length by aza-Michael reaction of enantiomerically pure alpha-amino esters and peptides with enantiomerically and geometrically pure N-4,4,4-trifluorocrotonoyl-oxazolidin-2-ones. The factors underlying the observed moderate to good diastereocontrol have been investigated. The conformations of model PMR-psi[NHCH(CF(3))]Gly tripeptides have been studied in solution by (1)H NMR spectroscopy supported by MD calculations, as well as in the solid-state by X-ray diffraction. Remarkable stability of turn-like conformations, comparable to that of parent malonyl-based retropeptides, was evidenced, as a likely consequence of two main factors: 1) severe torsional restrictions about sp(3) bonds in the [CO-CH(2)-CH(CF(3))-NH-CH(R)-CO] module, which is biased by the stereoelectronically demanding CF(3) group and the R side chain; 2) formation of nine-membered intramolecularly hydrogen-bonded rings, which have been clearly detected both in CHCl(3) solution and in some crystal structures. The former factor seems to be more important, as turn-like conformations were found in the solid-state even in the absence of intramolecular hydrogen bonding. The relative configuration of the -C*H(CF(3))NHC*H(R)- stereogenic centers has a major effect on the stability of the turn-like conformation, which seems to require a syn stereochemistry. X-ray diffraction and ab initio computational studies showed that the [-CH(CF(3))NH-] group can be seen as a sort of hybrid between a peptide bond mimic and a proteolytic transition state analogue, as it combines some of the properties of a peptidyl -CONH- group (low NH basicity, CH(CF(3))-NH-CH backbone angle close to 120 degrees, C-CF(3) bond substantially isopolar with the C=O) with some others of the tetrahedral intermediate [-C(OX)(O(-))NH-] involved in the protease-mediated hydrolysis reaction of a peptide bond (high electron density on the CF(3) group, tetrahedral backbone carbon).  相似文献

A theoretical study of the mechanisms associated with the synthesis of anticonvulsant symmetric N,N-substituted sulfamides is presented. Two possible synthetic routes are compared, which mainly differ in the use of pyridine as a nucleophilic agent in the reaction mechanism. Aldol reactions of titanium enolates of N-acyl-1,3-oxazolidin-2-ones with ethyl trifluoropyruvate occurred with low to good stereoselectivity depending on the steric properties of the N-acyl group. Attempts to transform the resulting aldols into peptidomimetics incorporating stereo-defined α-trifluoromethyl (Tfm)-malic units are described.  相似文献

The action of acyl halides on heterocyclic compounds of five-membered rings containing an O-M-O (M ? P, As, Sb) linkage is described. The reactions were either carried out in the presence of a solvent (benzene or toluene) or by direct heating of the reagent with the substrate. In the case of arsole, stibole and stannole derivatives, the cleavage of O-M bond and the formation of the respective mono- and diesters were always obtained, while with the phospholes, no cleavage of the compounds was observed. The products IIa-c (M = Sb) have been obtained in excellent yields starting from VIa-c and antimony trichloride. The structure of the compounds which were prepared was determined by analytical and spectroscopic methods and also by comparations with authentic samples where possible.  相似文献

Six novel metabolites, acremines A-F have been isolated from agar cultures of a strain of Acremonium sp. Their structures and stereochemistry were elucidated using a combination of 13C and 1H homo and heteronuclear 2D NMR experiments and X-ray analysis. The (3R,5S,6E,8S,10R)‐11‐amino‐3,5,8,10‐tetramethylundec‐6‐enoic acid (ATUA; 1 ), which was designed as a βII′‐turn mimic according to the concepts of allylic strain and 2,4‐dimethylpentane units, was incorporated into a cyclic RGD peptide. The three‐dimensional structure of cyclo(‐RGD‐ATUA‐) (=cyclo(‐Arg‐Gly‐Asp‐ATUA‐)) 4 in H2O was determined by NMR techniques, distance geometry calculations and molecular‐dynamics simulations. The RGD sequence of 4 shows high conformational flexibility but some preference for an extended conformation. The structural features of the RGD sequence of 4 were compared with the RGD moiety of cyclo(‐RGDfV‐) (=cyclo(‐Arg‐Gly‐Asp‐D ‐Phe‐Val‐)). In contrast to cyclo(‐RGDfV‐), which is a highly active αvβ3 antagonist and selective against αIIbβ3, cyclo(‐RGD‐ATUA‐) shows a lower activity and selectivity. The structure of the ATUA residue in the cyclic peptide resembles a βII′‐turn‐like conformation. Its middle part, adjacent to the C?C bond, strongly prefers the designed and desired structure.  相似文献
