First-derivative spectrophotometric method with a “zero-crossing” technique of measurement has been used for the quantification of two-components mixtures of cephalothin and clavulanic acid. As the absorption bands of these drugs overlap, both direct and derivative spectrophotometric methods have been investigated and evaluated by a rigorous statistical analysis of the experimental data. The first-derivative spectrophotometric method was found to be more accurate, direct and reproducible. Beer's law was valid over the concentration range 2.0–28.0 mg/l for both compounds. The detection limits of cephalothin and clavulanic acid, at a 0.05 level of significance, were calculated to be 0.13 and 0.15 mg/l. The method was applied for determining these antibiotics in mixtures, some of them containing inject-able dosage forms of cephalothin, and so to determine both compounds in saline and glucosed physiological sera. 相似文献

Summary A new, rapid and simple method is described and applied to resolve and quantify mixtures of prednisolone, Zn-bacitracin and phenylephrine. The determination was accomplished by MEKC. The separation was carried out at 25 °C and 30 kV, using a 5 mM phosphate-5 mM borate buffer (pH=8.2), 40 mM SDS as background electrolyte. Under these conditions, the run time was 6.6 min and the limits of quantification were about 1.0 mg L–1 for every component. Repeatability and reproducibility studies were achieved showing no significant differences at 95% confidence level. The MEKC method has been applied for quantifying these compounds in different commercials pharmaceuticals products, without separations steps. 相似文献

A fluorescence method involving sample pre-treatment is investigated concerning the determination of cefoxitin. A fluorescent product is formed when samples containing cefoxitin are subjected to alkaline hydrolysis with 1.0M sodium hydroxide and heated for 60 min at 90 degrees . The fluorescence is measured in ethanol/water medium (50% v/v) at approximately pH 2.0 provided by adding of 0.1M hydrochloric acid. The fluorescence excitation and emission maxima were 317 and 400 nm, respectively. The quantitative range is between 0.020 and 1.40 mug/ml. A detection limit of 2 x 10(-3) mug/ml was found. The proposed method has been applied to the determination of cefoxitin in commercial injections, saline and glucosed physiological serum. 相似文献

Voltammetric behavior of zaleplon and its differential pulse polarographic determination in capsules

In this work both the electrochemical behavior and the analysis of the hypnotic pyrazolopyrimidine derivative zaleplon were studied. Zaleplon in ethanol-0.1M Britton Robinson buffer solution (30-70) showed 2 irreversible, well-defined cathodic responses in the pH range of 2-12 using differential pulse polarography (DPP), tast polarography, and cyclic voltammetry. From chronocoulometric studies, it was possible to conclude that one electron was transferred in each reduction peak or wave. For analytical purposes, the DPP technique working at pH 4.5 for peak I was selected, which exhibited adequate repeatability, reproducibility, and selectivity. The recovery was 99.97 +/- 1.52%, and the detection and quantitation limits were 5.13 x 10(-7)M and 1.11 x 10(-6)M, respectively. The DPP method was applied successfully to the individual assay of capsules in order to verify the content uniformity of zaleplon. Treatment of the sample is not required because the excipients do not interfere, the method is not time consuming, and it is less expensive than column liquid chromatography. 相似文献

Coordination compounds of copper have been invoked as major actors in processes involving the reduction of molecular oxygen, mostly with the generation of radical species the assignment for which has, so far, not been fully addressed. In the present work, we have carried out studies in solution and on surfaces to gain insights into the nature of the radical oxygen species (ROS) generated by a copper(II) coordination compound containing a thioether clip‐phen derivative, 1,3‐bis(1,10‐phenanthrolin‐2‐yloxy)‐N‐(4‐(methylthio)benzylidene)propan‐2‐amine (2CP‐Bz‐SMe), enabling its adsorption/immobilization to gold surfaces. Whereas surface plasmon resonance (SPR) and electrochemistry of the adsorbed complex indicated the formation of a dimeric CuI intermediate containing molecular oxygen as a bridging ligand, scanning electrochemical microscopy (SECM) and nuclease assays pointed to the generation of a ROS species. Electron paramagnetic resonance (EPR) data reinforced such conclusions, indicating that radical production was dependent on the amount of oxygen and H2O2, thus pointing to a mechanism involving a Fenton‐like reaction that results in the production of OH.. 相似文献

The aim of the work described in this paper was to provide a fast, easy, inexpensive, precise, and accurate method for the determination of melatonine in solid pharmaceutical dosage forms. The developed method is based on a UV first-derivative spectrophotometric determination, which exhibits excellent linearity in aqueous solutions (r2 = 0.996) for analyte concentrations of 1.5-4.5 mg/dL within a pH range of 5-9. Neither excipients present in the formulation nor indole adulterants, such as tryptophan (up to 5%), interfere with the assay. A study of variation parameters showed that sonication temperature was the main factor for successful determination. At temperatures of <45 degrees C, the sample dissolved completely, and accurate spectrophotometric measurements were obtained. A study was conducted of all the parameters established by the United States Pharmacopeia, 23rd Rev., to validate an analytical method for a solid pharmaceutical form, i.e., linearity, range, accuracy, precision, and specificity. All the parameters were in accordance with the acceptance criteria of the Comité de Guías Oficiales de Validación de la Dirección General de Control de Insumos para la Salud de Méjico. The design, synthesis, and use of two new, stable, functionalized chain transfer agents (CTA's) containing OH and amine end groups for the RAFT polymerization is reported: 2‐hydroxyethoxy‐carbonylphenylmethyl dithiobenzoate and 2‐(2‐(tert‐butoxycarbonyl)ethylamino)‐2‐oxo‐1‐phenylethyl benzodithioate, respectively. The RAFT polymerization of n‐hexyl acrylate (HA) using those CTA's, were compared to several other functionalized dithiobenzoate esters reported in the literature containing COOH and Ester groups. The performances of the dithiobenzoates were compared in terms of kinetics and molecular weight distribution control. Good control in polymerization of n‐hexyl acrylate with a linear increase of Mn with conversion mantaining polydispersity indices (PDI) below 1.1 was obtained by use of the new functionalized CTA's developed and also by use of some other CTA's tested, to produce well‐defined linear polymers with one specific chain‐end functionality: ? OH, ? COOH or Amine. Using a postpolymerization reaction with functionalized azocompounds in a 5 to 1 ratio, α,ω‐telechelic polymers, with ? OH or ? COOH as functional group at the second end were obtained. By using this synthetic strategy α,ω‐homotelechelic and heterotelechelic polymers were readily prepared. The chemical availability of functional end‐groups in the telechelics was demonstrated by reaction with methacrylic anhydride. © 2010 Wiley Periodicals, Inc.
