A range of benzylaminopurines naturally occur in plants and exhibit high biological activity. Others have been synthesized, such as 6-(2-hydroxy-3-methoxybenzylamino)purine riboside (2OH3MeOBAPR), which has shown interesting anti-cancer activity under in vitro conditions. In order to study the biological activity of this interesting compound in more detail, a rapid and highly efficient method for its purification from complex samples (e.g. blood and plant extracts) is needed. Therefore, we prepared monoclonal antibodies against 2OH3MeOBAPR. The antibody had undetectable cross-reactivity with all natural isoprenoid cytokinins, but relatively high cross-reactivity with aromatic cytokinins as well as some synthetic di- and tri-substituted 6-benzylaminopurines and the corresponding ribosides. The antibody also showed strong responses and specificity in enzyme-linked immunoassays (ELISAs). In addition, it was used to prepare, for the first time, an immunoaffinity sorbent with high specificity and capacity for aromatic cytokinins. A batch immunoextraction method was then developed and optimized for the purification of 2OH3MeOBAPR from murine blood samples. The high efficacy and simplicity of this method (in off-line combination with HPLC-MS) for the isolation of target analytes from biological material is demonstrated in this study.  相似文献

The synthesis and physicochemical properties of novel porphyrazines possessing an alternate system of two peripheral substituents, 2,5-dimethylpyrrol-1-yl and dimethylamino, are presented. All the macrocycles were subjected to HPLC purity studies. Spectroscopic studies of magnesium(II) porphyrazine encompassed steady state absorption, emission measurements, including fluorescence decays, transient absorption spectra, and thermoluminescence. Additionally, magnesium(II) porphyrazine was found to be a moderate photosensitizer with singlet oxygen generation values of 0.12 and 0.14 in DMF and DMSO, respectively. Comparison of the quantum yields of singlet oxygen generation before and after deoxygenation showed that the photodynamic effect of magnesium(II) porphyrazine is governed by the photosensitization mechanism II. Magnesium(II) and manganese(III) porphyrazines were characterized using X-ray crystallography.  相似文献

Total synthesis of the natural d-ribo-phytosphingosine I and its 2-epimer III in the protected form was achieved through a common strategy. The aza-Claisen rearrangement of allylic thiocyanate (Z)-V incorporated the new stereogenic centre with nitrogen and the subsequent Wittig olefination constructed a non-polar side chain. Hydrogenation, followed by removal of protecting groups, completed the syntheses of I and III.  相似文献

The iron-porphyrin aggregates were studied by optical absorption and fluorescence method, infrared spectroscopy, X-ray absorption and Mössbauer spectroscopy. The aggregation of porphyrin molecules strengthens the Fe-ligands bonds and accelerates the spin-spin relaxations. A significant speeding-up of relaxation was observed with lowering the temperature down to 25 K. The comparison of the EXAFS (Extended X-ray Absorption Fine Structure) and Mössbauer spectroscopy results enabled some separation of the individual Fe vibration from its collective movement with ligands.  相似文献

We studied radiation effects on the stability of various states of hemoglobin (Hb) in red blood cells (RBC) irradiated with a very low dose of neutron rays, 50?μGy. We investigated RBCs isolated from blood of healthy donors. M?ssbauer spectroscopy was applied to monitor different forms of Hb. Our results show, for the first time, that oxyhemoglobin (OxyHb) and deoxyhemoglobin (DeoxyHb) are two Hb forms sensitive to such a low neutron radiation. Both Hbs change into a new Hb form (Hbirr). Additionally, OxyHb transfers into HbOH/H2O, which under our experimental conditions is resistant to the action of neutron rays.  相似文献

A common strategy for the total syntheses of the protected l-arabino- and l-ribo-C18-phytosphingosine (8 and 9, respectively), HCl salts of ent-2-epi-jaspine B (ent-6) and 3-epi-jaspine B (7) with efficient use of both flexible building blocks 26 and 27 was achieved. The key step of this approach was [3,3]-sigmatropic rearrangement of allylic trichloroacetimidate 21 and thiocyanate 22, which were derived from the known 2,3:5,6-di-O-isopropylidene-d-mannofuranose 18 as the source of chirality. The side chain functionality was installed utilizing a Wittig reaction.  相似文献
