Synthesis and biological activities of analogs of a lipid A biosynthetic precursor: 1-O-phosphonooxyethyl-4'-O-phosphono-disaccharides with (R)-3-hydroxytetradecanoyl or tetradecanoyl groups at positions 2, 3, 2' and 3'.

Two novel analogs of a biosynthetic precursor of lipid A (2) were synthesized. The one analog (3) has acyl groups identical to those of 2, and the other (4) has tetradecanoyl groups in place of the (R)-3-hydroxytetradecanoyl groups of 2. Both 3 and 4 possess an alpha-glycosidically-bound phosphonooxyethyl group in place of the alpha-glycosyl phosphate group of 2. Compounds 3 and 4 exhibited definite antitumor activity against Meth A fibrosarcoma and low toxicity in rabbits, as the original compound 2 does. The replacement of the hydroxytetradecanoyl groups with tetradecanoyl groups barely affected the antitumor activity, but slightly enhanced the toxicity in rabbits.     

Simple and rapid determination of hydrogen peroxide using phosphine-based fluorescent reagents with sodium tungstate dihydrate.

A simple batch method for the fluorometric determination of hydrogen peroxide using phosphine-based fluorescent reagents has been developed. A rapid, mild and selective derivatization reaction was achieved by adding sodium tungstate dihydrate to the reaction mixture of hydrogen peroxide and a phosphine-based fluorescent reagent. When 4-diphenylphosphino-7-methylthio-2,1,3-benzoxadiazole was used as a reagent, the derivatization reaction was completed after 2 min at room temperature. The calibration curve was linear between 12.5 and 500 ng hydrogen peroxide in a 10 microL sample solution. This method is accurate and has potential for on-line applications.     

Photo-initiated base-formation in a polymer matrix

It was found that amines were formed efficiently by the photolysis of O-acyloximes followed by hydrolysis in polystyrene films and the relationship between structures of O-acyloximes and yields of amines were investigated. O-phenylacetyl acetophenone oxime (PaApO), O-pivaloyl acetophenone oxime (PApO), and O-benzoyl acetophenone oxime (BApO) as monofunctional O-acyloximes and O,O′-succinyl diacetophenone oxime (SDApO) and O,O′-glutaryl diacetophenone oxime (GDApO) as bifunctional O-acyloximes were examined. The yields of amines for PaApO and SDApO under N₂ were ca. 70%, which was the highest among O-acyloximes examined in this experiment. On the other hand, the yields for PApO, BApO, and GDApO were less than 15% and it was verified that the hydrogen abstraction by imino radicals via 6-membered cyclic intermediates resulted in the lowering of yields. Although the effect of oxygen under photolysis on the yields of amine for PaApO was negligible under 50% conversion of PaApO, the yield decreased with further increase in the conversion and was 50% at 90% conversion. © 1994 John Wiley & Sons, Inc.     

Significance of anti-HBc IgM in acute hepatitis and hepatitis B associated chronic liver disease

To determine the diagnostic value of anti-HBc IgM in acute viral hepatitis or chronic liver disease B, Anti-HBc IgM was measured by a RIA and an ELISA in 32 patients with acute hepatitis (4 with type A, 15 with type B and 13 with type non A non B), 18 patients with chronic hepatitis and 19 patients with liver cirrhosis. In acute hepatitis B, anti-HBc IgM (both RIA and ELISA) was positive in 14(93%) of 15 patients and its cut-off index value was very high. However, anti-HBc IgM was always negative in one patient with typical course of type B. In 1 of 4 patients with acute hepatitis A and 2 of 13 with non A non B, anti-HBc IgM (RIA and/or ELISA) was positive. These 3 patients were positive for anti-HBs at the onset of disease, so we could not made the diagnosis of acute hepatitis B. Anti-HBc IgM was positive in 21(51%) of 37 patients with HBsAg-positive chronic liver disease by RIA and in 11 (30%) by ELISA, and its cut-off index value was relatively low. These results suggest that when adequate cut-off index value is established, anti-HBc analysis is useful for differentiating recent and current infections from remote infections.     

Total synthesis of (+)-tubelactomicin A. 2. Synthesis of the upper-half segment and completion of the total synthesis

[reaction: see text]. We have completed the total synthesis of natural (+)-tubelactomicin A (1), a 16-membered macrolide antibiotic. This Letter presents a highly efficient synthesis of the upper-half segment (C14-C24) and the completion of the total synthesis featuring a high-yielding Stille coupling for the connection of the upper-half and lower-half segments and Mukaiyama macrolactonization for the construction of the entire structure of 1.     

Selective wet air oxidation of diluted aqueous ammonia solutions over co-precipitated transition metal-aluminium catalysts

Catalytic wet air oxidation of ammonia over a co-precipitated transition metal-aluminium catalyst was investigated. Copper-aluminium (Cu-Al-O) catalyst exhibited the highest activity and N₂ selectivity among those prepared from Co, Fe, Mn, and Ni. 50% of 1500 ppm of ammonia could be removed from wastewater of pH 12 at 503 K under 2.0 MPa of air by using 4.0 g of catalyst without formation of toxic nitrogen containing compounds. Cu and Al ions were not found in solution after the reaction. It has been found that the catalytic performance of Cu-Al-O catalyst was strongly dependent on the preparation methods. The co-precipitated Cu-Al-O catalysts showed high N₂ selectivity. The presence of CuO is concluded to promote the reaction and CuAl₂O₄ in bulk phase is needed to stabilize the catalyst.     

Chemical structure of escherichia coli lipid A

The chemical structure of E. coli lipid A was elucidated to be $\text{2}$ by determination of the nature of the individual acyl groups bound to the two hydroxyl groups in positions 3,3′ and the two amino groups of the D-glucosamine disaccharide phosphate backbone.     

Conformation of poly(<Emphasis Type="SmallCaps">l</Emphasis>-glutamic acid) in bipolar amphiphiles with alkyl ammonium cations of different hydrophobicity

The conformation of the sodium salt of poly(l-glutamic acid) (P(Glu)) in solutions of the cationic bipolar amphiphile 1, 20-isosanediylbis(alkylammonium chloride) (C ₂₀(RA) ₂) with different alkyl head groups as a function of amphiphile concentration was investigated using circular dichroism (CD). RA included methylammonium (MA), ethylammonium (EA), propylammonium (PA), butylammonium (BA), and pentylammonium (PeA) cationic groups. The CD spectrum of each C ₂₀(RA) ₂ had a double minima corresponding to the a-helix of P(Glu), which was replaced by CD spectra with a single minimum at wavelengths larger than 222 nm as concentration increased. These changes in the CD spectrum were ascribed to the conformational change from random coil to a-helix and to aggregates of helices. In C ₂₀(EA) ₂ solutions, a step-like change in the CD intensity was observed at 222 nm as a function of the ratio of C ₂₀(RA) ₂ to P(Glu). At the step, the CD spectrum of the complete a-helix was observed. At 10–35 °C, an a-helix was induced in P(Glu) in the order: C ₂₀(EA) ₂>C ₂₀(MA) ₂>C ₂₀(PA) ₂>C ₂₀(BA) ₂>C ₂₀(PeA) ₂. This order was ascribed to the best fit of ethylammonium to the P(Glu) side chain.     

Effect of polymer on electron-donating character of polymer donor. II. Formation of charge-transfer complex between polymers containing N-dimethylaniline group and trinitrotoluene

Formation of charge-transfer complexes with trinitrotoluene (TNT) as a common acceptor was studied in detail by using dimethyltoluidine (DMT), poly-N-dimethyl-p-aminostyrene (poly-ASt), and also copolymers of aminostyrene (ASt) and styrene (St) as donors. A smooth bathochromic shift in λ_max was observed with increasing ASt unit content in copolymers. Values of the constant for charge transfer complex formation K_CT were found to increase smoothly with ASt unit content. However, the K_CT value with DMT did not coincide with the value extrapolated from the plot of K_CT value versus ASt unit content to zero ASt unit content, but was found to be much higher than the limiting value, in contradiction to the results obtained with maleic anhydride (MAnh). The entropy of complex formation with DMT was found to be exceptionally small; this small value may be responsible for the high K_CT value with DMT.
