Enzymes that degrade pectin are called pectinases. Pectinases of microbial origin are used in juice clarification as the process is cost-effective. This study screened a pectinase-producing bacterium isolated from soil and identified as Bacillus subtilis 15A B-92 based on the 16S rRNA molecular technique. The purified pectinase from the isolate showed 99.6 U/mg specific activity and 11.6-fold purity. The molecular weight of the purified bacterial pectinase was 14.41 ± 1 kD. Optimum pectinase activity was found at pH 4.5 and 50 °C, and the enzyme was 100% stable for 3.5 h in these conditions. No enzymatic inhibition or activation effect was seen with Fe2+, Ca2+, or Mg2+. However, a slight inhibition was seen with Cu2+, Mn2+, and Zn2+. Tween 20 and 80 slightly inhibited the pectinase, whereas iodoacetic acid (IAA), ethylenediaminetetraacetate (EDTA), urea, and sodium dodecyl sulfate (SDS) showed potent inhibition. The bacterial pectinase degraded citrus pectin (100%); however, it was inactive in the presence of galactose. With citrus pectin as the substrate, the Km and Vmax were calculated as 1.72 mg/mL and 1609 U/g, respectively. The high affinity of pectinase for its substrate makes the process cost-effective when utilized in food industries. The obtained pectinase was able to clarify orange and apple juices, justifying its application in the food industry.  相似文献

Abstract

This study focuses on isolation of pigment producing bacteria from fresh water sediment. The isolated bacteria were grown in nutrient broth and the maximum absorbance of 2.512 was obtained for the extracted pigment at 500 nm. The effective strains were optimized, pH 11 and temperature 30 °C was found to be more favorable for its maximum growth. The isolates were identified based on their molecular characterestics as Microbacterium arborescens and Sphingomonas paucimobilis, molecular size of the amplified 16S rRNA gene sequence was found to be approximately 1270 and 765 bp respectively. The antioxidant property of the pigment was analyzed using DPPH and ABTS assay. The IC50 value of Microbacterium arborescens was higher in all the three assays in comparison with Sphingomonas paucimobilis. The extracted pigment was characterized for the presence of compounds using GC-MS and FTIR analysis to determine the functional groups. As the pigment obtained from M. arborescens had shown better antioxidant activity it may be used as colorant in food industrial applications.  相似文献

The combination of light activation and N‐heterocyclic carbene (NHC) organocatalysis has enabled the use of acid fluorides as substrates in a UVA‐light‐mediated photochemical transformation previously observed only with aromatic aldehydes and ketones. Stoichiometric studies and TD‐DFT calculations support a mechanism involving the photoactivation of an ortho‐toluoyl azolium intermediate, which exhibits “ketone‐like” photochemical reactivity under UVA irradiation. Using this photo‐NHC catalysis approach, a novel photoenolization/Diels–Alder (PEDA) process was developed that leads to diverse isochroman‐1‐one derivatives.  相似文献

Noble metal nanoparticles like Au have long been admired for their brilliant colour, significantly influenced by plasmon resonance. When embedded in metal oxides, they exhibit unique properties which make them an excellent choice for sensing in high-temperature and harsh environment atmospheres. In this review, the various morphologies of Au nanoparticles (AuNPs) used in combination with metal oxides for sensing gases at temperatures greater than 300 °C are discussed. Theoretical discussions on the plasmon resonance properties of AuNPs as well as computational techniques like finite difference time domain (FDTD), are often used for understanding and correlating their extinction spectra and are briefed initially. The sensing properties of AuNPs embedded on a metal oxide matrix (such as TiO2, SiO2, NiO etc) for quantifying multiple analytes are then elucidated. The effect of high temperature as well as gas environments including corrosive atmospheres on such nanocomposites, and the different approaches to comprehend them are presented. Finally, techniques and methods to improve on the challenges associated with the realization and integration such Au-metal oxide plasmonic nanostructures for applications such as combustion monitoring, fuel cells, and other applications are discussed.  相似文献

Linde Type A (LTA) zeolites have been synthesized in the current study by simple sol–gel technique. The crystal growth has been controlled by varying the hydrogel synthesis time and annealing temperature. The resulting products obtained at various crystallization times and temperatures have been studied using X-ray powder diffraction (XRD) method, High resolution transmission electron microscopy images, scanning electron microscopy (SEM) micrographs, energy dispersive study and Brunauer–Emmett–Teller (BET) analysis. The TEM images of the final LTA zeolite annealed at 500 °C revealed the formation of cubic structure. XRD analysis revealed that the crystallinity improved with annealing. BET analysis revealed that the synthesized LTA is a well crystallized 4A zeolite. LTA zeolites were dispersed in poly ethylene glycol in the ratio 3:100 and 5:100 and coated on porous alumina beads for the formation of membrane. The SEM images revealed excellent formation of fine structure LTA zeolite membrane with uniform coating. The membrane consisted of a top layer with thickness of 1.14–2.0 µm. Crystals in the top layer showed cubic morphology and amorphous phase was observed at the grain boundaries present between LTA zeolite and alumina substrate.  相似文献
