A mild, efficient and convenient extraction method of using 2-mercaptoethanol contained extractant solution combined with an incubator shaker for determination of mercury species in biological samples by HPLC–ICP-MS has been developed. The effects of the concentration of 2-mercaptoethanol, the composition of the extractant solution and the shaking time on the efficiency of mercury extraction were evaluated. The optimization experiments indicated that the quantitative extraction of mercury species from biological samples could be achieved by using 0.1% (v/v) HCl, 0.1% (v/v) 2-mercapoethanol and 0.15% (m/v) KCl extractant solution in an incubator shaker for shaking overnight (about 12 h) at room temperature. The established method was validated by analysis of various biological certified reference materials, including NRCC DOLT-3 (dogfish liver), IAEA 436 (tuna fish), IAEA MA-B-3/TM (garfish filet), IAEA MA-M-2/TM (mussel tissue), GBW 08193 (bovine liver) and GBW 08572 (prawn). The analytical results of the reference materials were in good agreement with the certified or reference values of both methyl and total mercury, indicating that no distinguishable transformation between mercury species had occurred during the extraction and determination procedures. The limit of detection (LOD) for methyl (CH3Hg+) and inorganic mercury (Hg2+) by the method are both as 0.2 μg L−1. The relative standard deviation (R.S.D.s) for CH3Hg+ and Hg2+ are 3.0% and 5.8%, respectively. The advantages of the developed extraction method are that (1) it is easy to operate in HPLC–ICP-MS for mercury species determination since the extracted solution can be directly injected into the HPLC column without pH adjustment and (2) the memory effect of mercury in the ICP-MS measurement system can be reduced. 相似文献

通过熔融共混法制备了聚乳酸/微生物产β-羟基丁酸酯与β-羟基己酸共聚物的共混物(PLA/PHBHHx)。采用拉伸力学试验研究了共混物的力学性能。通过土壤悬浊培养降解法和扫描电子显微镜(SEM)分析对共混材料的生物降解性能进行了研究。实验结果表明,随着PHBHHx含量的增加,共混物的拉伸强度和杨氏模量降低,而生物降解速率却显著提高。但是,在175h之前,重量组成比为20/80的共混物降解速率比纯PHBHHx还要快。综合分析表明,共混材料PLA/PHBHHx的重量比为20/80时,具有优良的力学性能和生物降解性。 相似文献

Selective chemical modification of proteins plays a pivotal role for the rational design of enzymes with novel and specific functionalities. In this study, a strategic combination of genetic and chemical engineering paves the way for systematic construction of biocatalysts by tuning the product spectrum of a levansucrase from Bacillus megaterium (Bm-LS), which typically produces small levan-like oligosaccharides. The implementation of site-directed mutagenesis followed by a tyrosine-specific modification enabled control of the product synthesis: depending on the position, the modification provoked either enrichment of short oligosaccharides (up to 800 % in some cases) or triggered the formation of high molecular weight polymer. The chemical modification can recover polymerization ability in variants with defective oligosaccharide binding motifs. Molecular dynamic (MD) simulations provided insights into the effect of modifying non-native tyrosine residues on product specificity. 相似文献

建立了高效液相色谱(HPLC)和电感耦合等离子体质谱(ICP-MS)联用测定多种生物样品中的无机汞和甲基汞的方法,并对比了提取生物样品中无机汞和甲基汞的不同前处理方法。实验使用5 mol/L的盐酸超声波提取样品中的无机汞和甲基汞。高效液相色谱流动相为含有0.06 mol/L醋酸氨,20μg/L B i,0.1%(V/V)2-巯基乙醇的5%(V/V)甲醇-水溶液,色谱柱为C18反相柱(5μm,3.9 mm×150 mm)。提取液在液相色谱中分离后,进入电感耦合等离子体质谱检测其中无机汞和甲基汞的浓度。测定了人发(GBW 07601),对虾(GBW 08572),鱼肉组织(IAEA MA-B-3/TM)和牛肝(GBW 080193)4种生物标准参考物,结果与标准参考物的标准值相符。无机汞和甲基汞检出限分别为0.3和0.2μg/L。 相似文献

本文通过局部展开算法研究了多孔介质中排列结构对非线性渗流规律的影响.计算结果表明:规则排列结构的渗流标度律和渗流方向相关,然而非规则排列结构的渗流标度律不依赖于渗流方向而只依赖于多孔介质的几何结构.伴随着排列结构的非规则程度增强,不同方向的渗流阻力变化规律的差异性减小.当流场输运区域较为复杂且不存在较宽直通道时,渗流阻力在较宽雷诺数范围内体现出较为明显的标度律特性.周期性强非规则排列结构中不同区域大小的模型却具有相似的渗流阻力的标度律特性,同时标度律特性与孔隙度的大小相关性不大.阻力非线性标度律由多孔介质结构的不同而取值为2至3之间. 相似文献

在分析传统的双面立式动平衡机的振动结构的基础上,设计了一种新的立式动平衡机的振动结构(即摆架),采用新摆架结构的动平衡机可以实现静不平衡和偶不平衡的有效分离。文中详细分析了新摆架结构的特点,并在ANSYS7.0平台上对摆架进行了有限元模态分析和谐响应分析。由于新的摆架结构采用多块簧板,在多个部位刚度匹配,且采用多个传感器安装于测量点处,这样新的动平衡机可以直接测量静不平衡和偶不平衡,且相互影响极小。实践证明:采用新摆架结构的动平衡机特别适合于飞行物体的惯性测量,且克服了传统的双面立式动平衡机长期以来平面分离不佳的缺点,具有很高的推广价值。 相似文献

A series of new long-lasting phosphor Gd2O2S:xEr,Ti are prepared by the conventional high-temperature solid-state method and their luminescent properties are systematically investigated in this paper. The characteristic afterglow emission of Er, which is due to the transition of 4F9/2→4I15/2 and 4S3/2→4I15/2, is reported for the first time. XRD, photoluminescence, long-lasting phosphorescence and decay curves are used to characterize the synthesized phosphors. A new homoisoflavonoid, (3R)-5,7-dihydroxy-6-methyl-3-(2′-hydroxy-4′-methoxybenzyl)-chroman-4-one (1), namely polygonatone H, in addition to fourteen known homoisoflavones (2–15) were isolated from the rhizome of Polygonatum Cyrtonema Hua. The structures were identified with the aid of 1D/2D NMR spectroscopic technologies. Compounds 2, 6, 8, 10, 11, 13, and 15 were isolated from P. Cyrtonema for the first time. Compound 1 showed cytotoxicities to human cancer cell lines with IC50 values to comparable those of cisplatin. 相似文献
