Biosynthetic human Growth Hormone (B-hGH) is a protein comprising 191 amino acids. The molecular weight is 22,125 and the isoelectric point is close to pH 5. Due to the ready availability of closely related analogues B-hGH was used as a model protein thus allowing for the demonstration and evaluation of the high resolution capability of high performance capillary electrophoresis (HPCE). The same apparatus was used throughout the experiments and an optimum signal-to-noise ratio was found at 200 nm. Linearity was observed between peak area, retention time and the hGH concentration or sample introduction time. Baseline separation of hGH, desamido hGH and didesamido hGH was obtained. Examples showing analysis with 1 million theoretical plates per meter, high speed separation, simultaneous analysis of multiple samples, sample stacking, hGH tryptic digest, and hGH lysate are reported. The use of electrophoretic velocities instead of apparent velocities for peak identification is illustrated.  相似文献

Cyclodextrins (CDs) are cyclic oligosaccharides that have found widespread application in numerous fields. CDs have revealed a number of various health benefits, making them potentially useful food supplements and nutraceuticals. In this study, the impact of α-, β-, and γ-CD at different concentrations (up to 8% of the flour weight) on the wheat dough and bread properties were investigated. The impact on dough properties was assessed by alveograph analysis, and it was found that especially β-CD affected the viscoelastic properties. This behavior correlates well with a direct interaction of the CDs with the proteins of the gluten network. The impact on bread volume and bread staling was also assessed. The bread volume was in general not significantly affected by the addition of up to 4% CD, except for 4% α-CD, which slightly increased the bread volume. Larger concentrations of CDs lead to decreasing bread volumes. Bread staling was investigated by texture analysis and low field nuclear magnetic resonance spectroscopy (LF-NMR) measurements, and no effect of the addition of CDs on the staling was observed. Up to 4% CD can, therefore, be added to wheat bread with only minor effects on the dough and bread properties.  相似文献

The x-ray diffraction intensities of Bragg reflections have been measured at room temperature for thulium selenide, samarium sulphide, samarium selenide and samarium telluride. On the basis of a common amplitude approximation, the Debye-Waller factor, the mean amplitude of vibration and the Debye temperature have been evaluated. The values of the Debye temperatures and mean amplitudes of vibration are 176±16°K, 0·185 ± 0·017 Å (TmSe), 155 ± 7°K, 0·244 ± 0·012 Å (SmS), 153 ± 14°K, 0·221 ± 0·020 Å (SmSe) and 151 ± 20°K, 0·204 ± 0·027 Å (SmTe).  相似文献

MRI measurements of water diffusion and blood perfusion are increasingly used for the evaluation of organ functionality and tissue viability (e.g., in tumors). While diffusion-weighted imaging is performed without contrast agents, measurement of blood perfusion is normally performed based on the administration of paramagnetic substances such as gadolinium diethylenetriamine pentaacetic acid (Gd-DTPA). Simultaneous measurements of these two parameters are often preferred. However, it may be argued that Gd-DTPA causes constriction of small blood vessels or alters hemodynamic parameters such as blood viscosity, thereby corrupting subsequent measurements of the apparent diffusion constant (ADC). The objective of the current study was to investigate the possible changes in the ADC in tumors following intravenous administration of 0.2 and 0.4 mmol/kg of Gd-DTPA in mice. The study was conducted with C3H mouse mammary carcinomas inoculated in the right foot of the animal subjects. The results were compared with findings in a sham group, demonstrating that Gd-DTPA had no significant impact on the ADC as measured in a 7-T animal system.  相似文献

Scanning electron microscopy (SEM) studies on the poultry red mite, Dermanyssus gallinae (DeGeer), showed that the tip of the foreleg tarsus of both sexes and early instar stages (larvae, protonymph and deutonymph) bears a cluster of 10 blunt peg-type sensillae. Diversity in size, shape and surface features of the wall of these sensillae characterize the sensilla complex (sensory field). Two short setae (1 and 2) with smooth walls lack detectable pores, whereas three medium-sized setae (4, 5 and 8) bear pores in the wall. Four medium or long setae (3, 6, 7 and 9) have longitudinal grooves or ribs in the wall, whilst one seta (10) possesses both pores and grooves in the wall. Based on the morphology, it is suggested that the porous setae could be olfactory receptors, and the grooved setae could be mechanoreceptors.  相似文献

In the present work, we describe a collection system for the off-line coupling of capillary isoelectric focusing (CIEF) with matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry. In this system, the capillary effluent is directly deposited in fractions onto the MALDI target via the use of a sheath liquid. The collected fractions are subsequently supplemented with matrix and further analysed by MALDI-TOF mass spectrometry for mass assignment. The experimental set-up includes a fiber optic based UV detector operating at 280 nm, which allows the study of the influence of the sheath liquid composition on the CIEF separation. The influence of the carrier ampholyte concentration on the protein MALDI spectra was also evaluated and the feasibility of the collection method was finally demonstrated with a mixture of four standard proteins.  相似文献

A capillary-based model modified for characterization of monolithic cryogels is presented with key parameters like the pore size distribution, the tortuosity and the skeleton thickness employed for describing the porous structure characteristics of a cryogel matrix. Laminar flow, liquid dispersion and mass transfer in each capillary are considered and the model is solved numerically by the finite difference method. As examples, two poly(hydroxyethyl methacrylate) (pHEMA) based cryogel beds have been prepared by radical cryo-copolymerization of monomers and used to test the model. The axial dispersion behaviors, the pressure drop vs. flow rate performance as well as the non-adsorption breakthrough curves of different proteins, i.e., lysozyme, bovine serum albumin (BSA) and concanavalin A (Con A), at various flow velocities in the cryogel beds are measured experimentally. The lumped parameters in the model are determined by matching the model prediction with the experimental data. The results showed that for a given cryogel column, by using the model based on the physical properties of the cryogel (i.e., diameter, length, porosity, and permeability) together with the protein breakthrough curves one can obtain a reasonable estimate and detailed characterization of the porous structure properties of cryogel matrix, particularly regarding the number of capillaries, the capillary tortuousness, the pore size distribution and the skeleton thickness. The model is also effective with regards to predicting the flow performance and the non-adsorption breakthrough profiles of proteins at different flow velocities. It is thus expected to be applicable for characterizing the properties of cryogels and predicting the chromatographic performance under a given set of operating conditions.  相似文献

An all-fiber-integrated linear chirped-pulse amplifier system generating microjoule pulse energies is reported. It is seeded by an all-fiber dissipative-soliton laser and incorporates a newly developed fiber stretcher, whose dispersive properties match that of the grating compressor. Pulse durations of 189 fs with energies of 2.2 μJ were achieved after compression. The average power was 9.8 W at the repetition rate of 4.5 MHz.  相似文献

位于藏东南的帕隆藏布流域,处在东喜马拉雅构造结的北缘,是新构造强烈隆升区,断层、褶皱等地质构造发育。在青藏高原隆升及气候变化等内外动力作用下,研究区内堆积体分布广泛,具有独特的工程地质特征并成为该区内地质灾害的主要物源。由于河流的下切及川藏公路的修建,形成了许多高陡的堆积体边坡,以冰碛边坡最为典型。由于其特殊的物源特征及形成过程,冰碛物具有粒度分布范围广、粗粒含量多等与其他沉积物不同的粒度特征。冰碛物粒度分布范围广的特征,导致随着研究尺度的不同,其结构特征也不相同,具有明显的尺度效应。冰碛土的力学强度一般强于其他堆积体,但受不同粒径颗粒的含量及分布、形成时代的影响,其力学特征也有所差异。作者对川藏公路沿线部分典型的剖面进行了量测,所量测的剖面虽然只是整个斜坡的一部分,与经验数据相比,仍表现出高陡的特征,而且很多边坡在既高且陡的形态下仍能保持稳定。本文以极限平衡理论为基础,采用多种方法对这类边坡的稳定性进行计算分析,求出边坡的安全系数。考虑各种地震烈度下边坡的稳定性,得出这些边坡的安全系数与地震加速度具有严格的指数相关关系。  相似文献
