The present study aimed describing the ovaries of the sugarcane spittlebug Mahanarva fimbriolata which are meroistic telotrophic with nurse cells and oocytes located in the tropharium. SEM revealed paired ovaries located dorsolaterally around the intestine, and oocytes exhibiting shapes ranging from round (less developed) to elliptic (more developed), suggesting a simultaneous, although, asynchronous development. Based on histological data we classified the oocytes in stages from I to V. Stage I oocytes exhibit follicular epithelium with cubic and/or prismatic cells, fine cytoplasmic granules. Stage II oocytes present intercellular spaces in the follicular epithelium due to the incorporation of yolk elements from the hemolymph. Small granules are present in the periphery of oocytes while larger granules are observed in the center. Stage III oocytes are larger and intercellular spaces in the follicular epithelium are evident, as well as the interface between follicular epithelium and oocyte. Yolk granules of different sizes are present in the cytoplasm. During this stage, chorion deposition initiates. Stage IV oocytes exhibit squamous follicular cells and larger intercellular spaces when compared to those observed in the previous stage. The oocyte cytoplasm present granular and viscous yolk, the latter is the result of the breakdown of granules. Stage V oocytes exhibit a follicular epithelium almost completely degenerated, smaller quantities of granular yolk and large amounts of viscous yolk. Based on our findings we established the sequence of yolk deposition in M. fimbriolata oocyte as follows: proteins and lipids, which are first produced by endogenous processes in stages I and II oocytes. Exogenous incorporation begins in stage III. In stages I and II oocytes, lipids are also produced by follicular epithelial cells. The third element to be deposited is polysaccharides, mainly found as complexes. Therefore, the yolk present in the oocytes of this species consists of glycolipoproteins. Molecular weights of proteins present in M. fimbriolata oocytes ranged from 10 to 92 KDa, differently from vitellogenin, the most common protein present in insect oocytes, weighing approximately 180 KDa.  相似文献

Mutagenic cyclobutane pyrimidine dimers (CPDs) can be induced in DNA through either direct excitation or photosensitized triplet–triplet energy transfer (TTET). In the latter pathway, thymines are expected to receive the excitation energy from the photosensitizer and react with adjacent pyrimidines. By using state‐of‐the art analytical tools, we provide herein additional information on the formation of cytosine‐containing CPDs. We thus determined the yield of all possible CPDs upon TTET in a series of natural DNAs with various base compositions. We show that the distribution of CPDs cannot be explained only by excitation of individual thymines. We propose that the mechanism for TTET involves at least dinucleotides as the minimal targets. The observation of the formation of cytosine–cytosine CPDs also suggests that additional pathways are involved in this photosensitized reaction.  相似文献

The aim of this study was to evaluate the production and properties of novel extracellular polymeric substances (EPS) from glucose (EPS 1) and crude glycerol (CG) from castor beans oil (EPS 2). Cupriavidus necator IPT 027 cultivated with glucose and CG (35 °C, 150 rpm, 72 hr, pH 7, shaker) produced approximately 1.60 ± 0.01 and 2.83 ± 0.02 g l?1 EPS, respectively, with high‐molecular weight (3.89 × 105 and 1.89 × 107 Da) and constituted of different functional groups such as uronic acid, monosaccharides (glucose, mannose, arabinose, and fucose) and primary amine group. The composition of the fermentable substrate influenced the melting temperature (221.11 and 230.18 °C), crystallinity (34.36% and 37.11%), degradation temperature (255.06 and 296.62 °C) and morphology. EPS showed pseudoplastic non‐Newtonian fluid behavior in the aqueous solutions, presenting potential applicability biotechnological and industrial mainly in food industry as emulsifiers and biosurfactants. This is the first study on the production and characterization of EPS obtained by C. necator IPT 027 in culture with glucose and CG. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

The present study reports cytochemistry data about salivary glands of females (unfed, engorged, and at day three post-engorgement) and males (unfed, at day seven post-attachment, and at days three and seven post-detachment from the host) of the tick Rhipicephalus sanguineus. The results revealed nuclear changes in engorged females and at day three post-engorgement, and in males in all stages (except unfed). These changes were more prominent in females. Cytoplasmic changes were also observed in cells of all acini of males and females. In types II and III acini of engorged females, nuclear changes were observed in the shape (irregular, with blebs, fragmenting or fragmented), size (enlarged or reduced), and arrangement and condensation level of chromatin (marginal or as blebs). Changes were also detected in nucleoli, regarding their shape (fragmenting or fragmented), size (enlarged), and location (central, marginal or as blebs). Some nucleoli were also compacted or disorganized. In females at day three pos-engorgement, all acini exhibited similar changes to those observed in engorged females. RNA staining was stronger in cells of engorged females than those at day three post-engorgement. In males at day seven post-attachment, cells of types II, III, IV acini presented changes in the size of the nucleus and condensation level of chromatin similar to those of females. The shape of the nucleus was round, irregular or undergoing fragmentation, and the chromatin was located at the margin or throughout the nucleus. The changes in the nucleolus were similar to those of females, regarding size and organization, although round-shaped and in the central location. In males at day three post-detachment, cells of all acini exhibited nuclear changes similar to those of males at day seven post-attachment, in addition to the fragmentation of the nucleolus. At day seven post-detachment, changes were detected in all acini similar to the observed in males at day seven post-attachment. Regarding cytoplasmic RNA, staining was prominent in males at day seven post-attachment and weak in those at day seven post-detachment from the host. In females as well as males, different RNA staining patterns in the cytoplasm and nuclear changes characterized apoptotic cell death.  相似文献

Spittlebugs are insects that suck sap from plants and regurgitate saliva containing toxic enzymes into the leaves. As a consequence, the conductive channels are blocked resulting in dry leaves, thus giving a burned aspect to the plantation. This work performed ultrastructural analyses of the salivary glands of the sugarcane spittlebug Mahanarva fimbriolata, since these organs produce the enzymes that are injected into the plants, thus being responsible for the economic losses in the production of sugarcane. Three kinds of secretory cells are found in principal gland, forming the lobules I-IV. The main differences among these cells relate to size, morphology and electron density of the secretory vesicles. The accessory glands contain different secretory vesicles to those in the principal gland. Muscular cells are found around the entire gland. The different secretory vesicles found in both principal and accessory glands indicate that the gland produces different substances or that the secretion in the interior of cells passes through a maturation process.  相似文献

The mandibular glands of Hymenoptera are structures associated with the mandibles and constitute part of the salivary glands system. Histological studies in workers of Atta sexdens rubropilosa revealed that this gland contains two portions: a secretory and a storage portion or reservoir. Both portions are connected by means of canaliculi. The object of the present work was the study of the ultratructure of the mandibular glands of minima, media and soldier ant of A. s. rubropilosa by TEM techniques. The glands, in the three castes studied, possess a reservoir, constituted by a simple pavementous epithelium surrounded by the cuticular intima and the secretory portion is constituted by cells of rounded shape. The secretory cells, mainly of minima and soldier, were rich in smooth endoplasmic reticulum. The media worker and soldier presented a large number of mitochondria, of varying shape. Well-developed Golgi complexes were also present in the soldiers. The secretory cells in minima, media and soldier were provided with collecting intracellular canaliculi, which were linked to the reservoir through the extracellular portion. The cytoplasm of the canaliculi-forming cell was poor in organelles. In the individuals of the three castes of A. s. rubropilosa, the presence of lipid secretion granules suggested, beyond the other functions, also a possible pheromonal action. The different roles executed by the different insect castes are directly dependent on the glandular products and, consequently, on the secretory cellular characteristics.  相似文献
