Slow-channel congenital myasthenic syndromes (SCCMSs) are rare genetic diseases caused by mutations in muscle nicotinic acetylcholine receptor (nAChR) subunits. Most of the known SCCMS-associated mutations localize at the transmembrane region near the ion pore. Only two SCCMS point mutations are at the extracellular domains near the acetylcholine binding site, α1(G153S) being one of them. In this work, a combination of molecular dynamics, targeted mutagenesis, fluorescent Ca2+ imaging and patch-clamp electrophysiology has been applied to G153S mutant muscle nAChR to investigate the role of hydrogen bonds formed by Ser 153 with C-loop residues near the acetylcholine-binding site. Introduction of L199T mutation to the C-loop in the vicinity of Ser 153 changed hydrogen bonds distribution, decreased acetylcholine potency (EC50 2607 vs. 146 nM) of the double mutant and decay kinetics of acetylcholine-evoked cytoplasmic Ca2+ rise (τ 14.2 ± 0.3 vs. 34.0 ± 0.4 s). These results shed light on molecular mechanisms of nAChR activation-desensitization and on the involvement of such mechanisms in channelopathy genesis. 相似文献
Compositions based on chitosan/β-glycerophosphate hydrogels with highly porous polylactide granules can be used to obtain moldable bone graft materials that have osteoinductive and osteoconductive properties. To eliminate the influence of such characteristics as chain length, degree of purification, and molecular weight on a designed material, the one-stock chitosan sample was reacetylated to degrees of deacetylation (DD%) of 19.5, 39, 49, 55, and 56. A study of the chitosan/β-glycerophosphate hydrogel with chitosan of a reduced DD% showed that a low degree of deacetylation increased the MSCs (multipotent stromal cells) viability rate in vitro and reduced the leukocyte infiltration in subcutaneous implantation to Wistar rats in vivo. The addition of 12 wt% polylactide granules resulted in optimal composite mechanical and moldable properties, and increased the modulus of elasticity of the hydrogel-based material by approximately 100 times. Excessive filling of the material with PLA (polylactide) granules (more than 20%) led to material destruction at a ~10% strain. Osteoinductive and osteoconductive properties of the chitosan hydrogel-based material with reacetylated chitosan (39 DD%) and highly porous polylactide granules impregnated with BMP-2 (bone morphogenetic protein-2) have been demonstrated in models of orthotopic and ectopic bone formation. When implanted into a critical-size calvarial defect in rats, the optimal concentration of BMP-2 was 10 μg/mL: bone tissue areas filled the entire material’s thickness. Implantation of the material with 50 μg/mL BMP-2 was accompanied with excessive growth of bone tissue and material displacement beyond the defect. Significant osteoinductive and osteoconductive properties of the material with 10 μg/mL of BMP-2 were also shown in subcutaneous implantation. 相似文献
We announce a new result, contained in [10–12], concerning rough solutions to Einstein vacuum equations expressed relative to wave coordinates. Our methods blend paradifferential techniques with a geometric approach to Strichartz type inequlities, which takes advantage of the specific structure of the Einstein equations. The UV excited and persistent luminescence properties as well as thermoluminescence (TL) of Eu2+ doped strontium aluminates, SrAl2O4:Eu2+ were studied at different temperatures. Two luminescence bands peaking at 445 and 520 nm were observed at 20 K but only the latter at 295 K. Both Sr-sites in the lattice are thus occupied by Eu2+ but at room temperature efficient energy transfer occurs between the two sites. The UV excited and persistent luminescence spectra were similar at 295 K but the excitation spectra were different. Thus the luminescent centre is the same in both phenomena but excitation processes are different. Two TL peaks were observed between 50 and 250 °C in the glow curve. Multiple traps were, however, observed by preheating and initial rise methods. With longer delay times only the high temperature TL peak was observed. The persistent luminescence is mainly due to slow fading of the low temperature TL peak but the step-wise feeding process from high temperature traps is also probable. 相似文献

We presented exact analytical formulae and numerical calculation of diffusitivity curves with different law for a local pore behavior and have obtained critical exponent, different from conductivity. The connectivity percolation theory was built only on the conductivity, (the diffusion critical exponent was supposed to be equal to the conductivity exponent) and therefore sees only one side of problem-the scalar side. In many topological problems involving mechanical properties and fluid flow the connectivity scalar percolation geometry does not enough to apply.
One of the most useful aspects of percolation is that many very complicated systems have the same behavior with the same critical exponents. Universality of vector percolation is shown in the coincidence between the experimental measured relative hydraulic permeability of fluid and gas flow through unconsolidated sand and effective conductivity and diffusitivity curves of the bond–site percolation models. Comparisons of our calculation results to natural matches are quite good. We have argued that experimental data may be interpreted as a variant of pure vector percolation and to belong to the same universality class. 相似文献
We study von Karman evolution equations with non-linear dissipation and with partially clamped and partially free boundary conditions. Two distinctive mechanisms of dissipation are considered: (i) internal dissipation generated by non-linear operator, and (ii) boundary dissipation generated by shear forces friction acting on a free part of the boundary. The main emphasis is given to the effects of boundary dissipation. Under suitable hypotheses we prove existence of a compact global attractor and finiteness of its fractal dimension. We also show that any solution is stabilized to an equilibrium and estimate the rate of the convergence which, in turn, depends on the behaviour at the origin of the functions describing the dissipation. 相似文献
