Procedures for the determination of vinblastine (VBL), 4-O-deacetylvinblastine (DVBL) and 4-O-deacetylvinblastine-3-oic acid (DVBLA) in biological samples using high-performance liquid chromatography (HPLC) combined with selective sample clean-up are presented. VBL and DVBL were determined in plasma and urine using ion-exchange normal-phase HPLC with fluorescence detection. The limit of detection was 1 microgram/l for both compounds using a 500-microliter sample. Successful chromatographic analyses of DVBLA were achieved by using a glass column packed with 5-microns Hypersil ODS and acetonitrile-0.05 M phosphate buffer (pH 2.7) (23:77, v/v). Positive identification was supported by the use of diode-array detection. The limit of detection (at 270 nm) was 10 micrograms/l using 1-ml samples.  相似文献

Frequent in vivo sampling of blood proteins is often stressful, making it difficult to obtain more than a few samples. As a result, only limited time-profiles can be made. We have developed an ultrafiltration collection device (UCD) for continuous sampling. The UCD consists of a hollow fiber, a coil and a flow creator. Hollow fiber membranes are often hydrophobic and this can result in adsorption of protein and/or peptides, leading to clogged membranes. Adsorption was tested with a hydrophobic and hydrophilic peptide and two biocompatible hollow fibers made from different materials. The hollow fiber made from poly(ethylene) coated with ethylenevinyl alcohol gave near 100% recovery for both peptides. This was in contrast to the poly(sulfone) hollow fiber when sampling the hydrophobic peptide. Filling the coil with various peptide concentrations gave good recovery and insignificant diffusion even after storage for 6 d at 37 degrees C. Continuous pulse-free sampling was tested by vacuum. An average flow rate of 423 +/- 50 nl min(-1) over a period of 4 d was created using S-Monovette. The flow rate gradually declined during this period by <5% every consecutive day. In addition, we also examined a complex sample-serum in the poly(ethylene) hollow fiber. Serum and ultrafiltrate were spotted onto a protein chip and analyzed by surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS). Six proteins out of 64 were found to be significantly different between serum and the ultrafiltrate (p < 0.05). The UCD has the potential to be used for in vivo real-time monitoring.  相似文献

This review paper covers the low temperature wet growth of nano-engineered particles of ZnO-based mixed metal oxides, their growth mechanism, and characterization using X-ray diffraction, SEM, TEM and IR, UV–visible, and XPS spectral techniques. Main focus of this article is centered on low temperature semi-wet methods of synthesis that are suitable for large scale production of zinc oxide-based systems mixed with iron oxide, copper oxide, nickel oxide and cobalt oxide. These mixed metal oxides have broad industrial applications as catalyst, semiconductors, adsorbents, superconductors, electro-ceramics, and antifungal agents in addition to extensive applications in medicines. This paper discusses the low-cost and environment friendly synthesis of these mixed metal oxides, measurement of properties and applicability of these materials systems.  相似文献

Salts can damage building materials by chemical reactions or crystallization, which is a serious threat to cultural heritage. In order to develop better conservation techniques, more knowledge of the crystallization processes is needed. In a porous material, the size of a salt crystal is limited by the sizes of the pores. It has been predicted that as a consequence, the solubility of a salt increases with decreasing pore size. This increase seems to be related to an increase of the stress generated by a crystal on the pore wall. It has been suggested that the resulting stress could become high enough to induce failure. We have studied the crystallization of salts in porous materials with well-defined pore sizes. Samples were saturated at 40 degrees C with saturated Na2SO4 and Na2CO3 solutions. Next we have cooled the samples to 0 degrees C and waited for nucleation. After nucleation occurred, the solubility in the porous material was measured with nuclear magnetic resonance (NMR) as a function of the temperature. The measurements on Na2CO3 indeed show an increase in solubility with a decrease in pore size. For Na2SO4, we did not observe a pore size-dependent solubility. However, we have to remark that these results show a metastable crystal phase. The results can be used to calculate the actual pressure exerted by the crystals onto the pore wall.  相似文献

Crystallization pressure of salt in porous materials is one of the mechanisms that may induce serious damage, for example, weathering of buildings and monuments of cultural heritage. Since this pressure also causes the solubility of the salt inside a porous material to differ from the bulk solubility, it can be assessed experimentally by measuring the solubility inside the pores. We show that this is possible by NMR, and study Na(2)CO(3) and Na(2)SO(4) in a series of model porous materials. Using the solubility data the crystal-liquid surface energies are estimated as gamma = 0.09 N/m for Na(2)CO(3) . 10H(2)O and gamma = 0.06 N/m for Na(2)SO(4) . 10H(2)O. For pore sizes below about 30 nm, the resulting pressure exceeds the tensile strength of typical building materials (3 MPa). No pressure is induced by the metastable Na(2)SO(4) . 7H(2)O, which suggests for this crystal a value of gamma close to zero.  相似文献

The concern regarding the effect of chemicals on the environment has increased considerably in recent years. Nowadays, technological developments in the coating industry are largely influenced by environmental issues and subsequent legislation. One of these issues is the tendency to replace cobalt as a catalyst with more environmentally friendly alternatives, because studies have indicated possible carcinogenicity. Not much knowledge is available on the effects of catalysts (driers) on the in-depth drying of alkyd coatings. Therefore we have studied the effect of cobalt as a primary drier combined with Ca and Zr as secondary driers on the in-depth curing of high solid solvent-borne alkyds. The profiling of the curing of alkyd coatings is performed with a new high-spatial-resolution NMR setup. In this study, two effects observed in the solvent-borne alkyd coatings are investigated. One is that when Ca and Zr are added as secondary driers the speed of the observed cross-linking front increases. Second, in the deeper un-cross-linked region below the front, the signal of the NMR profiles was found to decrease proportional to . This could be explained by the presence of slowly reacting species that diffuse into the deeper uncured region of the coating, after which they cross-link. The model describing the effect of these reactive species also indicates that the signal decrease is inversely proportional to coating thickness L, which was confirmed by additional measurements.  相似文献

High spatial resolution NMR imaging techniques have been developed recently to measure the spatial inhomogeneity of a polymer coating film. However, the substrates of the polymer coatings for such experiments are generally required to be non-metallic, because metals can interact with static magnetic fields B(0) and RF fields B(1) giving rise to artifacts in NMR images. In this paper we present a systematic study on the effects of metallic substrates on 1D profiles obtained by high resolution NMR imaging. The off-resonance effect is discussed in detail in terms of the excitation profile of the RF pulses. We quantitatively show how the NMR signal intensities change with frequency offset at different RF pulse lengths. The complete NMR profiles were simulated using a Finite Element Analysis method by fully considering the inhomogeneities in both B(1) and B(0). The excellent agreement between the calculated and measured NMR profiles on both metallic and non-metallic substrates indicates that the experimental NMR profiles can be reproduced very well by numerical simulations. The metallic substrates can disturb the RF field of the coil by eddy current effect and therefore change the NMR profiles. To quantitatively interpret the NMR profile of a polymer layer on a metallic substrate, the profile has to be divided by the profile of a reference on the same metallic substrate located at the same distance from the coil.  相似文献

Background  

Short-term habituation of the startle response represents an elementary form of learning in mammals. The underlying mechanism is located within the primary startle pathway, presumably at sensory synapses on giant neurons in the caudal pontine reticular nucleus (PnC). Short trains of action potentials in sensory afferent fibers induce depression of synaptic responses in PnC giant neurons, a phenomenon that has been proposed to be the cellular correlate for short-term habituation. We address here the question whether both this synaptic depression and the short-term habituation of the startle response are localized at the presynaptic terminals of sensory afferents. If this is confirmed, it would imply that these processes take place prior to multimodal signal integration, rather than occurring at postsynaptic sites on PnC giant neurons that directly drive motor neurons.  相似文献

The influence of temperature gradients on the drying of water-saturated porous networks has been studied. We have focussed on the influence of the temperature on the drying process via the equilibrium vapor density rhoe, because this is the most sensitive parameter with respect to variations of the temperature T. We have used a 2D model which accounts for both capillary and buoyancy forces. Invasion events by air or water are handled by standard rules of invasion percolation in a gradient (IPG). Vapor fluxes are calculated by solving a discretized version of the Laplace equation. In the model the temperature T varies linearly from the open side T0 to the closed side TL. The temperature gradients strongly influence the cluster evolution during the process, because they facilitate vapor transport through wet regions. When T0TL, the front movement is enhanced and the air ingress in the wet region behind the front is inhibited. The behavior of 3D systems differs from that of 2D systems, because the point where air percolates the system and the point where the water network breaks up in isolated clusters do not coincide. Before the latter fragmentation point the temperature will mainly influence the drying rates. After this point also the water distribution becomes sensitive to the temperature profile.  相似文献
