The method used at LGC for analysis of “total” 19-norandrosterone (19-norandrosterone glucuronide plus “free” 19-norandrosterone)
in urine for the Comité Consultatif pour la Quantité de Matière Pilot Study (CCQM-P68) is described. The analytical method
used was a modified version of the method developed at the National Measurement Institute of Australia, which used a hydrolysis
and derivatisation procedure first described by the German Sports University. This method is routinely used by World Anti-Doping
Agency-accredited laboratories for sports drug testing. The main modifications made to the method were the use of 19-norandrosterone
glucuronide as a calibration standard and 19-norandrosterone glucuronide-d4 as an isotopically labelled internal standard,
and the use of a bench-top quadrupole gas chromatograph–mass spectrometer. The results produced by LGC (2.14 ± 0.15 ng g−1 expanded uncertainty, coverage factor k = 2) were in excellent agreement with those from other participating national metrology institutes and thus further validates
the exact-matching isotope-dilution mass spectrometric procedures used at LGC for a wide range of reference measurement applications,
including measurement of ng g−1 levels of steroids in a biological matrix. 相似文献

The design and development of a novel extractive electrospray ionisation (EESI) device for on-line reaction monitoring is described. The EESI apparatus uses a secondary, grounded nebuliser to produce an analyte aerosol and a Venturi pump is then used to transfer a sample of the aerosol to an electrospray source where it is ionised. The EESI apparatus was then tested with a variety of small, organic molecules to assess sensitivity, linearity and dynamic range. The performance of the technique will depend on the mass spectrometer used for the experiments; in the configurations used here it has a usable dynamic range of around 3.5 orders of magnitude with a linear range of around 2.5 orders of magnitude and is capable of analysing species present down to low μg/mL with signal-to-noise ratio greater than 2.5. The use of EESI for reaction monitoring was validated using a series of mock reaction mixtures and then used to monitor the base hydrolysis of ethyl salicylate to salicylic acid. 相似文献

Product ion spectra produced by collision-induced dissociation (CID) in tandem mass spectrometry can yield important structural information on organic compounds which can aid in their identification. However, differences in experimental conditions may have a strong effect on the degree of product ion formation and therefore on the features observed in product ion spectra. For this reason, a common approach for library building is the acquisition of several spectra, typically between 5 and 10, each at a different collision energy level. In this study, the use of an alternative approach was investigated, where a tuning point protocol was applied to tune the instruments in an attempt to standardise CID conditions prior to data acquisition. With this approach, the acquisition of a single mass spectrum was sufficient. The stability of the tuning point was investigated and the choice of a commercially available search package to assess spectral comparability was discussed. Finally, the product ion spectra of 33 compounds were acquired on twelve tandem-in-space instruments, including nine triple quadrupoles, one hybrid triple quadrupole linear ion trap and two quadrupole time-of-flight mass spectrometers, resulting in 2178 spectral comparisons being carried out. The results from the spectral comparisons suggest that the use of a tuning point enables the standardisation of the experimental conditions that affect the degree of product ion formation. Indeed, 84.5% of the comparisons demonstrated a good degree of spectral agreement with match scores greater than 700, which we believe is the minimum score for a tentative library match. 相似文献

Product ion spectra produced by collision-induced dissociation (CID) in tandem mass spectrometry experiments can differ markedly between instruments. There have been a number of attempts to standardise the production of product ion spectra; however, a consensus on the most appropriate approach to the reproducible production of spectra has yet to be reached. We have previously reported the comparison of product ion spectra on a number of different types of instruments - a triple quadrupole, two ion traps and a Fourier transform ion cyclotron resonance mass spectrometer (Bristow AWT, Webb KS, Lubben AT, Halket JM. Rapid Commun. Mass Spectrom. 2004; 18: 1). The study showed that a high degree of reproducibility was achievable. The goal of this study was to improve the comparability and reproducibility of CID product ion mass spectra produced in different laboratories and using different instruments. This was carried out experimentally by defining a spectral calibration point on each mass spectrometer for product ion formation. The long-term goal is the development of a universal (instrument independent) product ion mass spectral library for the identification of unknowns.The spectra of 48 compounds have been recorded on eleven mass spectrometers: six ion traps, two triple quadrupoles, a hybrid triple quadrupole, and two quadrupole time-of-flight instruments. Initially, 4371 spectral comparisons were carried out using the data from eleven instruments and the degree of reproducibility was evaluated. A blind trial has also been carried out to assess the reproducibility of spectra obtained during LC/MS/MS.The results suggest a degree of reproducibility across all instrument types using the tuning point technique. The reproducibility of the product ion spectra is increased when comparing the tandem in time type instruments and the tandem in space instruments as two separate groups. This may allow the production of a more limited, yet useful, screening library for LC/MS/MS identification using instruments of the same type from different manufacturers. 相似文献

This work investigates for the first time the potential of mixed-mode (anion-exchange with reversed-phase) high performance liquid chromatography coupled to inductively coupled plasma mass spectrometry (ICP-MS) for the simultaneous retention and selective separation of a range of inorganic and organically-bound selenium (Se) species. Baseline separation and detection of selenocystine (SeCys2), Se-methyl-selenocysteine (SeMC), selenomethionine (SeMet), methylseleninic acid (MSA), selenite, γ-glutamyl-methyl-selenocysteine (γ-glutamyl-SeMC), and selenate in a Se standard mixture by mixed-mode HPLC-ICP-MS was achieved by switching between two citrate mobile phases of different pH and ionic strength within a single chromatographic run of 20 min. Limits of detection obtained for these Se species ranged from 80 ng kg?1 (for SeMC) to 123 ng kg?1 (for selenate). Using this approach as developed for selenium speciation, an adequate separation of inorganic and organic As compounds was also achieved. These include arsenite, arsenate, arsenobetaine (AsB) and dimethylarsenic acid (DMA), which may coexist with Se species in biological samples. Application of the newly proposed methodology to the investigation of the elemental species distribution in watercress (used as the model sample) after enzymatic hydrolysis or leaching in water by accelerated solvent extraction (ASE) was addressed. Only SeMet, SeMC and selenate could be tentatively identified in watercress extracts by mixed-mode HPLC-ICP-MS and retention time matching with standards. Recoveries (n = 3) of these Se species from samples spiked with standards averaged 102% (for SeMC), 94.9% (for SeMet) and 98.3% (for selenate). Verification of the presence of SeMet and SeMC in an enzymatic watercress extract was achieved by on-line HPLC-ESI MS/MS in selected reaction monitoring (SRM) mode. 相似文献
