Unsteady and chaotic characteristics of natural convection field in vertical slots at large Prandtl number

In this study we examined the unsteady and chaotic characteristics of the natural convection field in vertical slots at large Prandtl number. In the computation, temperature dependence of viscosity was partially taken into consideration. As a result we discovered that the spatial region where a significant temperature fluctuation is detected corresponds to the spatial region where the secondary cells exist. The largest Lyapunov exponents calculated from the temperature on the center line have a local maximum at the lower region of the enclosure when the viscosity change was considered. These characteristics agree well with the experimental ones that have been observed by authors (Ishida, Kure and Kimoto 2001).     

Nonlinear Cauchy problems with small analytic data

We study the lifespan of solutions to fully nonlinear Cauchy problems with small real- or complex-analytic data. Our proofs are based on the method of majorants and the fixed point theorem for a contraction mapping.

     

Clinical usefulness of a trypsin radioimmunoassay kit

From the clinical use of RIA-gnost trypsin kit, the following results were obtained. 1. Standard curve showed a steep and good curve was shown. 2. Incubation: The condition for the first incubation was set at the room temperature for 10-24 hours and that for the second incubation at the room temperature for 3-5 hours. With these settings, satisfactory results were obtained. 3. Reproducibility and recovery: The C.V. of the reproducibility and the recovery were considered superior, and the values were below 10% and +/- 3%, respectively. 4. Correlation between trypsin and serum elestase-1: An excellent positive correlation (coefficient of correlation r = 0.889) was shown. 5. Serum trypsin concentration of normal and pancreatic diseases: The normal range was from 100 to 500 ng/ml. Acute pancreatitis rose obviously. Diabetes mellitus and chronic pancreatitis was below 500 ng/ml and the pancreatic cancer showed a tendency to scatter in the range of 50-1,250 ng/ml. The above results indicated that serum trypsin can be easily measured with high precision by using this method. Thus the method is considered useful for the diagnosis of pancreatic diseases.     

Clinical evaluation of immunoglobulin-E radioimmunoassay kit

An IgE RIA kit (Sandwich method; Dainabott), is used to obtain the following results. (1) Standard curve: Since the range of reproduction rate show 3.16-7.07% (C.V.), the curve become steep. (2) Incubations under controlled situation: Both of the incubations are controlled at 15-30 degrees C for 2 h. (3) Reproducibility test: Coefficients of variation (C.V.) of intra-assay and inter-assay variation are 2.32-3.94% and 2.92-3.92% respectively. (4) Recovery test: A result of the recovery test range between 100.1-101.7%. (5) Dilution test: Multiple dilution effects are observed. (6) Average counts of the serum IgE for the controlled and diseased groups: The average counts of the serum IgE for the controlled group, atopic diseased group, allergic rhinitis group and allergic bronchial asthma are 144.9 +/- 183.2 IU/ml, 1,099.0 +/- 2,782.4 IU/ml, 1,150.9 +/- 2,063.3 IU/ml and 600.7 +/- 686.4 IU/ml respectively. The value of the diseased groups have tendency to show higher averages than the controlled group. Since the controlled and diseased groups show wide distributions of the serum IgE level, there is no significant difference of two variations. However the diseased groups have tendency to show higher ratio of the serum IgE level in blood than the controlled groups. These basic researches are quite meaningful, because they are able to apply for a supplemental diagnosis of the atopic and parasitic disease.     

Selective foam separation of binary protein solution by SDS complexation method

A fundamental study about the selective foam separation of protein mixture was carried out. A solution containing two proteins, ovalbumin (OA) and lysozyme (LZ), and an anionic surfactant, sodium dodecyl sulfate (SDS), was adjusted to pH 6.0, which referred to an intermediate state between the isoelectric points of the proteins. The solution was processed by continuous foam separation. The results showed that a proper addition of SDS greatly improved the selective recovery of LZ to OA. The experimental data were well explained by a simple model that most of cationic protein molecules (LZ) are associated with SDS and the adsorption of all the species including LZ-SDS complexes are subjected to Langmuir adsorption isotherm. The results also showed that one of the Langmuir parameters, which means a kind of lyophillic property of adsorbed material, of LZ-SDS complexes was extremely large as compared with that of primary protein.

自七十年代以来,已从海绵中发现了许多结构独特的、具有强烈生理活性的新化合物,有些已发展成为特效的药物。我们在研究中国南海海绵生物活性成分的过程中,从南海海绵Seletta teuuis Lindgren中分离出24-亚甲基-27-甲基胆甾醇(1)。虽然这个化合物已由De Luca等于1972年从海绵(Aplysina aerophoba)中发现了它。次     

Zooxanthellamide Cs: vasoconstrictive polyhydroxylated macrolides with the largest lactone ring size from a marine dinoflagellate of Symbiodinium sp

Zooxanthellamide Cs (ZAD-Cs), C(128)H(220)N(2)O(53)S(2) (ca. 2.7 kDa), was obtained from a cultured marine dinoflagellate of the genus Symbiodinium as an inseparable isomeric mixture of polyhydroxylated 61- to 66-membered macrolides. The chemical structures of the components were clarified by detailed 2D NMR analysis to be the macrolactonized analogues of zooxanthellamide A (ZAD-A), which had been previously isolated from the same microalgae. Chemical lability of ZAD-Cs suggests that ZAD-A is an artifact derived from ZAD-Cs during the isolation steps. Three of the components possess the largest (63-, 64-, and 66-membered) ring sizes found to date among the natural macrolides. ZAD-Cs exhibited higher vasoconstrictive activity than that of the zooxanthellatoxins, the first vasoconstrictive macrolides from Symbiodinium sp. The structure-activity relationship suggests that the huge macrolactone structure is important for biological activity. The relationship between the structures of the polyol metabolites and the phylogenetic systematics of Symbiodinium sp. is also discussed.     

Thermally-induced radical rearrangement of 2-(2-thienylmethoxy)-and 2-(2-benzo[b]thienylmethoxy)tropones

Upon heating at ca. 160°, 2-(2-thienylmethoxy)tropone afforded 3- and 5(2-thienylmethyl)tropolones. Formation of 3,5-bis(2-thienylmethyl)tropolone and unsubstituted tropolone indicated a radical chain mechanism for the thermolysis.     

Cloning and characterization of an active fragment of luciferase from a luminescent marine alga, Pyrocystis lunula

Two marine dinoflagellates, Lingulodinium polyedrum and Pyrocystis lunula, emit light in a reaction involving the enzymatic oxidation of its tetrapyrrole luciferin by molecular oxygen. The characteristic properties of P. lunula luciferase have not been clarified, whereas L. polyedrum luciferase, which has three active domains, has been characterized. A cloned partial cDNA of the P. lunula luciferase encodes an active fragment corresponding to part of domain 2 and all of domain 3 of L. polyedrum luciferase. The homology of the amino acid sequence between the two luciferases in domain 3 is about 84.3%. A recombinant His-tagged luciferase fragment containing domain 3 (Mr = 46 kDa) catalyzed the light-emitting oxidation of luciferin (lambdamax = 474 nm). This protein was purified by a single affinity-chromatography procedure. The pH-activity profile and the bioluminescence spectrum of the recombinant enzyme having a third domain are almost identical to those of an extract from P. lunula cultured in vitro. The recombinant enzyme is active at pH 8.0, although the recombinant enzyme derived from the second domain of L. polyedrum luciferase is inactive at pH 8.0. Substitution of Glu-201 by histidine in the third domain of P. lunula luciferase showed a decrease of activity above pH 7.0, suggesting that histidine residues could be responsible for pH-sensitivity in dinoflagellate luciferase.
