Avian influenza viruses preferentially bind to sialic acid alpha-2,3-galactose (N-SA-α-2,3-Gal) receptors on epithelial cells. Herein, we describe a procedure we have developed for isolation of N-SA-α-2,3-Gal receptors from porcine trachea using magnetic nanoparticles (NPs) coated with chitosan (NP-Ch) and functionalized with Maackia amurensis lectin (NP-lectin). Magnetic nanoparticles were coated with chitosan in a one-step co-precipitation, and then M. amurensis lectin was immobilized covalently using glutaraldehyde. Lectin coated nanoparticles were incubated with sialic acid enriched fraction of tracheal homogenate, and N-SA-α-2,3-Gal receptor was extracted under magnetic field in two cycles. The presence of 66.4 kDa protein was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The interaction of immobilized receptor (NP-Ch-R) with M. amurensis lectin (NP-Ch-R-L) was demonstrated by Fourier transform infrared spectrometry (FTIR) and thermogravimetric analysis (TGA).  相似文献

The purpose of this study was to evaluate the effect of pre-heating resin composite photo-cured with light-curing units (LCU) by FT-IR. Twenty specimens were made in a metallic mold (4 mm diameter × 2 mm thick) from composite resin—Tetric Ceram® (Ivoclar/Vivadent) at room temperature (25°C) and pre-heated to 37, 54, and 60°C. The specimens were cured with halogen curing light (QTH) and light emitted by diodes (LED) during 40 s. Then, the specimens were pulverized, pressed with KBr and analyzed with FT-IR. The data were submitted to statistical analysis of variance and Kruskal-Wallis test. Study data showed no statistically significant difference to the degree of conversion for the different light curing units (QTH and LED) (p > 0.05). With the increase of temperature there was significant increase in the degree of conversion (p < 0.05). In this study were not found evidence that the light curing unit and temperature influenced the degree of conversion.  相似文献

The indirect adhesive procedures constitute recently a substantial portion of contemporary esthetic restorative treatments. The resin cements have been used to bond tooth substrate and restorative materials. Due to recently introduction of the self-bonding resin luting cement based on a new monomer, filler and initiation technology has become important to study the degree of conversion of these new materials. In the present work the polymerization reaction and the filler content of dual-cured dental resin cements were studied by means of infra-red spectroscopy (FT-IR) and thermogravimetry (TG). Twenty specimens were made in a metallic mold (8 mm diameter × 1 mm thick) from each of 2 cements, Panavia® F2.0 (Kuraray) and RelyX? Unicem Applicap (3M/ESPE). Each specimen was cured with blue LED with power density of 500 mW/cm2 for 30 s. Immediately after curing, 24 and 48 h, and 7 days DC was determined. For each time interval 5 specimens were pulverized, pressed with KBr and analyzed with FT-IR. The TG measurements were performed in Netzsch TG 209 under oxygen atmosphere and heating rate of 10°C/min from 25 to 700°C. A two-way ANOVA showed DC (%) mean values statistically significance differences between two cements (p < 0.05). The Tukey’s test showed no significant difference only for the 24 and 48 h after light irradiation for both resin cements (p > 0.05). The Relx-Y? Unicem mean values were significantly higher than Panavia® F 2.0. The degree of conversion means values increasing with the storage time and the filler content showed similar for both resin cements.  相似文献

The ordering of fuzzy sets over the real line is approached from the point of view of ordering intervals rather than ordering numbers. First, the maximax and maximin criteria which are commonly used for ordering intervals are expressed in terms of characteristic functions. These criteria and the Hurwicz criterion for decision making under complete ignorance are then reformulated in a manner that allows for their generalization to the fuzzy case. Transitivity is established for these ordering rules. A criterion based on the principle of diminishing marginal utility is also presented.  相似文献

The aim of this study was to evaluate the hardness of a dental composite resin submitted to temperature changes before photo-activation with two light-curing unite (LCUs). Five samples (4 mm in diameter and 2 mm in thickness) for each group were made with pre-cure temperatures of 37, 54, and 60°C. The samples were photo-activated with a conventional quartz-tungsten-halogen (QTH) and blue LED LCUs during 40 s. The hardness Vickers test (VHN) was performed on the top and bottom surfaces of the samples. According to the interaction between light-curing unit and different pre-heating temperatures of composite resin, only the light-curing unit provided influences on the mean values of initial Vickers hardness. The light-curing unit based on blue LED showed hardness mean values more homogeneous between the top and bottom surfaces. The hardness mean values were not statistically significant difference for the pre-cure temperature used. According to these results, the pre-heating of the composite resin provide no influence on Vickers hardness mean values, however the blue LED showed a cure more homogeneous than QTH LCU.  相似文献

The purpose of this study was to investigate the effect of Er:YAG laser on surface treatment to the bond strength of repaired composite resin after aged. Sixty specimens (n = 10) were made with composite resin (Z250, 3M) and thermocycled with 500 cycles, oscillating between 5 to 55°C. The specimens were randomly separated in six groups which suffered the following superficial treatments: no treatment (GI, control), wearing with diamond bur (GII), sandblasted with aluminum oxide with 27.5 μm particles (GIII) for 10 s, 200 mJ Er:YAG laser (GIV), 300 mJ Er:YAG laser (GV), and 400 mJ Er:YAG laser (GVI), with the last 3 groups under a 10 Hz frequency for 10 s. Restoration repair was done using the same composite. The shear test was done into the Universal testing machine MTS-810. Analyzing the results through ANOVA and Tukey test, no significant differences were found (p-value is 0.5120). Average values analysis showed that superficial treatment with aluminum oxide presented the highest resistance to shear repair interface (8.91MPa) while 400 mJ Er:YAG laser presented the lowest (6.76 MPa). Fracture types analysis revealed that 90% suffered cohesive fractures to GIII. The Er:YAG laser used as superficial treatment of the aged composite resin before the repair showed similar results when used diamond bur and sandblasting with aluminum oxide particles.  相似文献

The aim of this study was to determine the effect of two light-curing units (QTH and LED) on microleakage of Class II composite resin restorations with dentin cavosurface margins. Twenty extracted mandibular first premolars, free of caries and fractures were prepared two vertical “slot” cavities in the occluso-mesial and -destal surfaces (2 mm buccal-lingually, 2 mm proximal-axially and cervical limit in enamel) and divided into 4 equal groups (n = 8): GI and GII: packable posterior composite light-activated with LED and QTH, respectively; GIII and GIV: micro-hybrid composite resin light-activated with LED and QTH, respectively. The composite resins were applied following the manufacturer’s instructions. After 24 h of water storage specimens were subjected to thermocycling for a total of 500 cycles at 5 and 55°C and the teeth were then sealed with impermeable material. Teeth were immersed in 0.5% Basic fuchsin during 24 h at room temperature, and zero to three levels of penetration score were attributed. The Mann-Whitney and Kruskal-Wallis tests showed significant statistically similar (P > 0.05) from GI to GII and GIII to GIV, which the GII (2.750) had the highest mean scores and the GIII and GIV (0.875) had lowest mean scores. The use of different light-curing units has no influence on marginal integrity of Class II composite resin restorations and the proprieties of composite resins are important to reduce the microleakage.  相似文献

Luting materials provides the retention of endodontic post. However, the failures of endodontic posts predominantly occurred are the losses of retention. Thus, the alternating use to remove the smear layer, open the dentine tubules, and/or etch the inter-tubular dentine can be provided by EDTA. This study was performed to evaluate effect of EDTA on bond strength of glass fiber post cemented with different resin cements to root canal. Fifty bovine incisors were selected and the crowns were removed to obtain a remaining 14-mm-height root. The roots were randomly distributed into five groups: GI: RelyX? ARC/LED; GII: RelyX? U100/LED; GIII EDTA/RelyX? U100/LED; GIV: Multilink?; and GV: EDTA/Multlink?. After endodontic treatment, the post space was prepared with the drills designated for the quartz-coated-carbon-fiber post Aestheti-Post®. Before application of resin cements, root canals were irrigated with 17% EDTA (GIII and GV) during 1 min, rinsed with distilled water and dried using paper points. The light-cured materials were light-activated with UltraLume LED 5 (Ultradent, South Jordan, Utah) with power density of 1315 mW/cm2. Specimens were perpendicularly sectioned into approximately 1 mm thick sections and the stubs were performed on Universal Testing Machine. The analysis of variance (ANOVA) and Tukey’s post-hoc tests showed significant statistical different between RelyX? ARC (GI) and RelyX? U100 independent of the pre-treatment (GII to GIII) (P < 0.05). The Multlink? showed between RelyX? ARC and RelyX? U100 (GI to GIII; GII to GV) (P < 0.05). The ANOVA showed significant statistical similar (P > 0.05) to all resin cements between the Cervical to Apical regions (GI to GV). The use of 17% EDTA showed no difference significant between the resin cements evaluated (GII to GIII; GIV to GV). Within the limitations of the current study, it can be concluded that the use of EDTA did not provide efficiency on bond strength. The RelyX? ARC showed higher bond strength values than RelyX? The interaction between influenza virus hemagglutinins and host cell with terminal sialic acid linked receptors, SA-α-2,6-Gal for human strains is important to obtain insights into this infectious disease. Sambucus nigra lectin has high affinity for SA-α-2,6-Gal receptors. The goals of this work were: to extract the SA-α-2,6-Gal receptors from porcine airways; to perform receptors immobilization and study their storage stability; and to determine some parameters of interaction between the receptor and S. nigra lectin. The receptor isolation was monitored by means of bound sialic acid (BSAc) detection. A major band of protein at 66.7 kDa was clearly visible in SDS-PAGE assay. Eighty-one percent of isolated glycoproteins were immobilized on magnetic nanoparticles. The kinetics of BSAc storage stability at 4 °C was approximated as the first order reaction with kinetic constant and half-life estimated as 0.062 day?1 and 11.2 days, respectively. The dissociation constant (K d) calculated from Scatchard's plot was 2.47?×?10?7 M, and the receptor concentration was equal to 7.92?×?10?5 M. Procedure for N-SA-α-2,6-Gal -receptors extraction based on their affinity to S. nigra lectin with magnetic nanoparticles, and their immobilization in active form, was not described previously, and may have wide application in designing biosensors or virus removal from areas or contaminated samples.  相似文献
