The photoionization of (pro)(n)DHB (pro = proline, DHB = 2,5-dihydroxybenzoic acid, n = 0, 1, 2 or 4) clusters was studied both experimentally and computationally. Experimentally the (pro)(n)DHB clusters are generated in the gas phase by laser desorption and supersonic jet entrainment. The photoionization thresholds are then determined by the mass-selective measurement of both one- and two-color photoionization efficiency curves. These experiments demonstrate that the ionization energies (IEs) of the (pro)(n)DHB clusters are substantially reduced in comparison with the IE of free DHB. Computational studies of the (pro)(n)DHB clusters provide insights into the mechanism of IE reduction. For the (pro)DHB system the IE reduction results from spin delocalization in the ion state of the cluster. In contrast, for the (pro)(2)DHB and (pro)(4)DHB clusters the IE reduction results from an inductive delocalization of electron density from pro to DHB in the ground state of the cluster. This latter effect, which is a result of the specific hydrogen-bonding interactions occurring in the mixed clusters, leads to IE reductions of >1 eV. Finally, determination of the energetics of the (pro)(2)DHB radical cation demonstrate that the DHB-to-proline proton transfer reaction is a barrierless, exoergic process in the ion state and that energetic demands for cluster dissociation to protonated (pro)(2) plus a deprotonated DHB radical are substantially lower than those for cluster dissociation to (pro)(2) plus DHB(+*). Cumulatively, these studies provide new energetic and mechanistic insights into both primary and secondary MALDI ionization processes.  相似文献

The microscopic and macroscopic versions of fluid mechanics differ qualitatively. Microscopic particles obey time-reversible ordinary differential equations. The resulting particle trajectories {q(t)} may be time-averaged or ensemble-averaged so as to generate field quantities corresponding to macroscopic variables. On the other hand, the macroscopic continuum fields described by fluid mechanics follow irreversible partial differential equations. Smooth particle methods bridge the gap separating these two views of fluids by solving the macroscopic field equations with particle dynamics that resemble molecular dynamics. Recently, nonlinear dynamics have provided some useful tools for understanding the relationship between the microscopic and macroscopic points of view. Chaos and fractals play key roles in this new understanding. Non-equilibrium phase-space averages look very different from their equilibrium counterparts. Away from equilibrium the smooth phase-space distributions are replaced by fractional-dimensional singular distributions that exhibit time irreversibility.  相似文献

A Ka-band gyrotron oscillator powered by a compact pulseline accelerator has been operated using oscillator cavities with and without axial slots. The oscillator was operated at high voltage (~900 keV) and high current (~500 A) in the approximate frequency range of 20-50 GHz. The use of axial slots has been shown to suppress low-starting-current whispering-gallery modes, in particular, modes of the TEm2 type, allowing stable operation in a linearly polarized TE13 mode. The content of a polynomial f over a commutative ring R is the ideal c(f) of R generated by the coefficients of f. A commutative ring R is said to be Gaussian if c(fg) = c(f)c(g) for every polynomials f and g in R[X]. A number of authors have formulated necessary and sufficient conditions for R(X) (respectively, R?X?) to be semihereditary, have weak global dimension at most one, be arithmetical, or be Prüfer. An open question raised by Glaz is to formulate necessary and sufficient conditions that R(X) (respectively, R?X?) have the Gaussian property. Creation of lipid partitions by deposition of amphipathic viral peptides   总被引:1,自引:0,他引:1  
Phospholipid vesicles exhibit a natural characteristic to fuse and reform into a continuous single bilayer membrane on hydrophilic solid substrates such as glass, mica, and silica. The resulting solid-supported bilayer mimics physiological tendencies such as lipid flip-flop and lateral mobility. The lateral mobility of fluorescently labeled lipids fused into solid-supported bilayers is found to change upon deposition on the membrane surface of an amphipathic alpha-helical peptide (AH) derived from the hepatitis C virus (HCV) NS5A protein. The binding of the AH peptide to a phospholipid bilayer, with the helical axis parallel to the bilayer, leads to immobilization of the bilayer. We used AFM to better understand the mechanistic details of this specific interaction, and determined that the diminished fluidity of the bilayer is due to membrane thinning. Utilizing this specific interaction between AH peptides and lipid molecules, we demonstrate a novel process for the creation of lipid partition by employing AH peptides as agents to immobilize lipid molecules, thus creating a patterned solid support with partition-defined areas of freely mobile lipid bilayers. This architecture could have a wide range of applications in novel sensing, biotechnology, high-throughput screening, and biomimetic strategies.  相似文献

A novel method for on-tissue identification of proteins in spatially discrete regions is described using tryptic digestion followed by matrix-assisted laser desorption/ionization (MALDI) imaging mass spectrometry (IMS) with MS/MS analysis. IMS is first used to reveal the protein and peptide spatial distribution in a tissue section and then a serial section is robotically spotted with small volumes of trypsin solution to carry out in situ protease digestion. After hydrolysis, 2,5-Dihydroxybenzoic acid (DHB) matrix solution is applied to the digested spots, with subsequent analysis by IMS to reveal the spatial distribution of the various tryptic fragments. Sequence determination of the tryptic fragments is performed using on-tissue MALDI MS/MS analysis directly from the individual digest spots. This protocol enables protein identification directly from tissue while preserving the spatial integrity of the tissue sample. The procedure is demonstrated with the identification of several proteins in the coronal sections of a rat brain.  相似文献

A new approach to on-line oxygen isotope analysis has been developed which utilises existing elemental analyser and mass spectrometry technology to produce a sample of carbon dioxide gas for oxygen isotople analysis. The method relies on on-line high temperature pyrolysis of the sample over a carbon source followed by a rapid, non-contributive partial catalytic oxidation over nickel powder at between 550 and 600 degrees C. Initial results demonstrate both good precision (better than 0.2 per thousand) and accuracy for both cellulose and silver nitrate samples. Copyright 1999 John Wiley & Sons, Ltd.  相似文献
