The synthesis of azadirachtin: a potent insect antifeedant

We describe in full the first synthesis of the potent insect antifeedant azadirachtin through a highly convergent approach. An O-alkylation reaction is used to unite decalin ketone and propargylic mesylate fragments, after which a Claisen rearrangement constructs the central C8-C14 bond in a stereoselective fashion. The allene which results from this sequence then enables a second critical carbon-carbon bond forming event whereby the [3.2.1] bicyclic system, present in the natural product, is generated via a 5-exo-radical cyclisation process. Finally, using knowledge gained through our early studies into the reactivity of the natural product, a series of carefully designed steps completes the synthesis of this challenging molecule.     

Analysis of the Inhibition of the Ergosterol Pathway in Fungi Using the Atmospheric Solids Analysis Probe (ASAP) Method

Fungal cells treated with various inhibitors to the ergosterol pathway were analyzed using an Orbitrap mass spectrometer equipped with an atmospheric solids analysis probe (ASAP). The technique is a rapid means for determining which of the multiple steps in the ergosterol pathway were interrupted by an inhibitor. Furthermore, in an inhibitor concentration study, the ASAP method was able to rapidly provide an estimate of the effectiveness of inhibition. In this method the cells are inserted directly into a hot nitrogen stream, thus eliminating extensive sample workup before analysis. Data indicating the point of pathway interruption are obtained in seconds.     

The Total Synthesis of Eleutherobin: A Surprise Ending

An “sp ² –sp ³ Stille coupling” of the vinyl triflate 1 and the stannyl compound 2 is a key step toward the completion of the total synthesis of eleutherobin, a natural product exhibiting taxol-like cytotoxic activity.     

Use of principal components analysis for displaying variation between pyrograms of micro-organisms

Principal components analysis is used to display variations between batches of pyrograms of micro-organisms represented by sets of peak heights. Although it is not usually suitable for discriminating genus or species groups, it provides a means of detecting aberrant analyses, unsuspected trends in the data and of assessing reproducibility. It can also provide information relating specific peaks or sets of peaks to clusters of samples.These uses of principal components analysis are not confined to pyrolysis—gas—liquid chromatography; the technique can be applied widely to quantitative chromatographic and mass spectrometric data.     

The first three anisotropy constants of nickel

The technique of ferromagnetic resonance at 23 GHz has been used to determine the first three anisotropy constants of pure Ni down to 4.2K. A temperature and orientation dependent linewidth has also been observed.     

Imaging the long-range dipolar field in structured liquid state samples

We describe imaging experiments in which the pattern of the dipolar field generated by spatially modulated nuclear magnetization is directly visualized in simply structured phantoms. Two types of experiment have been carried out at 11.7 T using (1)H NMR signals. In the first, the field from a single spin species is imaged via its own NMR signal. In the second, the NMR signal from one spin species is used to image the field generated by a second species. The field patterns measured in these experiments correspond well with those calculated using simple theoretical expressions for the dipolar field. The results also directly demonstrate the spatial sensitivity of the signal generated using dipolar field effects, indicating that the range of the field depends upon the inverse of the spatial frequency with which the magnetization is modulated.
