High-resolution Hadamard transform microscope fluorescence imaging: quantifying the DNA content in single cells

This study presents a novel miniaturized Hadamard transform fluorescence imaging microscope, by combining a conventional fluorescence microscope with Hadamard transform multiplexing encoding using a one-dimensional movable mask to realize spatial resolution and a linear CCD for multichannel detection. The microscope can provide high-resolution automatically-generated 0–255 gray level HT images for morphological analysis and visualization of a single cell, and normalized HT images for cellular quantitative measurements. The microscopes imaging capability was applied to measure the DNA content in human lymphocyte, chicken erythrocyte and eel erythrocyte, and a comparative study was performed. The results show that the calibrated DNA content in a chicken erythrocyte is 2.32 pg when human lymphocyte is used as the standard, and eel erythrocyte may be a potentially reliable and novel standard for determining DNA contents in other species because it has a stable DNA value of 2.06 pg, with a CV of 4.3% when 20 eel erythrocytes are measured. The results also demonstrate that the HT imaging microscope should be valuable in the fields of medicine and cell biology.     

稀土配合物的时间分辨光谱分析法研究(Ⅰ)TFA-TOPO-农乳-100体系用于测定铕和钐

本文采用自己组装的时间分辨荧光光谱测定装置,研究了铕(钐)-三氟乙酰丙酮-三正辛基膦化氧(Eu(Sm)-TFA-TOPO)在非离子表面活性剂农乳-100存在下的光谱特性、荧光衰减动力学特性和发光机理。提出了能量传递机制和第二配体TOPO在荧光发射过程中的作用,制定出TFA-TOPO-农乳-100体系测定痕量铕和钐的时间分辨荧光光谱分析方法,用于测定高纯氧化钇中的痕量铕和钐,结果令人满意。

Preparation and evaluation of <Superscript>131</Superscript>I-quercetin as a novel radiotherapy agent against dedifferentiated thyroid cancer

Here we reported the radiolabeling and evaluation of a novel ¹³¹I-radiolabeled quercetin for the treatment of dedifferentiated thyroid cancers. The human thyroid cancer cell lines (FTC-133, TT and DRO) experienced much higher uptake of ¹³¹I-quercetin as compared to the free ¹³¹I. And the proliferation inhibition rate of ¹³¹I-quercetin on in vitro DRO cell line was 86.87 ± 7.15%. Biodistribution and SPECT analysis demonstrated that the injected radioactivity mainly accumulated in tumors. The tumor volume in the treatment group was dramatically inhibited in comparison with the control group.

Binding and cytotoxicity of 131I-labeled gastrin-releasing peptide receptor antagonists modified by cell penetrating peptides

Gastrin releasing peptide receptors (GRPRs) are one of the most interesting targets over expressed in various tumors. Due to the superior potential of the GRPR antagonist analogs, they have been studied in the tumor radio imaging and therapy field. However, typical antagonists suffered the shortcomings of no internalization and poor binding affinity which hampered their applications in radiotherapy. Therefore, we attempted to introduce Oligoarginines (cell penetrating peptides) to RM26, aiming to increase the binding affinity or even trigger the internalization of the peptides on cells. The results showed Arg₆ as the most potent CPP, significantly enhanced the binding avidity of RM26 to the GRPR.

本文用自制的Hadamard变换显微图象分析仪分析了AO和FITC染色的单细胞的荧光光谱,用该仪器获取了单个洋葱细胞核内DNA的分布图象,并在535nm处分别测定了AQ染色的鸡红血细胞及洋葱表皮细胞核的荧光强度。所得结果与生物学结果吻合,显示了该仪器在单细胞试样分析中的应用价值。     

Analysis of DNA in Single Pollen Cell by Hadamard Transform Microscopic Fluorescence Imaging

A noncommercial Hadamard transform microscopic image analysis system was used to measure fluorescence emitted by single cells. A method for subtracting the native fluorescence of theZephyranthes candida(Lindl.) Herb. pollen cell was developed based on its correlation to pollen area. Acridine orange and ethidium bromide were used as fluorescence probes for pollen DNA quantitative analysis, and the results obtained by the two methods were compared.
