The methodology for using DPASV to study cadmium complexation in seawater is evaluated using EDTA as a model ligand and by analysing natural samples. The results show that the methodology gives an accurate evaluation of metal complexation when inert complexes are studied, both as regards the ligand concentration and the conditional stability constant; the error for both the parameters is lower than 10% at a ligand concentration of about 10(-8) M and a conditional stability constant of 10(9) M-1. Cadmium complexes with ligands present in natural seawater show an evident kinetic lability that may lead to underestimation of the conditional stability constant when a working electrode characterised by a very thick diffusion layer is used. The conditional stability constant in one water sample of the Adriatic coast ranged between 0.14 and 1.4 l/nmol using a rotating disk electrode at rotation rates of 300 and 6000 rpm. The results of cadmium complexation obtained for samples collected in coastal seawater show that the ligands present low specificity for the metal.  相似文献

Two species of edible clams Venerupis aurea laeta and Cerastoderma edule glaucum from Ganzirri Lake (Sicily, Italy) were investigated to determine OCP and PCB residues by GC-MS. Thirty-five samples were examined for two years in order to perform: a) their actual contamination; b) the daily dietary exposure of seafood consumers to the pollutants in question. In all the clams analysed in this study, PCB and OCP (4,4′-DDE) residues are always below the limits fixed by law. It seems that there is some seasonality of concentrations observed. In fact the more high concentrations of residues are detected in the hot months, for both species and years. Given the consumption of clams and the values obtained in this study for the two species analysed, it is possible estimate that the average daily dietary intake of NDL-PCB and of 4,4′-DDE for adults is really low.  相似文献

Angelman syndrome (AS) is a severe neurobehavioural disorder caused by failure of expression of the maternal copy of the imprinted domain located on 15q11-q13. There are different mechanisms leading to AS: maternal microdeletion, uniparental disomy, defects in a putative imprinting centre, mutations of the E3 ubiquitin protein ligase (UBE3A) gene. However, some of suspected cases of AS are still scored negative to all the latter mutations. Recently, it has been shown that a proportion of negative cases bear large deletions overlapping one or more exons of the UBE3A gene. These deletions are difficult to detect by conventional gene-scanning methods due to the masking effect by the non-deleted allele. In this study, we have used for the first time multiplex ligation-dependent probe amplification (MLPA) and comparative multiplex dosage analysis (CMDA) to search for large deletions affecting the UBE3A gene. Using this approach, we identified a novel causative deletion involving exon 8 in an affected sibling. Based on our results, we propose the use of MLPA as a fast, accurate and inexpensive test to detect large deletions in the UBE3A gene in a small but significant percentage of AS patients.  相似文献

L-Carnosine (β-alanyl-L-histidine, LCar) is the most widely and abundantly distributed copper(II)-coordinating endogenous dipeptide. Though its physiological role has not been completely understood yet, many functions have been proposed for this compound. LCar might be crucial in the potential reduction or prevention of several pathologies in which the metal ions are thought to be involved. The potential therapeutic applications of LCar are drastically limited because of hydrolysis by specific dipeptidases (carnosinases). D-Carnosine (DCar), the enantiomer of the naturally occurring dipeptide, shows the same properties as those of LCar, but it is not hydrolyzed by carnosinases. Chemical modification of LCar has been proposed as a promising strategy to reduce its enzymatic hydrolysis; conjugation of a carbohydrate moiety may also improve site-specific transport to different tissues, which would enhance the peptide bioavailability. On this basis, we have functionalized DCar with β-cyclodextrin (CDDCar) and characterized the compound via NMR. The copper(II) binding properties of the new DCar derivative were investigated by spectroscopic techniques (UV-vis, circular dichroism, electron paramagnetic resonance) and potentiometric measurements. The results surprisingly revealed a pronounced difference from the analogous LCar derivative (CDLCar), especially concerning the dimeric species. The spectroscopic data show that this stereoselectivity is driven by noncovalent interactions, namely, hydrogen bonds, CH-π interactions, and steric and hydrophobic effects of the cyclodextrin cavity.  相似文献

The well known correspondence between even cycles of an undirected graph and polynomials in a binomial ideal associated to a graph is extended to odd cycles and polynomials in another binomial ideal. Other binomial ideals associated to an undirected graph are also introduced. The results about them with topics on monomial ideals are used in order to show decision procedures for bipartite graphs, minimal vertex covers, cliques, edge covers and matchings with algebraic tools. In the present study we report on the optimization and validation of a sensitive high performance liquid chromatography atmospheric pressure chemical ionization mass spectrometry (HPLC–APCI–MS) method for the determination of 8 bioactive amines (histamine, tyramine, tryptamine, 2-phenylethylamine, cadaverine, putrescine, spermidine and spermine) in donkey milk samples. The method involves donkey milk pre-treatment to remove proteins and pre-column dansylation of the amines. HPLC in reversed phase mode has been used for bioactive amines separation and the operating condition of the APCI–MS system proved to be powerful and very efficient for peak assignment. The separation was accomplished in a short time with an excellent resolution for all the amine peaks. Quantification was carried out by monitoring the characteristic [M+H]+ ion of each amine derivative. The method sensitivity, linearity and repeatability were assayed with satisfactory results. The detection limits of the analysed amines ranged from 0.5 μg L−1 to 15 μg L−1; the highest LOD was for spermine. Also remarkably good recovery values were obtained; at the lowest spiking level (1 μg L−1) the percent mean recoveries ranged from 77.7 to 109.7. Furthermore, as the investigations relate to a complex matrix as donkey milk, suitable studies on matrix effect were performed. Finally, the developed and validated method was applied to analyse 13 donkey milk samples. Among the identified bioactive amines, putrescine, spermine and spermidine proved to be the main amines in donkey milk. Their concentration levels in the present study were lower than the values determined in mature human, cow and sow milk.  相似文献

Dried seeds of four varieties of Phaseolus vulgaris, three of Vigna unguiculata ssp. unguiculata and two of Vigna angularis grown and marketed in Italy, Mexico, India, Japan, Ghana and Ivory Coast were analysed for fatty acids content. In oils from seeds of P. vulgaris, the main fatty acids were linolenic (34.7–41.5%) and linoleic (30.7–40.3%), followed by palmitic (10.7–16.8%). The first three aforementioned fatty acids in the lipid fraction of V. unguiculata varieties were 28.4, 28.7 and 26.2%, respectively; while in V. angularis varieties, main fatty acids were linoleic (36.4–39.1%) and palmitic (26.9–33.3%), followed by linolenic (17.9–22.2%). Statistical analyses indicate that botanical species play a rule in bean fatty acids distribution, while the same was not verified for geographical origin. Furthermore, the atherogenic index (AI) and the thrombogenic index (TI) were investigated for health and nutritional information. The results showed that these wide spread legumes have functional features to human health.  相似文献

The concentrations of 19 elements in wild and genetically modified Nicotiana langsdorffii (N. langsdorffii) exposed to Chromium (VI) and to water deficit were determined and compared to provide new information about their response to abiotic stress.

Genetic modifications by GR and RolC genes (encoding for the rat glucocorticoid receptor and for Agrobacterium rhizogenes RolC, respectively) were investigated because they induce significant, but only partially known changes in the plant response to stress.

Simultaneous determination of Al, As, Ba, Ca, Cd, Co, Cr, Cs, Cu, Fe, K, Mg, Mn, Na, Pb, Rb, Sr, V and Zn was carried out by ICP-MS equipped with a collision/reaction cell (ICP-ORS-MS). The methodology was optimised by testing the grinding, homogenisation, digestion and analysis procedures, to reduce the uncertainty of the experimental results and to identify statistically significant differences between nine sample pools, for a total of 75 samples. The quality control procedure was carried out by blank control and by evaluating the detection limits and repeatability. Trueness was assessed by analysing certified reference material, NIST 1573a.

Significant differences were observed in the uptake and accumulation of several elements in the wild-type N. langsdorffii samples, either with respect to the plants submitted to water deficit and exposure to Cr(VI) or with respect to the genetically modified plants. The differences were highlighted by principal component analysis (PCA). The analysis of the element content of the whole plant, combined with the data found in the literature, allows us to hypothesise effects on the metabolic mechanism controlling the uptake and translocation of elements inside the vegetal organism. Because genetic and chemical stress decreases the nutrient concentration in the whole plant, we can say that the uptake at root level is affected. The increase in concentration of elements such as As, Sr and Al indicates a decreased selectivity in the uptake of potentially toxic elements and, consequently, highlights the effects on the plant’s metabolic processes.  相似文献
