Effect of aeration on lignin peroxidase production by Streptomyces viridosporus T7A

The effect of aeration on lignin peroxidase production by Streptomyces viridosporus T7A was studied in a bench-scale bioreactor using a previously optimized growth medium (0.65% yeast extract and 0.1% corn oil, pH7.0) at 37°C and natural pH. Airflow rates of 0.3, 1.0, and 1.5 vvm and a fixed agitation of 200 rpm were initially studied followed by 1.0 vvm and 200, 300, 400, and 500 rpm. The use of 1.0 vvm and 400 rpm increased enzyme concentration 1.8-fold (100–180 U/L) and process productivity 4.8-fold (1.4–6.7 U/[L·h]) in comparison with the use of 200 rpm and 0.3 vvm. The inexpensive corn oil, used as carbon source, besides its antifoam properties, proved to be nonrepressive for enzyme production.     

Tetraarylester der μ-Imidodiphosphorsäure und ihre Thioderivate — Strukturuntersuchungen

Tetraarylesters of μ-Imido-Diphosphoric Acid and its Thio Derivatives — Structure Investigations New O,O′,O″,O?-tetratolyl- and ditolyl-diphenylesters of the μ-imido-diphosphoric acid and its mono and dithio derivatives were synthesized, compared with the corresponding tetraphenylesters and investigated by ¹H, ¹³C, and ³¹P NMR spectroscopy and X-ray crystal structure analysis. Structures of the O,O′,O″,O?-tetrakis-(2-methyl-phenyl)-μ-imidodiphosphate, 1b , as well as of the corresponding ortho-, meta- and para-tolylesters of the μ-imido-monothiodiphosphoric acid ( 2a , 2b , 2c ) were determined. All the compounds form dimers via N? H…?O hydrogen bonds in the crystal as well as in nonpolar solvents. The distances around the phosphorus atoms rise with decreasing electronegativity of the phosphorus substituents. Signs of the ²J_{P? N? P} coupling constants were determined by ¹³C{¹H, ³¹P} triple resonance experiments for some compounds. These constants become more negative owing to substitution of a phosphoryl by a thiophosphoryl group.     

De novo synthesis of 4'-ethoxy nucleoside analogues

Butenolides 5a and 13 were used as optically active templates in the de novo synthesis of 4'-disubstituted nucleoside analogues. The butenolides were reduced and acylated in situ to give acetates 10 and 14. Vorbrüggen coupling gave the protected nucleoside analogues 11 and 15. Reduction of 11 gave 4'-ethoxy-2',3'-dideoxythymidine (6) and deprotection of 15 gave 4'-ethoxy-2',3'-dideoxydidehydrothymidine (7). The cis-dihydroxylation of a variety of butenolides occurred with the major product formed from oxidation of the beta-face.

Zusammenfassung Der Bleigehalt von pflanzlichem Material lÄ\t sich nach einer modifizierten photometrischen Bestimmungsmethode schnell und zuverlÄssig erfassen. Das extraktive Dithizonverfahren für Blei wurde hierzu systematisch getestet und hinsichtlich Schwierigkeitsgrad und Zeitbedarf der Arbeitsoperationen entscheidend vereinfacht. Absolutmengen bis zu einer unteren Grenze von 2 g bzw. 1 g Pb können in 2 Arbeitsbereichen mit einer Standardabweichung von ± 0,5 g bzw. ± 0,35 g Pb sicher bestimmt werden. Die Arbeit zeigt darüber hinaus den Gebrauch und den Wert einfacher mathematisch-statistischer Hilfsmittel für den Vergleich und die Beurteilung von Test- und Analysendaten.

Summary A modified photometric method is recommended for the rapid and reliable determination of lead in plant material. The dithizone extraction has been tested systematically and has been essentially simplified with regard to difficult operations and time consumption. Quantities down to a lower limit of 2 g or 1 g of Pb can be safely determined within two working ranges with a standard deviation of ±0.5 g or 0.35 g of Pb, respectively. Furthermore, the use of simple mathematicalstatistical means for comparison and evaluation of analytical data is shown.
