In our work we performed a combinatorial solid-phase synthesis in aqueous medium to prepare peptide libraries from which to select an amino acid sequence with binding properties towards aflatoxins.We used polystyrene beads, functionalised with carboxylic groups as solid support and eight amino acids as monomers. During the first step 64 different sequences of two amino acids were prepared by exploiting the principles of combinatorial chemistry; then the binding properties of all sequences towards aflatoxin B(1 )were checked. We determined binding constants towards aflatoxin B(1) and towards aflatoxins B(2), G(1) and G(2). Results were promising, so we prepared a new library by using the selected dipeptide as the starting solid phase. After selecting the best tetrapeptide sequences, we determined binding constants towards the quoted aflatoxins. We obtained binding constants ( K>10(4) M(-1)) similar to those shown by human serum albumin for similar compounds. Preliminary studies on an extraction column were promising for the development of an SPE system and for its application in food matrices.  相似文献

The ion chemistry of 1-bromo-1-chloro-2,2,2-trifluoroethane (the common anesthetic halothane) in air plasma at atmospheric pressure was investigated by atmospheric pressure chemical ionization mass spectrometry (APCI-MS). The major positive ion observed at low declustering (API interface) energies is the ionized dimer, M(+.)M, an unexpectedly abundant species which possibly is stabilized by two H-bonding interactions. At higher energies [M--HF](+.) and [M--Br](+) prevail; the former, corresponding to ionized olefin [ClBrC=CF(2)](+.), appears to originate from M(+.)M and is quite stable towards fragmentation. The latter fragment ion ([M--Br](+)) and its analogue, [M--Cl](+), which is also observed though at much lower abundance, are originally ethyl cations (+)CHX--CF(3) (X = Br, Cl) which, upon collisional activation, rearrange and fragment to CHFX(+) via elimination of CF(2). All of the above described ions are also observed in humid air: in addition, the oxygenated ion [ClBrC=CFOH](+.) also forms in humid air via water addition to [ClBrC=CF(2)](+.) and HF elimination, as observed earlier for ionized trichloroethene. In contrast with similar chloro- and fluoro-substituted ethanes, halothane does not react with H(3)O(+) in the APCI plasma, a result confirmed by selected ion APCI triple-quadrupole (TQ) experiments. Major negative ions formed from halothane in the air plasma are Br(-) and, to a lesser extent, Cl(-), and their complexes with neutral halothane. APCI-TQ experiments indicated that Br(-) and Cl(-) are formed via reaction of halothane with O(2) (-.), O(2) (-.)(H(2)O) and O(3) (-.), possibly via dissociative electron transfer or nucleophilic substitution. Competing proton transfer was also observed in the reaction with O(2) (-.) and, at high halothane pressure, also with O(2) (-.)(H(2)O); at lower pressures the molecular anion M(-.) was observed instead. The other minor anions of the air plasma, NO(2) (-), N(2)O(2) (-.) and NO(3) (-), were found to be unreactive towards halothane.  相似文献

The current opinion about molecular imprinted polymers (MIPs) is that their molecular recognition properties are due to the presence of nanocavities formed during a polymerization process developed in the presence of a template molecule. According to this principle, the shape of these nanocavities is complementary to that of the template and non-covalent interactions are established between the binding site and a single template molecule. Nevertheless, there are some experimental indications that the real molecular recognition mechanism involves clusters of template molecules being packed into the binding site. Recently, it has been proposed that template molecules covalently linked to the binding site can act as nucleation points, enhancing the formation of these molecular clusters.We have tested this hypothesis by studying the adsorption isotherms of polymers prepared by imprinting them with 2,4,5-trichlorophenoxyacetic acid (2,4,5-T). Three different polymers were considered: P0, prepared without the template, P1, whose template was represented by 2,4,5-T molecules, and P2, whose template was 1/3 constituted by the polymerisable 2-(2,4,5-trichlorophenoxyacetoxy)-ethylmethacrylate (2,4,5-TEMA) and 2/3 by 2,4,5-T. The polymers were prepared by thermoinduced polymerization of template mixtures, 4-vinylpyridine and ethylene dimethacrylate. The crushed polymers were packed into HPLC columns and frontal chromatographic runs were performed by eluting the columns with a mobile phase containing variable amounts of 2,4,5-T.The experimental adsorption isotherms were fitted by using several isotherm models, and the Freundlich-Langmuir model was found to give the best fitting in terms of F-test. All the models considered showed a significant difference between the affinity constant values measured for the polymer P1 and P2, with a higher value for the polymer P2 (for Freundlich-Langmuir model: polymer P1, k=(2.00±0.43)×104 M−1; polymer P2, k=(1.93±0.0535)×105 M−1; ratio P2/P1, 9.65±2.09). Such experimental results support the hypothesis that a polymer prepared with a limited amount of template covalently attached to the binding site shows an increased affinity for the template itself.  相似文献

The preliminary results described here show the complete transfer of Hg(II) ions, in the absence of a chelating agent in 1-alkyl-3-methylimidazolium hexafluorophosphate ionic liquids; the lag time required to gain the quantitative metal ion partition turned out to be strongly dependent both on alkyl chain length on the imidazolium ring and on the working temperature.  相似文献

In this work we present a method for confirmatory analysis of chloramphenicol (CAP) in bovine and buffalo raw milk. CAP is extracted in acetonitrile and purified by affinity chromatography on an alpha-1-acid glycoprotein (AAG) column, then is identified and determined by ion-trap liquid chromatography/electrospray ionisation tandem mass spectrometry (LC/ESI-MS/MS) analysis in the negative ion mode. CAP was identified at the minimum required performance limit (MRPL) of 0.30 ppb, by monitoring the [M-H]- ion and at least two product ions, meeting the qualitative and quantitative criteria set by the European Commission in Decision 2002/657/EC for confirmation of prohibited veterinary drugs. The trueness and within-day and between-day repeatability data are also reported. Moreover, the loading capacity of affinity columns towards CAP was tested. This method, based on the molecular recognition between drug and AAG during the purification step to improve sample cleanup, represents a quantitative and repeatable procedure for confirmatory analysis, and fits the requirements for the routine official control of CAP residues in raw milk.  相似文献

Summary Treatment of [Mo{HB(3,5-Me2C3HN2)3}(NO)I2] with one or two moles of AgPF6 in acetonitrile afforded the paramagnetic (one unpaired electron) complex [Mo{HB(3,5-Me2C3NH2)3}(NO)(NCMe)2][PF6]. The structure of this complex was determined crystallographically, and the six-coordinate geometry of the complex cation confirmed.  相似文献

In this paper, we report a spectroelectrochemical investigation of proton-coupled electron transfer in flavodoxin D. vulgaris Hildenborough (Fld). Poly-L-lysine is used to promote the binding of Fld to the nanocrystalline, mesoporous SnO(2) electrodes. Two reversible redox couples of the immobilized Fld are observed electrochemically and are assigned by spectroelectrochemistry to the quinone/semiquinone and semiquinone/hydroquinone couples of the protein's flavin mononucleotide (FMN) redox cofactor. Comparison with control data for free FMN indicates no contamination of the Fld data by dissociated FMN. The quinone/semiquinone and semiquinone/hydroquinone midpoint potentials (E(q/sq) and E(sq/hq)) at pH 7 were determined to be -340 and -585 mV vs Ag/AgCl, in good agreement with the literature. E(q/sq) exhibited a pH dependence of 51 mV/pH. The kinetics of these redox couples were studied using cyclic voltammetry, cyclic voltabsorptometry, and chronoabsorptometry. The semiquinone/quinone reoxidation is found to exhibit slow, potential-independent but pH-sensitive kinetics with a reoxidation rate constant varying from 1.56 s(-)(1) at pH 10 to 0.0074 s(-)(1) at pH 5. The slow kinetics are discussed in terms of a simple kinetics model and are assigned to the reoxidation process being rate limited by semiquinone deprotonation. It is proposed that this slow deprotonation step has the physiological benefit of preventing the undesirable loss of reducing equivalents which results from semiquinone oxidation to quinone.  相似文献
