An RPLC was developed to rapidly determine lipophilicity of neutral and basic compounds using three base deactivated RPLC stationary phases particularly designed for the analysis of basic compounds, namely, Supelcosil ABZ(+)Plus, Discovery RP Amide C16, and Zorbax Extend C18. The work consisted of three sets of experiments. In the first log kw values of neutral compounds were extrapolated using hydroorganic mobile phases at different compositions. Good correlation between log kw and log Poct indicated that the method was appropriate for these supports, without adding a silanol masking agent. In the second set of experiments, isocratic log k values of neutral and basic compounds were measured with three different mobile phases. The best estimation of lipophilicity was obtained for neutral and basic compounds when the secondary interactions were strongly reduced (i. e., when basic compounds were under their neutral form). In the third set of experiments, isocratic retention factors of basic compounds (in their neutral form) were measured with a high-pH mobile phase, on a chemically stable support (Zorbax Extend C18). Under these chromatographic conditions, correlation between the isocratic retention factors and log Poct (log D10.5) for basic compounds was similar to that for neutral compounds. 相似文献

Flavins were extracted from sporangiophores of the lower fungus Phycomyces blakesleeanus and identified by HPLC with fluorescence detection. In the wild-type strain NRRL1555 they were found to be present at the following concentrations: riboflavin (5.5 x 10(-6) M), flavin mononucleotide (FMN) (4.0 x 10(-6) M) and flavin adenine dinucleotide (1.4 x 10(-6) M). The HPLC elution profiles of the wild type were compared to a set of behavioral mutants (genotype mad) with specific defects in their light-transduction pathway. The photoreceptor mutants C109 (madB), C111 (madB) and L1 (madC) had normal amounts of flavins. The most prominent changes were found in single mutants with a defective madA gene which contained about 25% of riboflavin and about 10% of FMN and FAD normally found in the wild type. A hypertropic mutant with a defective madH gene contained instead 80% of riboflavin and 120% of FMN and FAD. The double mutant L52 (madA madC) and the triple mutant L72 (madA madB madC) had normal amounts of FAD and FMN. This indicates that the madC mutation, which itself causes loss of light sensitivity and which affects the near-UV/blue-light receptor (Galland and Lipson, 1985, Photochem. Photobiol. 41, 331-335) functions as a restorer of the flavin content in a genetic madA background. The double mutant L51 (madA madB) had about 40% of FMN and FAD, suggesting that the madB mutation functions as a partial restorer of flavin content. The status of near- UV/blue-light research is described and critically assessed. It is argued that the generally accepted proposition of flavins acting as near- UV/blue-light photoreceptors is reasonable, but that also other pigments, specifically pteridines and pterins, should be considered. In view of the fact that a receptor assay, applicable to any organism and near-UV/blue-light reaction, is not yet at hand, caution appears to be indicated as to what one can accept as a reasonable “criterion” for the involvement of any proposed photoreceptor pigment. 相似文献

The coupling between an electrochemical cell (EC) and a mass spectrometer (MS) is a useful screening tool (EC-MS) to study the oxidative transformation pathways of various electroactive species. For that purpose, we showed that the EC-MS method, carried out in the presence and absence of isotope 18O labeled water leads not only to a fast identification of oxidation products but also leads to a fast elucidation of the mechanism pathway reaction. We examined herein the case of the electrochemical hydrolysis of activated aromatic ether. Acebutolol (β-blockers) was selected herein as model of activated aromatic ether, and its electrochemical oxidation was examined in both the presence and absence of isotope 18O labeled water. To elucidate electrochemical hydrolysis pathway reaction: O-dealkylation or O-dealkoxylation, our approach was used to prove its applicability. The electrochemical oxidation mechanism was then elucidated showing an O-dealkoxylation reaction. It is generally assumed that astatide (At?) is the predominant astatine species in basic aqueous media. This assumption is questioned in non‐complexing and non‐reductive aqueous solutions by means of high‐pressure anion‐exchange chromatography. Contrary to what is usually believed, astatide is found to be a minor species at pH=11. A different species, which also bears a single negative charge, becomes predominant when the pH is increased beyond 7. Using competition experiments, an equilibrium constant value of 10?6.9 has been determined for the formation of this species from AtO(OH) with the exchange of one proton. The identification of this species, AtO(OH)2?, is achieved through relativistic quantum mechanical calculations, which rule out the significant formation of the AtO2? species, while leading to a hydrolysis constant of AtO(OH) in excellent agreement with experiment when the AtO(OH)2? species is considered. Beyond the completion of the Pourbaix diagram of astatine, this new information is of interest for the development of 211At radiolabeling protocols. 相似文献
