Losartan, a highly effective blood pressure-lowering agent, has been widely used for the treatment of hypertension. A fast and reliable method for the determination of losartan was highly desirable to support formulation screening and quality control. A first-derivative UV spectroscopic method and HPLC were developed for the determination of losartan in the tablet dosage form. The first-derivative spectrum recorded between 220 and 320 nm and a zero-crossing technique for first-derivative measurement at 232.5 nm were selected. The selectivity and sensitivity of the method was in desirable range. In comparison with the direct UV method, first-derivative UV spectroscopy has a definite trough without any interference from UV absorbing-excipients. This method is also fast and economical in comparison with the more time-consuming HPLC method regularly used for formulation screening and quality control and can be used routinely by any laboratory possessing a spectrophotometer with a derivative accessory. The linear concentration ranges were 2-50 microg ml(-1), (D(1)=-0.0159C-0.0056, r=0.9994, n=6). Between-days CV of < or =2.9%, within-day CV of < or =2.1%, and analytical recovery close to 98.1% show the suitability of the method for determination in quality control.  相似文献

A novel sensitive method for detection of DNA methylation was developed with thioglycollic acid (TGA)-capped CdTe quantum dots (QDs) as fluorescence probes. Recognition of methylated DNA sites would be useful strategy due to the important roles of methylation in disease occurrence and developmental processes. DNA methylation occurs most often at cytosine-guanine sites (CpG dinucleotides) of gene promoters. The QDs significantly interacted with hybridized unmethylated and methylated DNA. The interaction of CpG rich methylated and unmethylated DNA hybrid with quantum dots as an optical probe has been investigated by fluorescence spectroscopy and electrophoresis assay. The fluorescence intensity of QDs was highly dependent to unmethylated and methylated DNA. Specific site of CpG islands of Adenomatous polyposis coli (APC), a well-studied tumor suppressor gene, was used as the detection target. Under optimum conditions, upon the addition of unmethylated dsDNA, the fluorescence intensity increased in linear range from 1.0?×?10??10 to 1.0?×?10??6M with detection limit of 6.2?×?10??11 M and on the other hand, the intensity of QDs showed no changes with addition of methylated dsDNA. We also demonstrated that the unmethylated and methylated DNA and QDs complexes showed different mobility in electrophoresis assay. This easy and reliable method could distinguish between methylated and unmethylated DNA sequences.  相似文献

The electrochemical behavior of D ‐penicillamine (D ‐PA) studied at the surface of ferrocene carboxylic acid modified carbon paste electrode (FCAMCPE) in aqueous media using cyclic voltammetry and double step potential chronoamperometry. It has been found that under optimum condition (pH 7.00), the oxidation of D ‐PA at surface of such an electrode is occurred about 420 mV less positive than that an unmodified carbon paste electrode (CPE). The catalytic oxidation peak current was linearly dependent on the D ‐PA concentration and a linear calibration curve was obtained in the ranges 7.5×10?5 M – 1.0×10?3 M and 6.5×10?6 M?1.0×10?4 M of D ‐PA with cyclic voltammetry (CV) and differential pulse voltammetry (DPV) methods respectively. The detection limits (3σ) were determined as 6.04×10?5 M and 6.15×10?6 M. This method was also used for the determination of D ‐PA in pharmaceutical preparation (capsules) by standard addition method.  相似文献

In this article, a novel triple-stimuli hydrogel was prepared by simultaneous formation of super paramagnetic iron oxide nanoparticles (SPION) and crosslinking of poly (acrylic acid) grafted onto kappa carrageenan (κC-g-PAA). The structure, thermal stability, surface morphology, and magnetic property of the κC-g-PAA/SPION hydrogel were characterized by Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy with energy dispersive X-ray analysis (SEM–EDAX), transmission electron microscopy (TEM), thermal gravimetric analysis (TGA), and vibrating sample magnetometer (VSM). Moreover, swelling capacity of the obtained hydrogel was measured at different temperature, pH, and magnetic-field to assess the sensitivity of κC-g-PAA/SPION hydrogel. This synthetic hydrogel was also examined as a controlled drug delivery system and defrasirox release was investigated at different temperature, pH, and magnetic-field. The in vitro antibacterial activity of κC-g-PAA/SPION hydrogel was studied against Escherichia coli and Staphylococcus aureus bacteria where the results showed no antibacterial activity of this new hydrogel. In vitro biocompatibility experiments were undertaken using human bladder epithelial cell line HTB 5637. These results indicated the synthesized κC-g-PAA/SPION hydrogel are nontoxic that will be useful for biomedical applications.

The dynamic von Karman equations are used for nonlinear analysis of a thin circular plate made of a functionally graded material. The thickness of the plate is constant and the properties of the functionally graded material depend on temperature and vary throughout the thickness. It is assumed that the plate oscillates with large amplitudes. The forces and moments in the plate are determined in solving the equations for harmonic vibrations. Relevant results are obtained in the case of stead-state free vibrations.
