Procion red HE-3B (RR120) is an example of dye currently used in affinity purification. A method is described for determining trace amounts of RR120 dye contaminant in human serum albumin by cathodic stripping voltammetry. The method is based on a measure of a well-defined peak at −0.58 V, obtained when samples of HSA protein (0.01-2% w/v) containing dye concentrations are submitted to a heating time of 330 min at 80 °C in NaOH, pH 12.0 and the samples are removed to a solution containing Britton-Robinson buffer, pH 4.0. Using an optimum accumulation potential and time of 0 V and 240 s, respectively, linear calibration curves were obtained from 1.0×10−9 to 1.0×10−8 mol l−1 for RR120 dye. Leakage/hydrolysis of reactive red 120 from an agarose support (e.g. at pH 2 or 12) can also be conveniently determined at very low levels (sub-μg ml−1) by means of cathodic stripping voltammetry, which involves adsorptive accumulation of the dye onto the hanging mercury-drop electrode.  相似文献

Poly(vinyl chloride) membrane electrodes containing the specified substances are suitable as indicator electrodes in titrations of basic dyes with sodium tetraphenylborate solution and of acidic dyes with crystal violet solution. Useful curves are obtained even at the 10-3 M level.  相似文献

Oxidized glutathione (GSSG) can be determined after previous accumulation on the HMDE at E > -0.2 V (vs. the Ag AgCl reference electrode). GSH is formed during the accumulation, possibly by a mercury-ion-assisted hydrolytic disproportionation of GSSG. In the subsequent cathodic scan GSH is released and catalyses the reduction of nickel ion, giving a peak located at -0.6 V. This enables the determination of GSSG by differential-pulse cathodic stripping voltammetry at pH 7.0 in the phosphate acetate or MOPS buffer containing 0.5-1.0 mM Ni(II). The detection limit is 10 nM. The calibration graph is linear even in the presence of small amounts of human serum albumin, HSA. However, HSA increases the detection limit (20 nM for 3 x 10(-4)% HSA). Acetyl-cysteine in small excess or Cu(II) present as reagent impurity do not interfere. Glutathione, cysteine and similar compounds, which accumulate as mercury salts and form stable nickel complexes, will interfere. The method is put forward as a novel alternative stripping voltammetric method to those involving accumulation and determination as mercury or copper salts and complexes, in the knowledge that it may have advantages in particular analytical situations. The behavior of (Z)-3-p-tolylsulfinylacrylonitrile (1) as a chiral dienophile has been evaluated from its reactions with furan and acyclic dienes. Electrostatic interactions of the cyano group with the sulfinyl one restrict the conformational mobility around the C-S bond, thus controlling the pi-facial selectivity, which is almost complete in all cases, the approach of the diene from the less-hindered face of the dienophile (that bearing the lone electron pair) in the predominant rotamer being the favored one. The regioselectivity is also completely controlled by the cyano group. Additionally, the reactivity of compound 1 as well as its endo-selectivity are both higher than those observed for the corresponding (Z)-3-sulfinylacrylates, thus proving the potential of sulfinylnitriles as chiral dienophiles.  相似文献

The use of xanthene, triphenylmethane and other basic dye cations as reagents for the determination of anions is reviewed. Elements and anions determined include Sb, Tl, Ga, Au, Te, In, Zn, Ta, Hg, Re, Sn, U, Os, B, Cr, Ag, ClO(4)(-), PO(4)(-), SO(4)(2-) and NO(3)(-). Difficulties encountered in developing satisfactory procedures involving basic dyes are discussed. Procedures for the purification and analysis of basic dye samples are outlined.  相似文献

ABSTRACT: BACKGROUND: The peri-adolescent period is a crucial developmental moment of transition from childhood to emergent adulthood. The present report analyses the differences in Power Spectrum (PS) of the Electroencephalogram (EEG) between late childhood (24 children between 8 and 13 years old) and young adulthood (24 young adults between 18 and 23 years old). RESULTS: The narrow band analysis of the Electroencephalogram was computed in the frequency range of 0--20 Hz. The analysis of mean and variance suggested that six frequency ranges presented a different rate of maturation at these ages, namely: low delta, delta-theta, low alpha, high alpha, low beta and high beta. For most of these bands the maturation seems to occur later in anterior sites than posterior sites. Correlational analysis showed a lower pattern of correlation between different frequencies in children than in young adults, suggesting a certain asynchrony in the maturation of different rhythms. The topographical analysis revealed similar topographies of the different rhythms in children and young adults. Principal Component Analysis (PCA) demonstrated the same internal structure for the Electroencephalogram of both age groups. Principal Component Analysis allowed to separate four subcomponents in the alpha range. All these subcomponents peaked at a lower frequency in children than in young adults. CONCLUSIONS: The present approaches complement and solve some of the incertitudes when the classical brain broad rhythm analysis is applied. Children have a higher absolute power than young adults for frequency ranges between 0-20 Hz, the correlation of Power Spectrum (PS) with age and the variance age comparison showed that there are six ranges of frequencies that can distinguish the level of EEG maturation in children and adults. The establishment of maturational order of different frequencies and its possible maturational interdependence would require a complete series including all the different ages.  相似文献

We propose a picture of Wigner function scars as a sequence of concentric rings along a two-dimensional surface inside a periodic orbit. This is verified for a two-dimensional plane that contains a classical hyperbolic orbit of a Hamiltonian system with 2 degrees of freedom. The stationary wave functions are the familiar mixture of scarred and random waves, but the spectral average of the Wigner functions in part of the plane is nearly that of a harmonic oscillator and individual states are also remarkably regular. These results are interpreted in terms of the semiclassical picture of chords and centers.  相似文献
