Two lots of mangoes of the Nahng Glahng Wahn variety from Thailand were irradiated at the Thai Irradiation Center (TIC) at 0.49 to 0.77 kGy. Following this, one batch was retained in Thailand while the other was shipped to the Canadian Irradiation Center (CIC) for investigation of the same variables during storage. This way, it was hoped to compare the effects of gamma radiation with hot water dip (HWI) or without (I) before and after transportation on the ripening, vitamin C and vitamin A content and sensory qualities of mangoes. The results indicate that the I and HWI treatments delayed the ripening of the mangoes. The irradiated groups appeared to have a slightly higher content of ascorbic acid on the first day after irradiation than their corresponding controls. The sensory evaluation indicates that the panelists mostly favoured the I and HWI irradiated groups for the overall appearance and texture of whole mangoes and pulp and in taste and palatability of the pulp alone. Transportation stress did not appear to have much of an effect on the ascorbic acid and dehydroascorbic acid levels and the sensorial tests. The mangoes stored and studied in Canada appeared to have a lower vitamin A content. 相似文献

This study was undertaken to determine if a combined treatment (marinating in natural plant extracts or vacuum) with irradiation could have a synergetic effect, in order to reduce the dose required for complete elimination of Salmonella on fresh poultry. The effect of these combined treatments on the shelf-life extension was also evaluated. The fresh chicken legs were irradiated at 0, 3 and 5 kGy. The poultry underwent microbial analysis(mesophilic and Salmonella detection). For each treatment, the total microbial count decreased with increase of irradiation dose. The marinating treatment have a synergistic effect with irradiation treatment to reduce the total microbial count and controlling the proliferation during storage at 4°C. Irradiation of fresh chicken pieces with a dose of 3 kGy appears to be able to extend the microbial shelf-life by a factor of 2. When the chicken is marinating and irradiated at 3 kGy or when irradiated at 5 kGy without marinating, the microbial shelf-life is extended by a factor of 7 to 8. No Salmonella was found during all the experiment in the chicken in air and marinated. However, a presence of Salmonella was found in samples irradiated at 5 kGy under vacuum, in unirradiated samples and samples irradiated at 3kGy in air and under vacuum. 相似文献

Group IV metallocene triphenylsiloxy chlorides, Cp2MCl(OSiPh3) (1, M=Ti; 2, M=Zr; 3, M=Hf), and cyclic organohydroborates, Cp2M(OSiPh3){(μ-H)2BC8H14} (4, M=Zr; 5, M=Hf), were synthesized and characterized. The new hafnocene chloride derivative 3 was obtained by treating Cp2HfCl2 with triphenylsilanol and piperidine. The 18-electron cyclic organohydroborates 4 and 5 were afforded by reacting 2 and 3 with K[H2BC8H14], the potassium salt of the 9-BBN dimer. Reaction of 1 with K[H2BC8H14] causes reduction of the Ti(IV) center and produces the well-known Ti(III), 17-electron, paramagnetic dimer [Cp2Ti(μ-Cl)2TiCp2] (6). The evolution of the texture of silica aerogels during sintering is studied by thermoporometry for both neutral and base catalysed materials.During the densification the macroporous volume drops and the analysis of the mesopore size distribution evolution shows that the collapse of the smallest mesopores is responsible for the macropore volume transformation. In order to enhance the shelf-life of edible mature mushrooms Agaricus bisporus, 2 kGy ionising treatments were applied at two different dose rates: 4.5 kGy/h (I−) and 32 kGy/h (I+). Both I+ and I− showed 2 and 4 days shelf-life enhancement compared to the control (C). Before day 9, no significant difference (p>0.05) in L* value was detected in irradiated mushrooms. However, after day 9, the highest observed L* value (whiteness) was obtained for the mushrooms irradiated in I−. Analyses of phenolic compounds revealed that mushrooms in I− contained more phenols than I+ and C, the latter containing the lower level of phenols. The polyphenol oxidase (PPO) activities of irradiated mushrooms, analysed via catechol oxidase and dopa oxidase substrates, resulted in being significantly lowered (p0.05) compared to C, with a further decrease in I+. Analyses of the enzymes indicated that PPO activity was lower in I+, contrasting with its lower phenol concentration. Ionising treatments also increased significantly (p0.05) the phenylalanine ammonia-lyase (PAL) activity. The observation of mushrooms cellular membranes, by electronic microscopy, revealed a better preserved integrity in I− than in I+. It is thus assumed that the browning effect observed in I+ was caused by both the decompartimentation of vacuolar phenol and by the entry of molecular oxygen into the cell cytoplasm. The synergetic effect of the residual active PPO and the molecular oxygen, in contact with the phenols, allowed an increased oxidation rate and, therefore, a more pronounced browning in I+ than in I−. 相似文献

Over the past few years, a large number of studies have been prepared that describe the analysis of peptides and proteins using capillary electrophoresis (CE) and laser-induced fluorescence (LIF). These studies have focused on two general goals: (i) development of automatic, selective and quick separation and detection of mixtures of peptides or proteins; (ii) generation of new methods of quantitation for very low concentrations (nm and subnanomolar) of peptides. These two goals are attained with the use of covalent labelling reactions using a variety of dyes that can be readily excited by the radiation from a commonly available laser or via the use of noncovalent labelling (immunoassay using a labelled antibody or antigen or noncovalent dye interactions). Targeted mutagenesis and gene knock-out can be mediated by triple helix-forming oligonucleotides (TFO) linked to mutagenic agents, such as psoralen. However, this strategy is limited by the availability of homopurine/ homopyrimidine stretches at or near the target site because such sequences are required for high-affinity triplex formation. To overcome this limitation, we have tested TFO conjugated to psoralen via linker arms of lengths varying from 2 to 86 bonds, thereby designed to deliver the psoralen at varying distances from the third strand binding site present at the 3'end of the supFG1 mutation reporter gene. Following triplex formation and UVA irradiation, mutations were detected using an SV40-based shuttle vector assay in human cells. The frequency and distribution of mutations depended on the length of the linker arm. Precise targeting was observed only for linker arms of length 2 and 6, which also yielded the highest mutation frequencies (3 and 14%, respectively). Psoralen–TFO with longer tethers yielded mutations at multiple sites, with the maximum distance from the triplex site limited by the linker length but with the distribution within that range influenced by the propensity for psoralen intercalation at A:T base-pair-rich sites. Thus, gene modification can be extended beyond the site of third strand binding but with a decrease in the precision of the targeting. 相似文献

The cuttlefish belongs to the mollusk class Cephalopoda, considered as the most advanced marine invertebrates and thus widely used as models to study the biology of complex behaviors and cognition, as well as their related neurochemical mechanisms. Surprisingly, methods to quantify the biogenic monoamines and their metabolites in cuttlefish brain remain sparse and measure a limited number of analytes. This work aims to validate an HPLC‐ECD method for the simultaneous quantification of dopamine, serotonin, norepinephrine and their main metabolites in cuttlefish brain. In comparison and in order to develop a method suitable to answer both ecological and biomedical questions, the validation was also carried out on a phylogenetically remote species: mouse (mammals). The method was shown to be accurate, precise, selective, repeatable and sensitive over a wide range of concentrations for 5‐hydroxyindole‐3‐acetic acid, serotonin, dopamine, 3,4‐dihydroxyphenylacetic acid and norepinephrine in the both extracts of cuttlefish and mouse brain, though with low precision and recovery for 4‐hydroxy‐3‐methoxyphenylethylene glycol. Homovanillic acid, accurately studied in rodents, was not detectable in the brain of cuttlefish.
