Mirror-image oligonucleotide ligands (Spiegelmers) that bind to the pharmacologically relevant target gonadotropin-releasing hormone I (GnRH) with high affinity and high specificity have been identified using the Spiegelmer technology. GnRH is a decapeptide that plays an important role in mammalian reproduction and sexual maturation and is associated with several benign and malignant diseases. First, aptamers that bind to D-GnRH with dissociation constants of 50-100 nM were isolated out of RNA and DNA libraries. The respective enantiomers of the DNA and RNA aptamers were synthesized, and their binding to L-GnRH was shown. These Spiegelmers bind to L-GnRH with similar affinity to that of the corresponding aptamers that bind to D-GnRH. We further demonstrated dose-dependent inhibition of GnRH-induced Ca(2+) release in Chinese hamster ovary cells that were stably transfected with the human GnRH receptor. 相似文献

Abstract Tissue samples were collected from ten healthy mature female wood bison (B. bison athabascae) for examination. Livers and kidneys were tested for toxic heavy metals and trace metals considered as essential nutrients for successful reproduction, while fat samples were analyzed for persistent chlorinated hydrocarbons. No elevated levels of toxic heavy metals (arsenic, cadmium, lead, mercury) were found and essential trace elements (copper, manganese, selenium, zinc) were within the acceptable ranges established for healthy cattle. All fat samples tested contained trace levels of α-BHC (mean value, 23 ppb) and lindane (mean value, 3 ppb). 相似文献

The interaction of solitons and magnons (linear modes) of the Sine-Gordon equation is studied. The phase shift of the magnon in presence of a soliton gives rise to a shift of the position of the soliton, if magnon wave packets are considered. This leads to a stochastic part of the soliton motion in a gas of thermally activated magnons yielding an effective velocity and a diffusion constant. The influence of this effect on the solitonic part of the correlation function is discussed and compared with the recent experiment by Steiner and Kjems. 相似文献
