Size-exclusion chromatography (SEC) has proven its capability to refold a variety of proteins using a range of gel filtration column materials, demonstrated in the growing body of experimental evidence. However, little effort has been allocated to the development of mechanistic models describing size-exclusion chromatographic refolding reactors (SECRR). Mechanistic models are important since they provide a link between process variables like denatured and reduced protein feed concentration (Cf,D&;R), flow rate, column length, etc., and performance indicators like refolding yield (YN), thereby opening the possibility for in silico design of SECRRs. A critical step, in the formulation of such models, is the selection of an adequate reaction mechanism, which provides the direct link between the separation and the refolding yield. Therefore, in this work we present a methodology using a SEC refolding reactor model, supported by a library of reaction mechanisms, to estimate a suitable reaction scheme using experimental SEC refolding data. SEC refolding data is used since it provides information about the mass distribution of monomers and aggregates after refolding, information not readily available from batch dilution refolding data alone. Additionally, this work presents (1) a systematic analysis of the reaction mechanisms considered using characteristic time analysis and Damköhler maps, revealing (a) the direct effect of a given reaction mechanism on the shape of the SEC refolding chromatogram (number of peaks and resolution) and (b) the effect that the competition between convection, refolding and aggregation is likely to have on the SEC refolding yield; (2) a comparison between the SECR reactor and the batch dilution refolding reactor based on mechanistic modeling, quantitatively showing the advantages of the former over the latter; and (3) the successful application of the modeling based strategy to study the SEC refolding data of an industrially relevant protein. In principle, the presented modeling strategy can be applied to any protein refolded using any gel filtration material, providing the proper mass balances and activity measurements are available. 相似文献

This study examines the difficulties college students experience when creating and interpreting graphs in which speed is one of the variables. Nineteen students, all preservice elementary or middle school teachers, completed an upper‐level course exploring algebraic concepts. Although all of these preservice teachers had previously completed several mathematics courses, including calculus, they demonstrated widespread misconceptions about the variable speed. This study identifies four cognitive obstacles held by the students, provides excerpts of their graphical constructions and verbal interpretations, and discusses potential causes for the confusion. In particular, misconceptions arose when students interpreted the behavior and nature of speed within a graphical context, as well as in situations where they were required to construct a graph involving speed as a variable. The study concludes by offering implications for the teaching and learning of speed and its interpretation within a graphical setting. 相似文献

The SH + CH(3) product channel for the photodissociation of CH(3)SH at 204 nm was investigated using the sliced velocity map ion imaging technique with the detection of CH(3) products using state selective (2+1) resonance enhanced multiphoton ionization (REMPI). Images were measured for CH(3) formed in the ground and excited vibrational states (v(2) = 0, 1, and 2) of the umbrella mode from which the correlated SH vibrational state distributions were determined. The vibrational distribution of the SH fragment in the SH + CH(3) channel at 204 nm is clearly inverted and peaks at v = 1. The highly negative anisotropy parameter of the CH(3) (v(2) = 0, 1, and 2) products is indicative of a fast dissociation process for C-S bond cleavage. Two kinds of slower CH(3) products were also observed (one of which was partly vibrationally resolved) that are assigned to a two-step photodissociation processes, in which the first step is the production of the CH(3)S (X(2)E) radical via cleavage of the S-H bond in CH(3)SH, followed by probe laser photodissociation of nascent CH(3)S radicals yielding CH(3)(X(2)A(1), v(2) = 0-2) + S((3)P(j)/(1)D) products. 相似文献

Fast atom bombardment, combined with high-energy collision-induced tandem mass spectrometry, has been used to investigate gas-phase metal-ion interactions with captopril, enalaprilat and lisinopril, all angiotensin-converting enzyme inhibitors.Suggestions for the location of metal-binding sites are presented. For captopril, metal binding occurs most likely at both the sulphur and the nitrogen atom. For enalaprilat and lisinopril, binding preferably occurs at the amine nitrogen. This paper describes a rapid technique for reconstruction of the internal area function of a duct using blockage-induced eigenvalue shifts determined from eigenfrequencies measured under two sets of duct termination boundary conditions. A single broad band maximum length sequence (MLS) measurement of short duration is utilized to obtain the transfer function of the duct, which in turn can be utilized to determine its eigenvalue shifts and subsequently its internal area function using an inverse perturbation technique. The reconstruction results display the same order of accuracy as those obtained previously using swept sine measurements of extended duration. An expression for the determination of the area function is presented utilizing resonant frequency information alone, thus rendering duct length determination unnecessary. A computational routine further simplifies the process such that the accuracy of the technique could be ascertained for a range of configurations including longer ducts and ducts that initially have nonuniform internal cross section over their length. Development of a relationship between obstacle length and wavelength of the lowest eigenfrequency required for successful reconstruction is also described. This is an important result for longer ducts where measurement of lower eigenfrequencies may present problems using standard measurement equipment. 相似文献

This work demonstrates that the type of ion-exchanger (anion or cation), the mode of operation (bind-and-elute or flow-through), and the operational pH of ion-exchange chromatography (IEX) can be selected in a fast and rational way by analytical pH-gradient IEX operations, thereby eliminating the need for pH scouting or high-throughput screening. The developed approach was applied for the selection of an IEX process for the capture of a monoclonal antibody (MAb) from hybridoma cell culture supernatant (CCS). It was found within a day that MAb can optimally be captured by bind-and-elute mode cation-exchange chromatography (CEX) at pH 4.5 or anion-exchange chromatography (AEX) at pH 7.2 without lowering the salt concentration in the CCS. The performance of both CEX and AEX was predicted to be equal for this particular MAb capture. 相似文献

The extraction behavior of Nd(III) and Eu(III) with 0.05 mol dm−3 furosemide in benzyl alcohol as single acidic extractant and then with equimolar (0.05 mol dm−3) synergic mixture of furosemide as acidic extractant and tribenzylamine as neutral donor in benzyl alcohol has been studied from aqueous solutions of pH 1 to 6. The effect of various parameters and of various cations and anions on the extraction of these metal ions was investigated. The composition of the extracted adducts was determined by slope analysis method that came out to be [(M(FS)2)+ (CH3COO)−] and [M(FS)3·3TBA] where M = Nd(III) and Eu(III).
