The novel NAD+-linked opine dehydrogenase from a soil isolate Arthrobacter sp. strain 1C belongs to an enzyme superfamily whose members exhibit quite diverse substrate specificites. Crystals of this opine dehydrogenase, obtained in the presence or absence of co-factor and substrates, have been shown to diffract to beyond 1.8 ? resolution. X-ray precession photographs have established that the crystals belong to space group P21212, with cell parameters a = 104.9, b = 80.0, c = 45.5 ? and a single subunit in the asymmetric unit. The elucidation of the three-dimensional structure of this enzyme will provide a structural framework for this novel class of dehydrogenases to enable a comparison to be made with other enzyme families and also as the basis for mutagenesis experiments directed towards the production of natural and synthetic opine-type compounds containing two chiral centres.  相似文献

Large single crystals of NbP have been prepared. A single-crystal X-ray diffraction study shows that it crystallizes in tetragonal symmetry with space group I4(1)md (No. 109) and lattice parameters a = 3.3324(2) ?, c = 11.3705(7) ?, and Z = 4. A full matrix least-squares refinement based on a unique data set of 285 reflections (I> 2sigma(I)) yielded R(F) = 0.017 and R(w)(F(2)) = 0.046 for nine variables. The unit cell consists of one unique Nb and one P, each in trigonal prismatic coordination with the other element. There are two short and four long bond distances of Nb-P. The Nb-Nb bond distances are significantly shorter than R(c) = 4.09 ?, the critical distance required for good Nb-Nb 4d orbital overlap for niobium metal-metal bonds. NbP shows metallic behavior with rho = 4.5 x 10(-)(5) Omega cm at room temperature. Magnetic susceptibility measurements on a collection of randomly oriented single crystals indicate very weak Pauli paramagnetism ( approximately 10(-)(5) emu/mol). A discussion of the structure as well as the physical properties of NbP compared with those of previous results are presented. The band structure of NbP based on the extended Hückel (tight-binding) calculations is presented along with an analysis that reveals that the valence band is built up from three center bonds localized within Nb(3) triangles.  相似文献

Precursors of the pincer-ligated iridium species, (PCP)Ir, react with nitrobenzene or acetophenone at ambient temperature to give O,C-chelated complexes resulting from addition of an aryl C-H bond and coordination of a nitro or acetyl oxygen. The C-H additions appear to be completely regioselective for the position ortho to the functional group; however, structural characterization and low-temperature NMR studies demonstrate that the reaction does not proceed via coordination of the functional group followed by C-H addition. In the case of nitrobenzene, kinetic preference for the para and meta positions is demonstrated at low temperature. Addition occurs more slowly at the ortho position, without assistance by the functional group; the ortho-C-H addition product is then trapped by chelation.  相似文献

The early lanthanide benzenefluorothiolates (Ln(SC(6)F(5))(3); Ln = La, Ce, Pr, Nd, Sm, Gd) react with Hg(SC(6)F(5))(2) in DME to form ionic heterometallic compounds with Ln cations and Hg anions. X-ray diffraction analyses of all compounds reveal an isostructural series with the general formula [(DME)(3)Ln(SC(6)F(5))(2)](2)[Hg(2)(SC(6)F(5))(6)]. In the structures, a fluorothiolate ligand has been extracted from the Ln coordination sphere that is saturated with three neutral DME donor ligands and a dative interaction between one ortho fluorine and the Ln. Distances between Ln and F do not vary simply with Ln ionic radius. There are two Ln cations with charge balanced by a Hg(2)(SC(6)F(5))(6) dianion composed of two distinctly nonideal Hg(II) tetrahedra, all connected through a series of pi-pi interactions that link cations with anions in a one-dimensional array and anions to anions in a more complex 2D network.  相似文献

The title compound, [2,6‐bis(di‐tert‐butylphosphino)phenyl‐1κ3P,C1,P′]di‐μ‐chlorido‐1:2κ4Cl:Cl‐(2η4‐cycloocta‐2,5‐diene)hydrido‐1κH‐diiridium(I,III) hexane hemisolvate, [Ir2(C8H12)(C24H43P2)Cl2H]·0.5C6H14 or [(tBuPCP)IrH(μ2‐Cl)2Ir(COD)][tBuPCP is κ3‐2,6‐(tBu2PCH2)2C6H3 and COD is η4‐2,5‐cyclooctadiene], is an IrIII/IrI dimer bridged by two chloride ions. The Ir2Cl2 framework is nearly planar, with a dihedral angle of 13.04 (4)° between the two Ir centers. The compound was isolated as a hexane hemisolvate. Cadmium sulfide (CdS) nanoclusters were prepared by a freeze drying method from two types of cadmium carboxylates. One was cadmium methacrylates that were part of poly(methyl methacrylate) (PMMA) ionomer. The other was cadmium acetates that were dispersed in PMMA. X-ray diffraction was mainly used to study the formation and the size of nanoclusters. The size of CdS made from the ionomer was 0.9 nm, whereas that from the composite of cadmium acetate and PMMA was 2 nm. This was consistent with the size difference of the precursors of CdS: i.e., Cd carboxylate nanoclusters (ionic aggregates) were smaller in the ionomer than in the PMMA mixture, because ionic groups in the ionomer were constrained due to their connectivity to backbone chains and thus forming smaller ionic aggregates. Once stabilized, however, CdS nanocluster sizes were unchanged despite thermal treatments at up to 220 °C for 24 h for both systems. Structural transformations from a freeze dried cadmium carboxylate powder, to a CdS-containing powder, and to a heat-treated CdS-containing sample are speculated for both types of systems.  相似文献

The amidation reaction of a tetrahydroisoquinolin-1-one-4-carboxylic acid is a key step in the multi-kilogram-scale preparation of the antimalarial drug SJ733, now in phase 2 clinical trials. In the course of investigating THIQ carboxamidations, we found that propanephosphonic acid anhydride (T3P) is an effective reagent, although the yield and byproducts vary with the nature and quantity of the base. As a control, the T3P reaction of a 3-(2-thienyl) THIQ was performed in the absence of the amine, and the products were characterized: among them are three dimeric allenes and two dimeric lactones. A nucleophile-promoted ketene dimerization process subject to subtle steric and stereoelectronic effects accounts for their formation. Two novel monomeric products, a decarboxylated isoquinolone and a purple, fused aryl ketone, were also isolated, and mechanisms for their formation from the ketene intermediate are proposed.  相似文献
