A new series of photoactivated DNA oxidizing agents in which an acridine moiety is covalently linked to viologen by an alkylidene spacer was synthesized, and their photophysical properties and interactions with DNA, including DNA cleaving properties, were investigated. The fluorescence quantum yields of the viologen-linked acridines were found to be lower than that of the model compound 9-methylacridine (MA). The changes in free energy for the electron transfer reactions were found to be favorable, and the fluorescence quenching observed in these systems is explained by an electron transfer mechanism. Intramolecular electron transfer rate constants were calculated from the observed fluorescence quantum yields and singlet lifetime of MA and are in the range from 1.06x10(10) s(-1) for 1 a (n=1) to 6x10(8) s(-1) for 1 c (n=11), that is, the rate decreases with increasing spacer length. Nanosecond laser flash photolysis of these systems in aqueous solutions showed no transient absorption, but in the presence of guanosine or calf thymus DNA, transient absorption due to the reduced viologen radical cation was observed. Studies on DNA binding demonstrated that the viologen-linked acridines bind effectively to DNA in both intercalative and electrostatic modes. Results of PM2 DNA cleavage studies indicate that, on photoexcitation, these molecules induce DNA damage that is sensitive to formamidopyrimidine DNA glycosylase. These viologen-linked acridines are quite stable in aqueous solutions and oxidize DNA efficiently and hence can be useful as photoactivated DNA-cleaving agents which function purely by the co-sensitization mechanism.  相似文献

Cellulose nanofibres (CNF) with diameter 10–60 nm were isolated from raw banana fibres by steam explosion process. These CNF were used as reinforcing elements in natural rubber (NR) latex along with cross linking agents to prepare nanocomposite films. The effect of CNF loading on the mechanical and dynamic mechanical (DMA) properties of NR/CNF nanocomposite was studied. The morphological, crystallographic and spectroscopic changes were also analyzed. Significant improvement of Young’s modulus and tensile strength was observed as a result of addition of CNF to the rubber matrix especially at higher CNF loading. DMA showed a change in the storage modulus of the rubber matrix upon addition of CNF which proves the reinforcing effect of CNF in the NR latex. A mechanism is suggested for the introduction of the Zn–cellulose complex and its three dimensional network as a result of the reaction between the cellulose and the Zinc metal which is originated during the composite formation.  相似文献

There is a need for evolving hydraulically efficient roof profile of bellmouth for high head sluice spillways, as sluice roof is susceptible to cavitation damage. In this paper, formulation and development of a numerical model for simulating the spillway flow and its application to a sluice spillway are presented. The main focus of the simulation study is to apply the developed model to investigate the effects of sluice roof profile geometry on the pressure distribution, the discharge coefficient and the nature of flow regime within the sluice bellmouth. From the analysis of results for eight different roof profiles by varying the entry and exit angles of elliptic bellmouth transition, some important observations have been suggested, which are of practical relevance to hydraulic design engineers. The numerical model results are compared for one profile with physical model study. The simulated results are in close agreement with the measured values. The aim of this study was to measure the shear modulus of the vocal fold in a human hemilarynx, such that the data can be related to direction of applied stress and anatomical context. Dynamic spring rate data were collected using a modified linear skin rheometer using human hemilarynges, and converted to estimated shear modulus via application of a simple shear model. The measurement probe was attached to the epithelial layer of the vocal fold cover using suction. A sinusoidal force of 3g was applied to the epithelium, and the resultant displacement logged at a rate of 1kHz. Force measurement accuracy was 20microg and position measurement accuracy was 4microm. The force was applied in a transverse direction at the midmembranous point between the vocal process and the anterior commissure. The shear modulus of the three female vocal folds ranged from 814 to 1232Pa. The shear modulus of the three male vocal folds ranged from 1021 to 1796Pa. These data demonstrate that it is possible to obtain estimates for the shear modulus of the vocal fold while preserving anatomical context. The modulus values reported here are higher than those reported using parallel plate rheometry. This is to be expected as the tissue is attached to surrounding structures, and is under natural tension.  相似文献

Phytochemicals serve as potential therapeutic agents for the prevention and treatment of diseases. In this study, we elucidate the renoprotective activity of compounds isolated from Eucalyptus globulus and Melaleuca styphelioides extracts in glucose- and oxalate-challenged NRK-49F cell model. The antioxidant potential of isolated compounds was evaluated based on their effect on antioxidant enzyme activities and lipid peroxidation levels. The results demonstrated that exposure of NRK-49F cells to glucose and oxalate stress augmented cell damage and attenuated antioxidant enzyme activities. The phytochemicals 2,2,8-trimethyl-6-formyl-chrom-3-ene-7-O-β-D-glucopyranoside, Cornusiin B and tellimagrandin I treatment restored antioxidant enzyme activity, significantly lowered lipid peroxidation levels and effectively protected cells from glucose and oxalate stress equivalent to the known antioxidant, N-acetyl cysteine. Pterocarinin A significantly reversed cellular damage owing to glucose stress. In conclusion, the compounds isolated from E. globulus and M. styphelioides showed potential cytoprotective and anti-oxidative property against glucose- and oxalate-induced oxidative stress in NRK-49F cells.  相似文献

Generalized incomplete Trojan-type designs are defined to be row–column designs in which each cell, corresponding to the intersection of row and column, contains more than one treatment and the rows are incomplete. NMR (13)C spin lattice relaxation (1/T(1)) rates of dipalmitoylphosphatidylcholine (DPPC) bilayers obtained from molecular dynamics simulations of 72 and 288 lipids are compared with each other, with experimental values from large liposomes obtained by magic angle spinning, and with previously published experimental data from small vesicles. The experimental results for multilayers and vesicles at the same frequencies differ only slightly. The simulation results indicate that T(1) relaxation in the 15.1 to 201.2 MHz carbon frequency range and up to 100 A length scale is dominated by fast isomerizations and slower lipid wobble (D perpendicular approximately 2.5 x 10(8) s(-1)). Rotational diffusion about the lipid long axis (described by D(parallel)) does not make a substantial contribution to the T(1). Modifications to the acyl chain torsional potential energy function used for the simulations substantially improve agreement with experiment.  相似文献

Over the past decade, organocatalysis has emerged as a powerful tool for stereoselective carbon-carbon bond formation under exceptionally mild conditions. The organocatalytic versions of a large number of traditional synthetic transformations are now well established and the quest for new applications of the basic concepts of organocatalysis continues. This review addresses the emergent interest in the organocatalytic vinylogous aldol reaction. While noteworthy progress has been made in this area, significant challenges lie ahead.  相似文献
