Conditions for converting a set of five standard proteins to electrochemically active sodium dodecylsulfate (SDS) complexes were worked out with the aim of using such complexes for conductivity detection with a a chip electrophoresis system. The results obtained were compared with standard capillary electrophoresis (37 cm (effective length 30 cm)×75 μm I.D. capillary, 10 kV, negative polarity at the inlet). The chip separations were run at 500 V per chip (100 V/cm) as compared to the standard capillary arrangement, which was run at 266.6 V/cm. For the capillary set-up the protein complexes were prepared in aqueous solution (Milli-Q water) made 10 mM with respect to SDS. If the SDS concentration was increased to 50 mM, the separation in the capillary was incomplete. On the other hand with the chip system both approaches yielded acceptable results. The chip separations were slightly (but not distinctly) shorter and offered better separations than the standard set-up. The concentration of the surfactant used for the preparation the complexes results in alternations of the elution sequence, which is preserved if the chip separation is used instead of the capillary set-up. Apparently the full capacity of protein–SDS binding is not exploited for the preparation of the adducts. 相似文献

The interaction between low molecular-mass homopeptides and mixtures of nonionic and anionic surfactants has been assessed by using reversed-phase thin-layer chromatography. The relative strength of interaction for mixtures of sodium dodecylsulfate and tridecylalcohol diglycolate (GNX) at the molar ratios of 8:2, 6:4, 4:6 and 2:8 has been calculated and its relationship with the physicochemical parameters (number of amino acid units, hydrophobicity, side chain bulkiness, electronic characteristics) of peptides has been computed by stepwise regression analysis. Each peptide interacted with each surfactant mixture the strength of interaction markedly depending on both the character of the peptide and the composition of the surfactant mixture. The hydrophobicity and electronic properties of the amino acid units exerted the highest influence on the strength of interaction at the highest concentration of the nonionic surfactant (GNX) whereas the number of amino acid units in the peptide molecule and the bulkiness of the amino acid side chain governed the strength of interaction at the lowest concentration of GNX. 相似文献

The relative strength of interaction between anionic (SDS) and nonionic surfactant (octaethoxylated oleyl alcohol, GEN) and homologous series of peptides was determined by reversed-phase thin-layer chromatography (RP-TLC) carried out on alumina layers impregnated with paraffin oil. The relative strength of interaction was calculated and was correlated with the physicochemical parameters of peptides. It was established that each peptide interacted with both surfactants and with their mixture (1:1, m/m). The relative strength of interaction depended on the number of amino acid units in the peptide, side chain bulk and electronic properties and hydrophobicity of the amino acids. The impact of individual parameters highly depended on the character of surfactant. The low and the high resolution electron impact (EI) and field desorption (FD) mass spectra (MS) of substituted 5- or 6-membered ring sultams are given and the main fragmentation pathways are interpreted. Sultams carrying polar substituents, such as dicarboxylic acids and their mono- and di-amides show either small or no molecular ions with EI-MS. Their existence, however, can be indirectly inferred by metastable defocusing. In contrast to these findings FD-MS display in all our examples the quasimolecular ions (M + 1)+ as the base peaks of the spectra. While the pattern for the EI fragmentations follows the generally known rules very closely, five general aspects for the fragmentation pathway of FD mass spectra have been established which demonstrate the characteristics of this modern analytical method. The use of high temperature activated emitters enables a choice to be made between obtaining only molecular peaks and producing significant fragments via a controlled thermal degradation. This is the first time that the complementary and different characters of the information obtained with the two ionization modes EI and FD have been demonstrated on a homologous series of compounds. 相似文献

Avian eggshell matrix proteins were studied by two analytical approaches. Peptide mapping was done by trypsin and pepsin followed by collagenase cleavage; analyses were carried out by capillary electrophoresis and reversed-phase high-performance liquid chromatography (HPLC). Comparison of peptide maps obtained by both methods revealed a complex mixture of peptides in the insoluble layers of the eggshell; it was concluded that there are at least three different insoluble protein/peptide layers in the avian eggshell (cuticle, palisade, and mammillary layer). Partial characterization of peptides in each layer was made by HPLC-mass spectrometry analysis. There is an evidence that the eggshell insoluble proteins contain species susceptible to collagenase cleavage, however, the sequences split by this enzyme probably are not those typical for the main triple-helical core of collagenous proteins. It is proposed that the action of collagenase upon eggshell proteins is caused by the side effect of collagenase described previously with synthetic peptides. Some of the proteins present are probably glycosylated. Fatty acid content in the insoluble eggshell layers (after decalcification) was in the range of 2-4% (which reflected both lipid and lipoproteins bound fatty acids).
