Human islet amyloid polypeptide (IAPP) is the major component of the amyloid deposits found in the pancreatic islets of patients with type 2 diabetes mellitus. After synthesis, IAPP is stored in the β-cell granules of the pancreas at a pH of approximately 5.5 and released into the extracellular compartment at a pH of 7.4. To gain insight into the possible consequences of pH differences for properties and membrane interaction of IAPP, we here compared the aggregational and conformational behavior of IAPP as well as IAPP-membrane interactions at pH 5.5 and pH 7.4. Our data reveal that a low pH decreases the rate of fibril formation both in solution and in the presence of membranes. We observed by CD spectroscopy that these differences in kinetics are directly linked to changes in the conformational behavior of the peptide. Mechanistically, the processes that occur at pH 5.5 and pH 7.4 appear to be similar. At both pH values, we found that the kinetic profile of IAPP fibril growth matches the kinetic profile of IAPP-induced membrane damage, and that both are characterized by a lag phase and a sigmoidal transition. Furthermore, monolayer studies as well as solid-state NMR experiments indicate that the differences in kinetics and conformational behavior as function of pH are not due to a different mode of membrane insertion. Our study suggests that a low pH prevents aggregation and membrane damage of IAPP in the secretory granules, most likely by affecting the ionization properties of the peptide. 相似文献

The lithium salt of the anionic SPS pincer ligand composed of a central hypervalent lambda4-phosphinine ring bearing two ortho-positioned diphenylphosphine sulfide side arms reacts with [Mn(CO)5Br] to give fac-[Mn(SPS)(CO)3]. This isomer can be converted photochemically to mer-[Mn(SPS)(CO)3], with a very high quantum yield (0.80+/-0.05). The thermal backreaction is slow (taking ca. 8 h at room temperature), in contrast to rapid electrode-catalyzed mer-to-fac isomerization triggered by electrochemical reduction of mer-[Mn(SPS)(CO)3]. Both geometric isomers of [Mn(SPS)(CO)3] have been characterized by X-ray crystallography. Both isomers show luminescence from a low-lying 3IL (SPS-based) excited state. The light emission of fac-[Mn(SPS)(CO)3] is largely quenched by the efficient photoisomerization occurring probably from a low-lying Mn-CO dissociative excited state. Density functional theory (DFT) and time-dependent DFT calculations describe the highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) of fac- and mer-[Mn(CO)3(SPS)] as ligand-centered orbitals, largely localized on the phosphinine ring of the SPS pincer ligand. In line with the ligand nature of its frontier orbitals, fac-[Mn(SPS)(CO)3] is electrochemically reversibly oxidized and reduced to the corresponding radical cation and anion, respectively. The spectroscopic (electron paramagnetic resonance, IR, and UV-vis) characterization of the radical species provides other evidence for the localization of the redox steps on the SPS ligand. The smaller HOMO-LUMO energy difference in the case of mer-[Mn(CO)3(SPS)], reflected in the electronic absorption and emission spectra, corresponds with its lower oxidation potential compared to that of the fac isomer. The thermodynamic instability of mer-[Mn(CO)3(SPS)], confirmed by the DFT calculations, increases upon one-electron reduction and oxidation of the complex. 相似文献

Reactions of Ln(BH4)3(THF)n and [Li(Et2O)]SPS(Me)], the lithium salt of an anionic SPS pincer ligand composed of a central hypervalent lambda4-phosphinine ring bearing two ortho-positioned diphenylphosphine sulfide sidearms, led to the monosubstituted compounds [Ln(BH4)2(SPS(Me))(THF)2] [Ln = Ce (1), Nd (2)], while the homoleptic complexes [Ln(SPS(Me))3] [Ln = Ce (3), Nd (4)] were obtained by treatment of LnX3 (X = I, BH4) with [K(Et2O)][SPS(Me)]. The [UX2(SPS(Me))2] complexes [X = Cl (5), BH4 (6)] were isolated from reactions of UX4 and the lithium or potassium salt of the [SPS(Me)]- anion. The X-ray crystal structures of 1.1.5THF, 2.1.5THF, 3.2THF.2Et2O, and 5.4py reveal that the flexible tridentate [SPS(Me)]- anion is bound to the metal as a tertiary phosphine with electronic delocalization within the unsaturated parts of the ligand. 相似文献

This short account summarizes recent results obtained in the coordination chemistry of phosphinines and emphasizes their analogy with CO ligands. Reduced complexes can be easily assembled through the reaction of reduced 2,2′-biphosphinine dianions with transition metal fragments. Theoretical calculations were performed to establish the oxidation state of the metal in these complexes. Though many reduced complexes are available, phosphinines proved to be too sensitive toward nucleophiles to be used as efficient ligands in most catalytic processes. However, the high electrophilicity of the phosphorus atom can be exploited to synthesize phosphacylohexadienyl anions which exhibit a surprising coordination chemistry. When phosphino sulfide groups are incorporated as ancillary tridentate anionic SPS ligands can be easily produced. These ligands can bind different transition metal fragments such as M-X (M = group 10 metal, X = halogen), Rh-L (L = 2 electron donor ligand), Cu-X and Au-X (X = halogen). Palladium(II) complexes proved to be active catalyst in the Miyaura cross-coupling reaction. Bidentate anionic PS ligands were also synthesized following a similar approach. Their Pd(II) (allyl) derivatives showed a very good activity in the Suzuki catalyzed cross-coupling process that allows the synthesis of biphenyl derivatives through the reaction of phenylboronic acid with bromoarenes. 相似文献

The structural and electronic transport properties of La1−x
Ce
x
MnO3 (x=0.0–1.0) have been studied. All the samples exhibit orthorhombic crystal symmetry and the unit cell volume decreases with
Ce doping. They also make a metal-insulator transition (MIT) and transition temperature increases with increase in Ce concentration
up to 50% doping. The system La0.5Ce0.5MnO3 also exhibits MIT instead of charge-ordered state as observed in the hole doped systems of the same composition. 相似文献
